Why To Preserve Bacteria?

 In nature there are only 1% bacteria which is patheogenic and harmful to


Animalia and Plantae
 99% of bacterial populations are of economic importance for human beings
and plant
 In soil for nutrient uptake in food industry,in sewage treatment, in medical
Industry.
 So preservation of bacteria is one of the most important profitable practice
economically as well as enviromentally
Pure Culture Method

 Spread Plate Method


 This is the best method to isolate the pure colonies.
 It is mixed with normal saline and serially diluted.
 0.1 ml of sample taken from diluted mixture,which is placed on the surface of
agar plate and spread evenly over the surface.
 Incubates the plates
 After inclubation ,colonies are observed on the surface of agar .
Preservation by overlaying culture with
mineral oil
 This is the simple and most economical method of maintaining pure culture.
 In this method,the sterile liquid paraffin is poured over the slope of the
culture and stored upright at room temperature .The layers of paraffin
ensures anaerobic condition and prevents dehydration of the medium.
 This condition helps pure culture to remain in dormant state and thus can be
preserved from months to year.
