The document provides instructions for preparing and studying plant cell structure under a light microscope using onion and Hydrilla leaf cells. Slides of onion epidermal cells and Hydrilla leaf cells are prepared, stained with iodine if needed, and observed under a microscope to view and label their cellular structures.

Materials : Onion, Hydrilla sp, iodine solution, distilled water, filter paper Apparatus : Light microscope, slides, cover slips, scalpel, forceps, dropper, mounting needle Procedure Procdure 1) A slide of epidermal cells of onion is prepared using the following steps. (a) A scalpel is used to cut a piece of onion scale measuring 1cm x 1cm (b) A pair of forceps is used to strip the epidermis layer. (c) It is mounted on a slide with a drop of water (d) The slide is covered with the help of a mounting needle. Procedure 2. A drop of iodine is put on one slide of cover slip. A piece of filter paper is put on the opposite side of the cover slip to draw the iodine in to stain the cells 3. The slide is observed under a light microscope 4. The observation is drawn and labelled 5. Another slide is prepared using the leaf of Hydrilla sp. The slide is observed under a light microscope. The observation is drawn and labelled. Observation
