1 System Installation

The Galileo Echo contains delicate electrical and mechanical


components. It must be unpacked and installed by a qualified service
engineer or application specialist.

1.1 Setting up the System

Do not position the equipment so that it is difficult to disconnect the


power supply
To ensure trouble free running of the Galileo Echo system, it is
important to locate the analyser in an appropriate environment.

The dimensions of the Galileo Echo system are:

The Galileo Echo system (Main instrument, fluids module, power


supply and PC) must be placed on a work bench that fits with the
size of the instrument in the laboratory. The bench must be strong
enough to support the weight of all the components and must be a
firm and level surface.

Adequate space should be provided for accessing the system,


both during operation and in case of servicing, a gap of at least 10
cm is required behind the analyser for ventilation.

1.1.1 Tools

The following equipment is required to unpack and set up the


instrument.

o Installation Qualification Document.


o American Standard hex wrench (Allen key) set .050”-3/16”, long
shaft ball end preferred
o Metric hex wrench (Allen key) set, long shaft ball end preferred

o 3/16”, 1/4”, 5/16”, 11/32”, 3/8”, 7/16” nut drivers (set with small
outside diameter will be most useful)
o 3/16”, 1/4", 5/16”, 11/32”, 3/8”, 7/16” open end wrenches (ignition
wrenches)
o Adjustable wrench 0-.94”
o Small channel lock pliers that will open to 1”
o Needle nose pliers w/ teeth
o Wire cutters
o Philips screw drivers #1 and #2, variety of lengths, one #2 w/
minimum 6” shaft
o Slotted screw drivers, variety for screws from #2 to 5/16-18
o Scissors/cutter
o Small weighing scale, Immucor part #0005996
o Tachometer, Immucor part #0005997
o Tempa-DOT thermometers, Immucor part #0087065
o 6 empty 10mL vials, and 7 empty test tubes, supplied with the
instrument

1.1.2 Procedure

The instrument comes in three crates (cardboard packages on one


pallet base), according to the packing list:

o Main Instrument
o Fluids Module and Instrument top cover
o Accessories

It is accompanied by five boxes containing PC and accessories.

1. Unpack the main instrument as follows:


a. Remove the smaller cardboard box from the delivery
package of the main instrument.
b. Open it and remove the protective cardboard frame.
c. Lift the instrument, using two handlers positioned on the
left and right sides of the unit.

Take care – 2 people are required for this step!

2. Place the instrument on the work bench on its back (rear side)
and remove the 7 screws (Flat head) which hold the wood
shipping frame in place. Then stand the instrument on its feet.
3. Remove the 5 shipping brackets (Fixed with 5/32 hex screws).

4. Remove the 3 hex screws which fix the centrifuge on the


instrument chassis (2 at the front and 1 at the rear of the
centrifuge module).

5. Unpack the fluidics unit as follows:

a. Remove the smaller cardboard box from the delivery


package of the fluids module.
b. Open it and remove the protective material.
c. Take the top cover away and put it in a safe place (Do not
fit the cover at this stage).
d. Lift the fluids module and remove the 5 hex screws
holding the wood shipping frame in place.

6. Place the fluids module on the bench to the left of the main
instrument.
7. Unpack the accessories. The box contains the following parts:

o Waste, shuttle and supply bottles (and support frame)


o Power supply
o Tubing and data cables
o Probe assembly
o Sample and reagent racks
o Trays and strip holders
o Camera drivers CD and instrument configuration CD

8. Connect the two components together with the supply tubing and
waste tubing according to the scheme located on the back panel
of the fluidics module. Be sure that the tubing is connected to the
correct orientation (e.g. top to top, bottom to bottom, left to left
and right to right). Also, be sure to connect the probe supply
tubing by screwing it into the back of the Fluidics module and the
back of the instrument.
9. Connect the two components together with the power and data
cables.

11. Place a strip holder that contains two CMT strips with barcode
IDs (prime holder) at the wash prime station (above the wash
manifold support).
12. Place the power supply device in a convenient location and
connect the power supply to the instrument.
13. Connect the power supply to the UPS provided.

The PC and its components come in five boxes:

o Personal Computer
o Touch Screen monitor
o Printer
o UPS
o Hand scanner and additional accessories

1. Unpack the computer and place on counter top.


a. Combine the keyboard cable and handheld scanner and
connect to the back of the computer.
b. Connect the mouse and power cord into the back of the
computer.
c. Do not plug power cord into power source.
d. Check that the supply voltage setting of the computer (Red
Selector near the power connector on the back) is set for the
correct mains voltage (230 volts for EU/110 volts for US).
2. Unpack the monitor and place on top of the Fluidics module.
a. Plug USB cable and blue monitor cable into monitor and the
back of the computer.
b. Connect the power cord into the monitor, but do not plug into
power source.
3. Unpack the printer and place on the counter near the computer.
a. Plug USB cable into printer and the back of the computer.
b. Plug the power cord into the printer, but not into the power
source.
4. Once all cables are plugged into all computer components, plug
the power cords into the power source and turn on all
components.
a. Unpack the UPS and place on the counter.
i. Remove the front cover and verify the battery is
connected.
ii. Plug the UPS into a designated power outlet.

The printer should not be plugged into the provided UPS. It


should be plugged directly into a wall outlet.

5. Insert the install disk into the DVD drive.


a. Double click on the “Setup.exe” file to load the G3Utility.
Confirm the installation prompts.
i. Go to folder C:\Program
Files\Biotek\Immucor G3Utility and create
a shortcut of the file G3Utility.exe.
ii. Move the shortcut to the Desktop.
b. Return to the D:\ drive and double click on the “Echo
Install.exe”.
i. Select the market specific version and press O OKK.
Press O OKK to begin installation.
ii. Press O OKK and Close when installation is complete.
c. From the installation CD open the printer folder and double
clicking on the “Setup.exe” file. Select the 1022n printer and
follow the installation prompts on the screen.
i. When prompted, connect the USB to the back of the
computer.
ii. Select C Coonnttiinnuuee AAnnyyw
waayy.
iii. When prompted to insert disk, click O OKK and then click
the BBrroow
wssee button. Choose the file path
D:\PrinterDriver and click O Oppeenn and then click
OOKK.
iv. Uncheck the Register box and click FFiinniisshh.

If the correct Printer driver is not available on the installation disk, use
the printer driver disk that was provided with the printer.

d. Remove the install CD from the DVD drive.

6. Load the monitor driver CD and confirm the installation directions
on the screen. Select the USB driver from the driver options
menu.
7. Restart the computer.
8. Press F10 when the HP logo appears in order to enter BIOS.
a. Select English and press the Enter key
b. Go to Advanced and select Power-on Options.
c. Disable the F9 prompt line, the F10 prompt line, and the F12
prompt line by pressing the right arrow key at each selected
line.
d. Press F10 to accept the changes
e. Go to Security and select Setup Password.
f. Enter the password OCHE, press the Tab key and re-enter
the password.
g. Press F10 to accept the changes.
h. Go to File and select Save Changes and Exit. Press the
Enter key and press F10 to confirm. At this point the
computer will re-boot.
9. Program the handheld scanner with the provided programming
sheet.
10. Set the date/time and Daylight Saving Time rule.
a. Go to the control panel and double click key on Date and
Time.
b. Click on the Time Zone tab and select the appropriate time
zone for the facility.
c. For geographic areas that do not follow Daylight Savings
Time, uncheck the prompt „Automatically adjust clock for
daylight savings changes‟.
d. Return to the Date & Time tab and adjust the time if
incorrect.
11. Plug the serial cable and the USB cable from the instrument into
the back of the computer.
12. When the USB cable is plugged in, the Found New Hardware
wizard will appear.

a. Select No, not at this time and press the N

be used.

1.1.3 Switching on the machine

1. Power on the instrument.


2. From the desktop, execute the G3 Utility software and open the
C:\ICL Files\IQ-PM folder.
3. Execute the UPLOAD AUTOCAL-CONFIG.ICL file (Click the R Ruunn
FFiillee button in the G3 utility software). This script uploads the
Autocal and Config parameters from the controllers to the PC.
4. Check the Step box in the G3 utility software and click R Ruunn
CCoom mmmaanndd for each step. Verify that the command output has a
status of 0 for each step.
5. Verify that the serial numbers in the command output box match
the serial numbers on the side of the instrument and fluidics
module.

6. Print screen the window.


7. Open the Vessel Config from the Configuration → Fluidics
menu
8. Verify that the „Microliter per Step‟ values are set to 13 for vessel
numbers 4, 5, 8 and 9 (See Figure 1-12 below).

9. If the values are wrong, copy the correct Config data files (Refer
to ETAN-2007-01: Galileo Echo „Config Data‟ files upgrade) and
download the configuration again to the controllers by executing
the DOWNLOAD CONFIG DATA.ICL file.
10. Open the STARTUP.ICL file. Click the R Ruunn FFiillee button in the G3
utility software.

1.2 Post Installation Checks (Qualification)

For the Post Installation Checks you will need to use the G3 Utility
software and ICL files (See chapter 6 'Alignments and Calibrations'
for more details on how to use G3 Utility and ICL files). Sections of
this chapter contain instructions for the verification of mechanical
movements and settings of the Galileo Echo components.

If a command line is blocked or a file is aborted, click on the


AAbboorrtt button and then run the STARTUP.ICL file before
continuing.

1.2.1 Robot (Strip transfer)

1. Place two strips in a strip holder and place it in position 1 of


ambient incubator tray 1. Load three empty strip trays in the
remaining positions of the ambient incubator.
2. Remove the wash prime holder
3. Run the ROBOT AUTOCAL VERIFY.ICL file.
4. Verify that all alignments are correct and that the robot transfers
the strip holder correctly.
5. If adjustments required refer to: chapter 6 'Alignments and
Calibrations – section 1.2.2 Autocalibration'.
6. Replace the wash prime holder on its station.
Shake 7. Place a strip holder in position 1 of tray 1 with a piece of
parameters reflective paper on top of some electric tape near the middle of
the strip holder.
8. Run the TEST SHAKE.ICL file.
9. With a tachometer, observe that the performance is as expected.
10. Record the measured value and compare with acceptable range
in IQ form.

1.2.2 Incubator

1. Place two strips plus one temperature strip in each of 3 strip


holders.
2. Put them in ambient incubator tray 1 from position 1 to 3: 1 for
top still air; 2 for forced air and 3 for bottom still air chambers.

3. Run the LOAD INCUBATOR.ICL file.
4. Wait at least 20 minutes for the strips to come at expected
temperature. Note that you can perform other IQ tasks during
this lapse of time.
5. Run the UNLOAD INCUBATOR.ICL file
6. After the strip holders are returned into the tray; record the
temperature displayed by the temperature strips in the
corresponding sections of the IQ form. Check that the measured
temperatures are within the acceptable range defined in the IQ
form.
7. Log the Tempa-dot serial number and expiration date on the IQ
form.

1.2.3 Centrifuge

1. Place a strip holder with two strips and a piece of reflective paper
stuck on the front of the holder in ambient incubator tray 1
position 1.
2. Open the centrifuge service door

3. Run the CENTRIFUGE TEST.ICL file.


4. With a tachometer verify that the centrifuge is spinning at the
following speeds:
o 1000rpm, 1500rpm, 2300rpm and 2800rpm
5. Record the spinning speed values in the IQ form. Check that
they are within the acceptable range as defined in the IQ form.
6. Open the CALIBRATE CENTRIFUGE.ICL file. Verify that the
Step box is checked. Select line 3 (HOME AXIS) and press the
RRuunn C

7. From the Command Output window, record the Centrifuge Home
Offset (CENTC value) in the IQ form and label the centrifuge with
the CENTC value.

1. Select the Diagnostics menu to open the Camera dialog box

2. Place a strip holder with two strips in ambient incubator position

3. Open the CAMERA ALIGN.ICL file.

6. Under the “Automatic Gain Mode”, click on the scroll control to


turn the AGC Off.
7. Verify the “Shutter Speed Control” is set to 1/60th.
8. Adjust the “Color Level” to 75%.
9. After the color of the image has stabilized, uncheck the “Auto WB
Tracking” box.
10. Use the “Manual Gain Control” to fine tune the brightness level of
the image (Starts with 50%). Increasing Manual Gain Control will
increase the RGB average values (Image brighter). Decreasing
Manual Gain Control will decrease the RGB average values

14. This will generate a Camera Image Calculation table. Verify the
RGB Averages are at 180 ± 5, the Max Diffs are less that 65 and
the Deviation is less than 15. „No errors or warnings detected‟
should be displayed in the output view.
15. If the calculated values are out of range, repeat steps 10 to 13.

Because the camera lamp remains on during camera setup in the


G3Utility, set the RGB Averages within the range of 184-188. This will
allow the Camera calibration through the Echo control software to set
the Averages closer to 180.

16. Adjusting the top chamber camera alignment:


a. Select the Diagnostics menu to open the Move
Robot/Probe dialog box to make adjustments to the
position of the strip holder and strips. On the left side of
this dialog box, there are step size selections. 16 are
equal to one full step. When adjusting the X and Y
positions, use 4 or 16 step sizes.
b. If necessary, adjust the X and Y directions until the four
wells are centred in the image. Refer to Figure 1-18.

c. When moving the Z axis position, this is adjusting the


focus of the strips and 100 step sizes are the most useful.
Inspecting the marks inside the strip will aid in visualizing
whether the strips are in focus or not.
d. If necessary, adjust the Z direction until the X‟s in the
bottom of the wells are in focus. Refer to Figure 1-18.
e. Once all adjustments have been made, select the
Configuration menu and then the Strip Transfer 
Autocal option to open the Strip Transfer Board Autocal
Data window:

17. Adjusting the bottom chamber read:
a. Continuing from the Top chamber adjustment, step all
command lines of the CAMERA ALIGN.ICL file through
the third „TRANSPORT_HOLDER‟ command. The
highlight bar should be located on the last
„TRANSPORT_HOLDER‟ command line. Verify in the
Camera dialog box view that this is located in the bottom
chamber of the Camera module and that the tabs are
visible on the strips.
b. Open the MIRROR ALIGN.ICL file in the G3 Utility and
adjust the mirror rotation so that the tabs and the first full
well are visible on the strips. This is done by un-checking
the Step box and selecting either the first or second
„MOVE_AXIS‟ command from the ICL file. The first
„MOVE_AXIS‟ command will move the image in an
upward direction and the second will move the image in a

9. Run the PROBE ALIGN.ICL file to perform the probe alignment
check. The probe should be centred with the alignment hole at
the top of the rinse station. Adjust the probe if necessary.
10. Remove the strips, sample racks and reagent racks when
complete.
11. Fill the supply bottle with PBS and connect the bottle to the
supply line (tubing and level sensor).
12. Run the LONG SYSTEM PRIME.ICL file.
13. Observe the rinse station and verify that fluid is being evacuated.
14. Observe the wash manifold and verify that fluid is being
dispensed to and aspirated from all wells.
15. Check for leaks at all plumbing connections and inspect tubing
for bubbles.
Peri-pump 16. From the Configuration menu (G3 Utility Options), turn off the
performance gating feature by un-checking the „Instrument Gating‟ option.

1.2.7 Stirrer mechanism

1. Place a reagent vial filled with water and with a stir ball in
position 4 of a reagent rack and insert it in lane 1 of the reagent
bay.
2. Run the TEST STIRRER.ICL file.
3. Observe that the stir ball always stays in contact with the bottom
of the bottle at any speed.
4. If the stir ball looses contact, the clearance between the moving
magnet and the base plate of the reagent bay must be adjusted.

1.3 Checks with the Application Software

A run of the application (Control) software should be performed to verify


additional functional aspects. Refer to the ‘Galileo Echo operating
manual’ on how to use the control software.

1.3.1 Loading Assay Files

1. Go to folder C:\G3\Assay and open the market specific folder


(i.e. EU, Japan, US).
2. Copy the assay files and paste them directly in the Assay folder.

1.3.2 Instrument initialization

1. Install the top cover on the main instrument from the front: -
engage the bottom left and right casters on the corresponding
guide rails and slide the cover all along the rails. It is not
necessary to restart the instrument after installing the cover.
2. Launch the Galileo Echo control software.
3. Login with appropriate service user name and password.
4. Initialize the instrument and verify that all components of
instrument initialization pass.
5. Re-check for leaking at plumbing connections.
6. Go to Tools-General options-Service and enter the instrument
serial number in the correct field.
7. Open the Camera report and view the Calibration image. Verify
that the RGB Averages are 180 ± 5, the Max Diffs are less than
65, and the Deviation is less than 15.

If Camera calibration fails during initialization, open the ‘Camera’


report from the menu. You will have access to the calibration
details: check if the top and bottom chambers are aligned properly
and check that the RGB values are close to 180. Repeat either the
camera or mirror alignment if necessary or the colour settings
adjustment as described in section 1.2.4.

3. Insert disk into DVD-ROM drive. Go to My Computer and right
click on the D:\ drive and select Format.
a. In the Label field, enter the instrument serial number,
backup and the date of the backup. I.e. M001 backup 01-
01-2006. Click SSttaarrtt and then click YYeess to start formatting.
b. Copy the ConfigInstrument folder from C:\Program
Files\Biotek\Immucor G3Utility\ to the backup
disk
c. Copy the Assay folder from C:\G3\ to the backup disk.
d. Copy the Basecode folder from C:\ICL Files\ to
the backup disk.
e. Copy the Image.dll and IIS.dll files from C:\G3\ to the
backup disk.
f. Copy all the configuration files (*.INI) from C:\G3\ to the
backup disk.
4. Label the outside of the disk with the same information as
described in step a. and place the disk in the disk sleeve on the
back of the instrument.

1.3.6 Debugging tool verification

1. Verify that the Dr. Watson debugging tool is turned on by clicking


Start from the Taskbar and clicking Run. Type in drwtsn32 and

2 System de-installation and re-packing

2.1 Preparation

1. Decontaminate the system (if it has been in contact with samples


and/or reagents) with the appropriate maintenance task.
2. Purge the instrument with the corresponding maintenance task.
3. Exit software by selecting „Shut down‟ procedure.
4. Switch off power supply.

2.2 Procedure

1. Remove power supply.


2. Disconnect the serial cable and USB cable from the PC.
3. Repack the computer.
4. Remove the top cover from the main instrument (Will be packed
with Fluidics unit).
5. Remove all the accessories: sample and reagent racks, strip
holders, ambient incubator trays, waste and supply bottles.
Don’t forget to remove the wash prime holder!

6. Remove the probe. Follow countdown steps 6 to 1 as described


in Figure 1-9: Instrument probe installation.
7. Disconnect the data cables from the main instrument and fluids
module.
8. Remove the supply tubing and waste tubing from the main
instrument and fluids module.
9. Pack the accessories as described in Figure 1-7: Accessories
shipping assembly
10. Pack the fluidics unit as described in Figure 1-6: Fluidics unit
shipping assembly
11. Fix the centrifuge module with the 3 hex screws on the
instrument chassis
12. Mount the 5 shipping brackets.
13. Pack the main instrument in respect of Figure 1-1: Main
instrument shipping assembly
