Operational Qualification Definitions

1. Definitions
1.1 Documentation
1. This document stipulates the procedures and the documentation of the Operational Qualification (OQ)
of the Shimadzu High-performance Liquid Chromatograph. When all procedures are executed and all
items pass the inspection, it is verified that the system operates to satisfy the intended purpose.
The contents of this document must be checked and approved by the Performer, Reviewer and
Manager before implementing the Operational Qualification.

2. The following are established as the responsible parties in the execution of inspections.
・Performer
The Performer is the individual who actually performs the procedures stipulated in the Installation
Qualification Protocol and the Operational Qualification Protocol.
The Performer shall be an individual who, having been trained by the Quality Assurance Department
of the Analytical & Measuring Instruments Division of Shimadzu Corporation, has been certified as
an IQ/OQ inspector by the General Manager of the Quality Assurance Department, or an individual
recognized by the Manager to be equipped with the ability to conduct this inspection.
・Reviewer
The Reviewer is an individual designated by the Manager charged with responsibility for the
system.
The Reviewer verifies the contents of the Installation Qualification Protocol and Operational
Qualification Protocol. In addition, the Reviewer shall attend the inspection procedures together with
the Performer, and evaluate the procedures.
The Manager can serve concurrently as the Reviewer.
・Manager
The Manager is the individual responsible for administering the entire HPLC system being
inspected.
As well as verifying the contents of the Installation Qualification Protocol and Operational
Qualification Protocol, the Manager shall also grant final approval to the inspection procedures.

3. The Performer shall first check the content of this document. After checking all items, the Performer
shall sign and date the cover sheet of this document and submit this document as the Operational
Qualification Protocol report. (OQ must be performed by a trained and knowledgeable individual in
accordance with 21 CFR Part 211 Section 211.25 (cGMP, USA promulgated 29 September 1978)).

4. The Reviewer shall first check the content of this entire document. After checking all items, the
Reviewer shall sign and date the cover sheet of this document as approval of the Operational
Qualification Protocol.

5. The Manager shall first check the content of this entire document. After checking all items, the
Manager shall enter a unique document control number in the Document No. column at the top right
of the cover sheet, and then sign and date the cover sheet of this document as final approval of the
Operational Qualification Protocol.

6. This document is comprised of 1. Definitions, 2. Preliminary Checks, 3. Operational Qualification


Outline, 4. Operational Qualification Protocol, and 5. Change List. All of these sections must be
included.

Operational Qualification Definitions

1.2 Scope
The Operational Qualification Protocol shall include the following.
・Operation check on each unit
Due to the modular nature of the system, the operation of each unit is checked separately.
・Operation check on overall system
Confirm that the system controller and workstation control each unit during analysis and that the
analysis results meet the prescribed criteria.
・Software and firmware check
The Operational Qualification also checks the software and firmware provided with each module.
The firmware checking is conducted based on version display and the Software Certificate of
Compliance. The software and firmware must be properly managed and change procedures must be
properly clarified

Analysis methods and data acquisition systems such as networks are not verified in the OQ of this system.
These must be separately verified by the customer.

1.3 Entries and Revisions


1. Making Entries
Use a black, permanent ballpoint pen, or equivalent, when recording results.
Names should be clearly printed.
Confirm that all items, except “Remarks” columns, are completed, and that there are no omissions.
Draw a diagonal line (from bottom left to top right) across all blank columns. (When making no entries, it
is not necessary to enter the “Change List No.__”, “Date” or “Signature”, or to enter the reason for the
change in the Change List of this document.)
Columns in which nothing has to be written in have been filled in with “N/A” in advance.
If choices are available within an item, place a checkmark in the box to select the appropriate decision.

2. Rounding of numeric values


If there are more significant figures in the measurement results than the specified criteria, round off the
result value at the number of significant figures required for the criteria. For example, if the criterion is
defined down to the first decimal place, round off the figure at the second decimal place of the result
value.

3. Deletion and correction of mistakenly filled items


To correct mistaken entries, delete the entire entry by drawing a diagonal line through it and enter the
correction in the margins. To correct numerical values or symbols (checkmark), draw 2 horizontal lines
across the mistake and re-write the entry. Moreover, write the date, and the signature, and the reason for the
change. (When deletion and correction of mistakenly filled items, it is not necessary to enter the “Change
List No.__”, “Date” or “Signature”, or to enter the reason for the change in the Change List of this
document.)

4. Changing or deleting steps, procedures or acceptance criteria


If there are sections of the steps or procedures which will not be applied, draw a diagonal line through the
entire item and write “Change List No. __” above the line and assign a number to it. Date and sign it.
Enter the necessary information in the Change List of this document.
When making a change, delete the existing description as explained above, enter the change point, and
then record the reason for the change in the Change List.

5. Revising entries
If the need to alter the contents of an entry arises, change the entry in accordance with “3. Deletion and
correction of mistakenly filled items”, and then write the reason for the change in the Change List of this
document.
When adding a new description, write it in the margins of the relevant page, and write “Change List No.
_”, the date and signature. Then, write the reason for the change in the Change List.

6. Handling Problems
Any problems that are identified in the Operational Qualification must be investigated and appropriate
actions must be taken. All such actions must be documented and be approved by the Manager.

7. In the event of a Fail


If troubles occur during an inspection and the item fails the inspection, place a checkmark in the “Fail”
box and describe the problem in the margins of the relevant page. Write “Change List No. _” and date and
sign the entry. Enter an outline of the problem in the Change List.
After the problem is solved, describe the solution and the result (“Pass”) in the margins of the relevant
page. Write “Change List No.__” and date and sign the entry. Enter the summary of the solution in the
Change List of this document.

8. Attaching data
Attached data are added by assigning page numbers as “A, B, C…” for each inspection item. If, for
example, there is a total of 3 sheets for attachment A, notate the pages as “73-A-1/3 - A-3/3”, ensuring that
no numbers are omitted. The Performer shall date and sign the attached data.

9. Other
Every item entered into the Change List shall be approved by the Reviewer and Manager.

Additional Information Significant Figures


The concept of significant figures is outlined below.

<What are Significant Figures?>


Significant figures are digits that are statistically significant in a measurement. The last digit of a significant
figure includes an error.
If we take, for example, a measured value 5.67, an error is included in the final digit, “7”. Despite the error in
the final digit “7”, the previous digit “6” is correct, and so the number of significant digits is three.
When making measurements, it is important to ascertain the number of significant digits for each measuring
instrument. For instruments with a digital display, the measurement sensitivity generally extends to the last
displayed digit, and so the last digit is considered to represent the lowest significant digit. However, the
sensitivity must be checked in the Instruction Manual.
For instruments with an analog display, the lowest significant digit corresponds to 1/10th of the smallest
increment on the scale. For example, measuring the length of an object with a ruler marked in 1-mm
increments permits measurement to the nearest 0.1 mm.

<Calculating Significant Figures>


Calculations based on measured values (such as calculating a mean value) result in more digits than the
number of significant digits used in the calculation. Such values must be rounded off to the number of
significant digits when the data is presented in reports. However, when a mean value, for example, is used in
further calculations, these calculations yield intermediate values with a larger number of digits, which are not
rounded during the calculations. No rounding for the purposes of bringing the figure in line with the
prescribed number of significant figures is conducted until the final result is obtained.
When quantities are being added or subtracted, the number of decimal places (not significant digits) in the
answer should be the same as the largest number of decimal places in the numbers being added or subtracted.
In a calculation involving multiplication or division, the number of significant digits in an answer should
equal the least number of significant digits in the numbers being multiplied, divided etc.

1. Writing a value as 0.00123000 indicates that it is accurate to six significant digits.
2. Subtracting 1.230 (four significant digits) from 1.231 (four significant digits) produces the result 0.001 ,
which is accurate to one significant digit (1 x 10-3).
3. Dividing 2.462 (four significant digits) by 1.231 (four significant digits) produces the result 2.000, which is
accurate to four significant digits (not 2).
4. If the number of significant digits of the measured result exceeds the number of significant digits of the pass
criteria, round the final result off to the number of significant digits of the pass criteria.

2. Preliminary Checks
2.1 Designation and Recording of Reviewer
Before conducting this inspection, the Manager shall designate the Reviewer. Record the Reviewer information
in the report. The Manager may also double as the Reviewer.

2.2 Performer Identification & Approval


The performer must be an individual who is trained and has adequate knowledge of this operational
qualification procedure. The Quality Assurance Department of the Analytical & Measuring Instruments
Division of Shimadzu Corporation performs such education, and recognizes individuals who are qualified to
perform the IQ/OQ procedures. An individual who is recognized as having been trained and who possesses
sufficient knowledge is certified by the General Manager of the Quality Assurance Department as a qualified
IQ/OQ Inspector, as proven by the certificate. A person who is recognized as having the ability to perform this
inspection may also perform this operational qualification procedure.
Before performing this operational qualification, the Performer’s qualification shall be confirmed and recorded
in the report.

2.3 Planning Items to be Evaluated in Operational Qualification


Prior to implementing this operational qualification, check the system configuration, determine the items to be
evaluated, record them in Operational Qualification Record and have them approved.

2.4 Calibration Certification and Instruments Used


The instruments and reagents used in the inspections are listed below.
When instruments controlled by the Shimadzu service company are to be used, copies of the Certificate of
Traceability and the Quality Assurance sheet will be submitted. However, if the assured performance
satisfies the required specifications, calibrated instruments controlled by the customer may be used in the
inspections.
(1) Items prepared by the Shimadzu Service Company
(1) Instruments for which the traceability certificate will be submitted
Certificates of traceability are attached to the following instruments controlled by the Shimadzu service
company.

* Thermometer and Recorder


The thermometer and recorder used for the Set Temperature Accuracy check on the Column Oven and
Autosampler Sample Cooler shall have a precision rating of 1.0C in the temperature range required for the
unit inspection (0C to 50C) as a guaranteed performance specification.

* Resistance temperature sensor


The resistance temperature sensor used for the Set Temperature Accuracy check on the Column Oven shall
conform to JIS Class A and have a precision rating of 0.5C in the temperature range required for the unit
inspection (0C to 50C).
* Thermocouple
The thermocouple used for the Set Temperature Accuracy check on the Column Oven and Sample Cooler shall
conform to JIS Class 1 and have a precision rating of 0.6C in the temperature range required for the unit
inspection (0C to 50C).
* DC voltage/current generator
The DC voltage/current generator used for the Chromatopac Hardware Validation check shall have a
precision rating of 0.15% at the test point as a guaranteed performance specification.

* Stopwatch
The stopwatch used for the pump Flow rate Accuracy check shall have a precision rating of 5'30" 0.3 s as a
guaranteed performance specification.

(2) Instrument and reagents for which Quality Assurance sheet is submitted upon request
A Quality Assurance sheet shall be submitted for the following instruments and reagents if requested by the
customer.

* Flasks
The 5 mL measuring flask used for the pump Flow rate Accuracy check.
(The amendment to the Measurement Law on November 1st 1993 excluded this item from the scope of the
Measurement Law.)

* Standard reagents for performance inspection


Acridine and anthracene (10 mg/L acetonitrile) 2 mL x 5 vials [228-32996-02]

2.6 Before Inspections


Follow the procedure below to wash the flow channels before commencing the inspection.
(1) Remove the column and connect a resistor tube instead.
(2) Pump 2-propanol to wash the flow channels.
If the mobile phase previously used has no phase solubility with 2-propanol or if salt deposition is expected
from the buffer solution, wash the flow channels with an appropriate intermediate washing solution, and then
replace it with 2-propanol.
Consider the instrument history when determining the washing solution flow rate and the washing times.

Information> ・Measurement method for the wavelength accuracy check
SPD-20A(V) uses zero-order light and the 486.0 nm and 656.1 nm emission lines of a
deuterium lamp (D2 lamp) for wavelength calibration. Strict calibration at these wavelengths
ensures wavelength accuracy across the UV range. Wavelength calibration using the emission
lines of a deuterium lamp (D2 lamp) is prescribed in the JIS standards, as follows:

5.2.1 Wavelength Scale Calibration (2) Method using lamp emission lines
Measure the light intensity of a low-pressure mercury lamp or deuterium discharge tube
to determine the wavelengths exhibiting the maximum intensity, and compare with the
emission line wavelengths to calibrate the wavelength scale.

This method is used for the wavelength accuracy test in Test 3 Initialization Test.

The wavelength accuracy is ensured by the method above. However, this Wavelength
Accuracy Test (Test 6) confirms the wavelength accuracy using reference wavelengths in the
UV range that is often used for actual analysis.

・Mercury Lamp and Caffeine Pass Criteria


The low-pressure mercury lamp with an emission line at 253.65 nm and caffeine with
maximum absorbance at 272 nm can both be used as the reference wavelengths in the UV
range. Caffeine was also adopted as it is stable in storage and is easy to obtain and handle.
The wavelength accuracy in the UV range is determined by comparing these wavelengths
with the measured peak wavelengths.
The pass criteria for the low-pressure mercury lamp is 2541 nm and for caffeine is 2722
nm. In contrast with the monochromatic absorption line of the mercury lamp, the caffeine
maximum absorbance has some width, which makes accurate measurement of the peak
wavelength difficult. For this reason, the pass criterion is set lower for caffeine.

・Selecting the Mercury Lamp or Caffeine Method


The use of the mercury lamp is recommended to determine the wavelength accuracy more
precisely.
Use the caffeine method if a mercury lamp is unavailable or to make measurements in a
situation similar to actual detector operation.

(Additional Information)
Use the mercury lamp designated by Shimadzu for the wavelength accuracy test. The test
evaluation is based on a direct comparison of the emission line (natural physical constant)
unique to the lamp. The following document prescribes the use of natural physical constants
for traceability.

2) Wet the temperature sensor under the leak sensor with cotton wool soaked in water.
3) Press the [Enter] key after 10 seconds.

(Criteria) “SENSOR GOOD” appears on the display.

Test 7
(Item) Sample cooler set temperature accuracy test

(Outline) Determines the difference between the sample cooler set temperature and actual
temperature.
This test is performed only for the SIL-20AC.

(Procedure) 1) Transfer 1mL of water to a 1.5mL glass sample vial, and set the vial in position 32 of
the standard accessory 1.5mL cooling rack.
2) Mount the rack in the main unit. Insert the sensor into the central vial mounted at
step 1) above. Pass the measurement sensor through the septum and put the sensor tip
in contact with the base of the vial. Ensure that the tip of the temperature
measurement sensor is not bent during these operations. In addition, take care with
the sensor cable routing to ensure that temperature measurement sensor is not pulled
out of the vial.
3) Set the sample cooler temperature to 4C and start temperature control. Leave the
autosampler door closed.
4) After about 1 hour, confirm that the sample cooler temperature has dropped to 4C.
5) Wait at least 30 minutes after the temperature drops to 4C and then read and record
the thermometer temperature. However, record the temperature sensor reading after
correcting it using the temperature measurement deviation value stated in the
temperature sensor inspection report.

(Criteria) At room temperature 30C max. and RH 60% max.

4.11 Data Processing Unit

4.11.1 LCsolution

1. Introduction
This section presents the standard operating procedure (SOP) for Operational Qualification of the
Workstation LCsolution. Refer to the Instruction Manual for details about the operation procedure and to
clarify uncertain points.
Follow section 1-3 Entries and Revisions to change or delete the contents of the procedures.

2. Inspection Procedures
・User Registration of Performer
Before conducting this Operational Qualification, the system administrator must complete user registration
for the Performer who conducts the work (OQ User). Account registration of the Performer is also necessary
to use Windows and Shimadzu application software. Disable the Performer's user registration when all
operations are complete.

・Windows Account Registration


Register two accounts in Windows. Create one account for the analyst who will belong to the group having
normal authority, and the other for the individual who will have administrator authority. The account
pertaining to the analyst authority group is used for checking Shimadzu Corporation application software,
including LCsolution and CLASS-Agent Manager software, while the account pertaining to the
administrator authority group is used for checking the computer etc.

・CLASS-Agent Manager Account Registration


A user with User Registration authority starts the CLASS-Agent Manager and logs in. Click [Tools] -> [User
Administration] to open the [User Administration] screen. If the OQ User is already created but disabled
(displayed gray), select the required user and click [Active/Invalid] to enable this user. Click [Edit] and
check that the authority shown below is set for the user. To register a new user, click [New] and register the
user as shown in the table below.

・LCsolution Account Registration


A user with System Administrator starts the LCsolution User Administration and logs in. Register the
LCsolution Users as shown in the table below. If the OQ user is already created, check that the
administration shown below is set for the user.

(Item) Shimadzu User Authentication Tool connection check

(Outline) Checks the Shimadzu User Authentication Tool connection using database access and log
registration.
* This test is conducted only if the Shimadzu User Authentication Tool is used with
LCsolution.
(Procedure) 1) Open the Windows Control Panel and double-click on the Shimadzu User Authentication
Tool icon to open the main menu.
2) Record the server name displayed at the bottom of the main menu and confirm that the
connected database is correct.

3) Log in to [View event log] as “OQ_USER_2” from the main menu. Set the test date
(Period) and test computer name (PC name) in [Condition]. Search the log and print out
the results.
4) Confirm that the user lockout generated in Test 2-4-4 is registered and that the software
name is “LCPrun.exe”.

10) Enter a password that is not set and click OK.
11) Check that login is not permitted and a message is displayed warning that the password is
incorrect.
12) Repeat steps 8) to 11) for the number of times designated for “Login Attempt Limit” in
the individual user lockout settings in the LCsolution system policy.
13) After the designated numbers of login failures, confirm that OQ_USER_1 is locked out
and cannot log in for the designated lockout time.
14) After the designated lockout time elapses, select LCsolution Postrun again. Enter
“OQ_USER_1” as the login ID and correctly enter the set password.
15) Confirm that login is possible.
16) Exit LCsolution Postrun. Select [Log Browser] from the LCsolution Launcher and login
as “OQ_USER_2”.
17) Display the log. If the Shimadzu User Authentication Tool is used with LCsolution, select
“User Authentication Server Log” as the log type. Otherwise, select [Each Instrument
Log] -> (PC name) -> [None]. Confirm that the login lockout in the procedure above and
the corresponding time appear in the displayed log.
18) If the # of Levels for Duplicate Password Checking and Login Lockout Duration settings
were changed, return them to their original settings.

Launcher. Login with the user ID “OQ_USER_2”.
2) Confirm that the analyzer parameters can be changed in the Instruments Parameters View
in the Data Acquisition screen, and then exit [Offline Editor].
3) Run [Offline Editor] again and login with the user ID “OQ_USER_1”. Confirm that the
Instrument Parameters cannot be changed in the Instruments Parameters View in the Data
Acquisition screen.

(Criteria) OQ_USER_2 can edit Instrument Parameters but OQ_USER_1 cannot edit them.

Test 2-6
(Item) System policy function check
(Outline) Checks the LCsolution system policy function.
The system policy function is verified by checking that the system policy settings for the audit
trail and save/overwriting of methods function correctly.
(Procedure) 1) Run [System Policy] from the LCsolution launcher. Login with the user ID
“OQ_USER_2”.
2) If no check mark appears by “Apply audit trail function when creating method file” or
“Prohibit overwriting of other method files”, enter a check mark and click [OK]. (Revert
to the original settings at the end of the test.)
3) Run [Offline Editor] for Analysis 1 from LCsolution Launcher. Login with the user ID
“OQ_USER_2”. Open the Data Acquisition screen.
4) Select [File] -> [New Method File] to create a new method file. Select [File] -> [Method
File Properties]. (The new file can be created using a template.)
5) Ensure that a check mark appears in the “Apply to the method file” and “Apply to the
data file acquired with this method file” checkboxes on the [Audit Trail] tab on the
displayed screen.
6) Select [File] -> [Save Method File As] and save the method file with the name
OQ_Method_xxx.lcm (where xxx is any number).
7) The following procedure is required for Test 2-7 Audit Trail Check.
Make arbitrary changes to the method parameters for the OQ_Method_xxx.lcm method
file and save it over the existing method file. A screen prompts for the input of the reasons
for the change. Enter “Audit trail test” and save.

and revert all settings to their original values.

(Criteria) A check mark appears for the audit trail settings in a newly created method file.
Overwriting other method files is prohibited.

(Outline) Checks the audit trail functions.

(Procedure) 1) Run [Offline Editor] for Analysis 1 from LCsolution Launcher. Login with the user ID
“OQ_USER_2”. Open the Data Acquisition screen.
2) Open the method file OQ_Method_xxx.lcm saved during Test 2-6.
3) Select [File] -> [Audit Trail] from the menu.
4) Check that the audit trail details (Date&Time, Modification, Reason) are displayed
correctly. Click [Print] on the screen to print the audit trail.

1. Introduction
This section presents the standard operating procedure (SOP) for Operational Qualification of the
Workstation CLASS-VP. Refer to the Instruction Manual for details about the operation procedure and to
clarify uncertain points.
Follow section 1-3 Entries and Revisions to change or delete the contents of the procedures.

2. Inspection Procedures
・User Registration of Performer
Before conducting this Operational Qualification, the system administrator must complete user registration
for the Performer who conducts the work (OQ User). Account registration of the Performer is also necessary
to use Windows and Shimadzu application software. Disable the Performer's user registration when all
operations are complete.

・Windows Account Registration


Register two accounts in Windows. Create one account for the analyst who will belong to the group having
normal authority, and the other for the individual who will have administrator authority. The account
pertaining to the analyst authority group is used for checking Shimadzu Corporation application software,
including CLASS-VP software, while the account pertaining to the administrator authority group is used for
checking the computer etc.

・CLASS-VP Account Registration


A user with System Administrator starts the CLASS-VP System Administration and logs in. Register the
CLASS-VP Users as shown in the table below. If the OQ user is already created, check that the authority
shown below is set for the user.

as Liquid B, C and D, as the mobile phase.
4) Degas the mobile phase.

pump for 10 minutes at 2.000 mL/min.

[Drift test]
4) Set the detector wavelength to 250 nm, response to 10 (2 s), and AUX RANGE to 2
(1.0 AU/V).
5) Set the data-processing unit recording fullscale to 4 mV (4 mAU) FS.
6) After the lamp lights, wait one hour for it to stabilize.
7) Record the baseline for one hour and record the amount of change over the hour as
the drift.

[Noise test]
8) Change the fullscale to 0.5 mV (1 mV) (0.5 mAU (1.0 mAU)) and record the

9) For SPD-20AV or 10AVvp, also conduct measurements at 600 nm wavelength.

equivalent to several times the loop volume.
9) Inject 10 L test sample. If using a manual injector, inject test sample equivalent to
several times the loop volume. Repeat the analysis several times.
10) Calculate the coefficient of variance (CV%) for the retention time and peak area over
the six analyses.
11) If the autosampler is connected, similarly perform six continuous measurements
using 5μL and 50μL injections, and calculate the retention time and peak area
variation coefficients (CV%).

* For a high-pressure gradient system with three pumps, replace Liquid B (pump B) with
Liquid C (pump C) and repeat steps 1) to 10) to compile more data.

2. Inspection Procedures
Test 1
(Item) Noise/drift test
(Outline) Measures the noise and drift while the flow cell is filled with water.
Note: Room temperature fluctuations 2C max. during this test.
(Preparation) (1) The following items and equipment are required:
(a) Water (distilled or ion-replacement)
Use water that has been placed in the measurement room temperature for several
hours in a container with its lid loosely attached. The refractive index of water is
dependent on ambient temperature and the amount of dissolved gases. Use air-
saturated distilled or ion-replacement water at room temperature for this test.
(b) Syringe
(c) Plugs (for inlet and outlet ports) x 2
(2) Turn on the detector. Set the controlled temperature to 40C and leave for at least two
hours with the power turned on. If room temperature exceeds 28C, set the controlled
temperature to (room temperature + 12C)
(Setting Procedure) Press the [func] key several times.
Enter “40.0” for <CELL TEMP>.
Press the [CE] key to return to the initial screen.
(Procedure) 1) Use the syringe to inject water into the cell.
During this process, turn RFLOW ON to replace the reference cell with water.
Ensure that no bubbles enter the flow channels.
2) Insert the plugs in the inlet and outlet ports.

(Outline) Checks the system repeatability of peak area and retention time for different injected
sample volumes.

(Procedure) 1) The following items are required:


a) Mobile phase   Water: Methanol =3: 2
Use degassed HPLC-grade methanol and water that passes through
the on-line degasser.
b) Column Shim-pack VP-ODS or LUNA C18(2) 150mm x 4.6mmID
c) Sample Caffeine solution (250mg/L)
d) Acetonitrile and isopropanol for washing
A column equivalent to the product specified above can also be used.

2) Wash the flow channels with a suitable solvent. At this stage, use a 1.6C coupling with
no column connected.
3) Pump the mobile phase and then replace the coupling with the column.
4) Set the unit parameters as follows:

Detector   AUX RANGE : 2 (1 x 10-3 RIU/V)

Note: Use Integration ON/OFF (LCsolution) as appropriate to avoid detecting peaks

Other parameters are identical to Test 1.
5)Wait until the baseline stabilizes. Inject 10 L mobile phase and check that no peak is
detected on the chromatogram. If using a manual injector, inject mobile phase
equivalent to several times the loop volume.
6) Inject 10 L of caffeine solution (250 mg/L). If using a manual injector, inject test
sample equivalent to several times the loop volume. Repeat the analysis several times.
7) Calculate the coefficient of variance (CV%) for the retention time and peak area over
the six analyses.
