Analyzing samples by ultra-violent visible

absorption

1. Deuterium or tungsten lamp produce constant amount of light sample.

2. The Monochromator selects the required wavelength to be absorbed by the sample.

3. The Beam splitter divides the light into two path, one passes through the sample cell and the
other passes through the reference cell.
4. Both path meets at the detector which compares the two beams and record it.
5. To find the concentration of and an unknown sample the instrument is first set at zero
absorbance using a blank sample.
6. Then a series of standard solution of known concentration are prepared. The absorbance of
each solution and the unknown sample are then measured to at a known maximum
wavelength.
7. A graph of absorbance versus known concentration of the series of standard is plotted to
generate a calibration curve.
8. The calibration curve is the used to determine the concentration of the unknown sample.
