Enzyme

IGCSE GRADE 10 SMA Pribadi Bandung

15 September 2020
Learning objectives:
1. Define the term catalyst as a substance that increases the rate of a chemical reaction and is not changed
by the reaction.
2. Define enzymes as proteins that function as biological catalysts.
3. Describe why enzymes are important in all living organisms in terms of reaction speed necessary to
sustain life.
4. Describe enzyme action with reference to the active site, substrate, enzyme-substrate complex and
product.
5. Describe and explain the specificity of enzymes in terms of the complementary shape and fit of the active
site of an enzyme with the substrate.
6. Investigate, describe and explain the effect of changes in temperature on enzyme activity in terms of
kinetic energy, shape and fit, frequency of effective collisions and denaturation.
7. Investigate, describe and explain the effect of changes in pH on enzyme activity in terms of shape and
fit and denaturation.

Enzyme Model of Action

Enzymes are highly specific:

They must bind to a specific
substrate before they can catalyze a
chemical reaction. Like a key into a
lock, only the correct size and shape
of the substrate (the key) would fit
into the active site (the key hole) of
the enzyme (the lock)
Denaturation of enzyme
is condition in which the shape of the
enzyme is altered, preventing it from
performing its function.
As the temperature rises, the enzymes continue to work faster in this
way;
the higher the temperature, the faster the enzyme movement, and the
greater the kinetic energy until it reaches the optimal temperature
