Background:

Biotin labeling can use to detect proteins using various methods


(i.e., non-radioactive immunoassays, cytochemical staining, cell
separation, isolation of nucleic acids, detection of specific DNA/
RNA sequences by hybridisation, probing of conformational
changes in ion channels).

Many reagents are available for labeling nucleic acids with bio-
tin. Biotin-CE Phosphoramidite can add multiple biotins to either
the 3’ or 5’ end of an oligonucleotide. Similarly, Biotin-TEG-CE
Phosphoramidite adds biotin to the 3’ and 5’- ends of an oligo.
5’-Biotin-CE Phosphoramidite and 5’-Biotin-CE Phosphoramid-
ite II can only add biotin to the 5’-end while 3’-Biotin CPG only
works at the 3’-end. For internal addition of biotin, Biotin-dT-CE
Phosphoramidite is suggested.

It should be noted that the DMT group of 3’-Biotin CPG can


be used for cartridge purifications. There is potential for cross
contamination in HPLC purifications--which should be avoided
when preparing biotinylated PCR primers.
