12.OBJECTIVES:

To prepare Hydrogel using PVA blends by solution casting methods for

use in skincare.
Evaluation of properties such as biocompatibility, biodegradability ,swelling
properties, morphology, water absorption,aging ,thermal stability,elasticity
..etc.
Casting of hydrogels with gypsum for bone fracture dressing application using
different models.
Enhancement of hydrogels for skin care using hydrogel blends of
turmeric,honey, aloe vera.

13.METHODOLOGY:
1.Crosslinking of PVA
Crosslinking process of polymers, which might be done by a chemical reaction (e.g.
free-radical polymerization, chemical reaction of complementary groups, using high
energy irradiation, or enzymatic reaction) or by a physical reaction. physical
crosslinking method has been chosen and preferred comparable with the chemical
crosslinking method for most crosslinked polymer preparation. Several attempts

have been done to prepare crosslinked PVA-based hydrogels including radiation

crosslinking , chemical reaction with glyoxal , bi-functional reagents with
glutaraldehyde , or reaction with borates.The earliest attempt for crosslinking of PVA
using the freezingthawing method.The crystallization degree of PVA hydrogels which
have been obtained using physical crosslinking by the freezethawing method, can
be calculated.
2. Determination of water content in PVA hydrogels
Water content in PVA hydrogels is not only to provide a local moist environment
(which is a key factor for wound healing rate), but also adjusting the permeation of
nutrients, drug, gases, or protein into the cells or targeted absorption site. Dried PVA
xerogels can swell in water or in saline up to more than 1000 times their own
weight . The amount of absorbed water is usually expressed as water uptake or
swelling ratio (SW%) as shown in the following equation .Blends such as PVAAlginate/Dextran/Chitosan/Glucan were prepared and different properties were
tested.
Water uptake or swelling ratio(SR%)=[(Ws-We)/We)]*100
Ws=weight of swollen hydrogels at different intervals,We=weight of dried
hydrogels.The water absorbed up to the equilibrium swelling state is called bulk
water or free water; it fills the network space chains and the center of large pores.
3.Test for Enzymatic degradation
Accelerated in vitro enzymatic degradation tests were performed through incubation
of film samples in 10ml of SBF (simulated body fluid) at 37 deg celsius using suitable
enzymesBefore the tests the samples were dried in an oven at 37 deg celsius until
constant mass
WEIGHT LOSS=[(W 1-WF)/W1]*100
W 1 and Wf corresponds to weight of film before and after
degradation.
4.Test for anti microbial properties
Prepare culture media.Inoculate bacterial and fungal colonies.Place the hydrogel
on the media and observe for the growth of colonies.Antimicrobial properties of
honey,aloe vera and turmeric were studied.Analyses explained following
characterstics
Honey possesses antimicrobial properties,helps in promoting autolytic
debridement,deodrizes malodorous wounds,its antioxidant activitymakes it ideal
for treating wounds.
Aloe vera ,traditional medicine contains salicylic acid and its salts
salicylateshelps to curb inflammation by inhibiting the productionof hormones like
prostagladins,reduces the stiffness of bones.The cooling effect of plant helps to
regenerate new cells and helps in rebuilding the damaged tissue.
5.Casting of gels into molds
Gels are casted by preparing molds of different thickness.Two types of molds are
prepared

1.long mold:for legs.

2.short mold:for hands
