EXCEL INSTRUCTIONS

1. FORMULA FOR CORRECTED ZONE OF INHIBITION


 ƒx = (B3/C3)*90
 copy top cell, select remainder of row of which this formula applies to, and
hit paste.

2. There are 3 readings for each concentration – we want to find a mean average for
zone of inhibition for each concentration.
 Highlight mean zone of inhibition diameter cell for concentration of 30
 Highlight the 3 rows in the first table and click average next to function
button
 Repeat this for each strength
 Place in bold the ones you have applied to prevent error of adding the wrong
cells to the formula

3. Calculate the log concentration of streptomycin


 Calculate the first cell in the log row for the first concentration and select the
log button in the function bar

Now we are ready to draw graphs

1. Highlight what you want to plot


2. Concentration of streptomycin and log concentration of streptomycin
3. Click insert – select scatter chart (click the first one)
4. Add axis labels – click chart, click design tab, clock chart element, select axis titles
 Click horizontal and vertical and rename to your desired axis title
 X-axis = concentration of streptomycin (mg/mL)
 Y-axis = mean zone of inhibition diameter (mm)
5. We then need to insert a trend line
 Click design, add chart element, select trendline, choose moving average
6. We then can use this graph to calculate our unknown concentrations of
streptomycin and silver nitrate and copper sulphate

Entering data into last table for unknowns.


1. Find mean zone of inhibition and diameter of plates for unknowns 1 and 2
2. Calculate corrected zone of inhibition (see function above - 1.)
3. Use the value of corrected zone on the graph we have generated earlier to estimate
the value of concentration of unknowns.
4. Use the silver nitrate and copper sulphate values to obtain an equivalent
concentration of streptomycin
Plotting the second graph.

1. Select concentration of streptomycin and log concentration rows simultaneous


2. Insert chart – scatter
3. Insert axis titles (ensure you state the x axis is log…)
4. Insert trend line – select linear
5. Obtain same data as in previous graph (estimate concentrations of unknowns and
heavy metals)
6. To add more lines to the graph in order to read the graph more easily
 Select chart elements
 Select gridlines, choose primary minor horizontal and vertical
 Do for both graphs

The last thing to do is to calculate the concentration of the unknowns from the log
concentrations
 Click on the last small box
 Enter into the log concentration the value you found when reading the log graph
 Then click the concentration cell, hit ƒx button and type 10^and click log conc cell
