PENGERTIAN

HPV merupakan virus DNA sirkuler

rantai ganda, berukuran kecil, tidak
memiliki selubung (envelope) dan masuk
dalam keluarga Papillomaviridae
Human Papilloma Virus (HPV) merupakan virus yang menyebabkan
keganasan kanker serviks. Virus ini bersifatonkogenik yang berpotensi
menyebabkan kanker serviks,
Kanker Serviks merupakan suatu bentuk keganasan yang terjadi pada
leher rahim (serviks) yang disebabkan oleh adanya pertumbuhan yang
abnormal dari jaringan epitel serviks akibat adanya infeksi yang persisten
human papillomavirus (HPV) tipe high risk (HR- HPV) onkogenik.
MORFOLOGI
HPV merupakan virus double stranded DNA
(dsDNA) yang tidak memiliki selubung dengan
diameter berkisar 55 nm. Kapsid dari HPV
berbentuk ikosahedral dan tersusun atas 72 capsomer
pentamerik (320 protein L1) dan 12 molekul protein
L2. Kapsid ini membungkus satu molekul dsDNA
sirkular. Molekul genom HPV berasosiasi dengan
molekul histon.

GEJALA KLINIS
Perdarahan vagina yang tidak normal, yaitu perdarahan ketika berhubungan seksual,
pasca-menopause, dan di luar siklus haid, vaginal discharge yang berwarna
keputihan, seperti nanah dan berbau, serta nyeri pada pinggul. Kutil kelamin
biasanya berwarna merah muda, lunak, permukaannya bervariasi ada yang datar atau
meninggi, berjumlah satu atau lebih, dan kadang berbentuk seperti kembang kol
(cauliflower-shaped). Lesi pada daerah genitalia perempuan umumnya terdapat pada
bagian belakang mulut vagina, kemudian menyebar ke vulva dan akhirnya vagina
dan serviks. Lesi ini menimbulkan rasa tidak nyaman akibat gatal.
 CARA PENGAMBILAN SAMPEL

Tahapan-Tahapan yang dilakukan dalam pemeriksaan HPV DNA:

- Pasien akan diminta untuk melepaskan celana atau rok serta celana dalam, lalu berbaring
telentang dengan lutut tertekuk serta tungkai terangkat dan disangga oleh penyangga.
- Dokter akan memasukkan alat yang disebut spekulum ke dalam vagina, sehingga dinding
vagina terbuka dan dokter dapat memeriksa bagian dalam vagina serta leher rahim. Tahap
ini dapat menimbulkan rasa tidak nyaman di bagian perut bawah.
- Dokter akan mengambil sampel sel dari leher rahim dengan menggunakan sikat atau spatula
khusus. Setelah itu, sampel akan dimasukkan ke dalam tabung kecil lalu dibawa ke
laboratorium untuk dianalisis.
 DIAGNOSA

Tes Asam Asetat Pap Smear

Dalam 1-5 menit lesi akan Tes pap serviks saat
berubah warna menjadi putih melakukan pemeriksaan
(acetowhite) pelvik untuk skrining

PCR Histologi
metode yang sensitif dan spesifik dalam
mendiagnosa infeksi HPV.. histologis menunjukkan kelainan
pada epidermis, termasuk akantosis
(menebalnya stratum spinosum)
