1. Introduction

Temperature Monitoring System consists with Temperature Monitoring Software and Temp
Recorder, a kind of thermometer, which can measure the temperature of the 96 well PCR
thermo block and analyze the temperature profile to provide the necessary information to
calibrate temperature.
Temperature Monitoring Software can recode and display the temperatures of each well in
the graph at real time by using the Temp Recorder. The measured temperature data are
used for providing proper off-set values when you calibrate the temperature of a ExicyclerTM
96.

2. System Components

please make sure it’s components.

3. Installation

Temperature Monitoring Software and USB Driver (FTDI CDM Driver Package) should be
installed on the computer when you install the product, and installation sequence is
independent of the system used. Please refer this chapter and follow the instructions when
initial installation and reinstallation.

3.1 Software Installation for Temp Recorder


To install Temperature Monitoring software and to check, follow the instruction below.

3.1.1 Software installation


1) After inserting the CD delivered with the system, open the
‘TempMonitoring‘ folder and double-click ‘TempMonitoring.exe’.

2) When the ‘Temperature Monitoring’ Install Shield Wizard starts, click ‘Next’ to
start the Installation. Then, the installation will proceed.

3) Click ‘Install’ button.


If you need to change any of installation setting, click ‘Back’. Click
‘Cancel’ to exit the Wizard.

4) Wait the installation finished.

5) When the installation is complete, click ‘Finish’ to finish the Installation Shield
Wizard.

3.1.2 Checking Software Installation


[On the desktop]
The new icon (Temperature Monitoring.exe) like below is created on the
desktop when the installation process is finished.

[On the Start menu]


The new menu (Temperature Monitoring.exe) like below is created on the
Start menu when the installation process is finished.

[From ‘Add or Remove Programs’ menu]


Select ‘Control Panel > Add or Remove Programs’. Then you can check the
list of ‘TempMonitoring’.

3.2 USB Driver(FTDI CDM Driver) Installation


To install USB Driver (FTDI CDM Driver) properly and to check it, follow the instruction
below.

3.2.1 FTDI CDM Driver Installation


1) After inserting the CD delivered with the system, open the ‘USB Driver’
folder and double-click ‘CDM20824_Setup.exe’.

2) At first step, a command line appears and displays ‘Installing driver....’

3) As soon, ‘FTDI CDM Driver Installation process completed.’ message is


displayed after finishing USB Driver installation.

3.2.2 ‘USB Serial Port’ driver Installation


1) To install ‘USB Serial Port’ driver, connect the USB data cable to the USB
ports between the computer and the Temp Recorder. Then, the driver is
installed automatically.

2) When the driver is installed completely, you can check the port number for
the driver like the below picture.

3.2.3 Checking ‘USB Serial Port’ driver installation


1) Select ‘Control Panel > System > Hardware > Device Manager’.
2) Select ‘Ports (COM & LPT)’ item.
3) Then, you can check the same port number for the driver which is displayed
above the picture.

4. Procedure for the Monitoring Temperature

The Temperature Monitoring software can measure the temperature of the 96 well PCR
thermo block by using a Temp Recorder and recode the temperatures of each well in real
time. The measured temperature data are used for temperature calibration.

4.1 Operating the Temperature Monitoring Software


This chapter explains how to use the Temperature Monitoring software for monitoring a
temperature interested.

4.1.2 Connecting to the Temp Recorder


1) Select the serial port number installed for USB communication at the ‘Serial
Port’ item like below.

2) Click ‘Connect’ button for connecting to the Temp Recorder.

4.1.3 Start Monitoring Temperature


1) Click ‘Run’ button to start monitoring temperature.

2) Enter a file name to record a temperature.

3) Then, it shows the temperature graph in real time.

4.1.4 Finishing the temperature monitoring.


To stop monitoring temperature, click ‘Stop’ button.

4.2 Analysis of the Temperature Profiles Recorded


All of the recorded temperature profiles can be analyzed at the Temperature Monitoring
software. This chapter explains how a specific temperature area can be selected, saved,
and converted to an image file.

4.2.2 Executing the Function of Temperature Analysis.


Select ‘Menu > Analysis > Temperature Analysis’ menu or click ‘Analysis’ button
at the toolbar of the software. Then, a new window titled as Temperature Analysis
appears.

4.2.3 Loading a Temperature Data


1) To load a temperature data, click ‘...’ button located right on the box of ‘CSV
data file’.

2) Select a temperature file to analyze.

3) Then, it shows temperature graph.

4.2.4 Analyzing a Temperature Profile


1) Select an area by dragging a mouse which you want to analyze. At that time, a
red vertical line indicates the start point of the area.

2) During the dragging, the red line shows the end point of the area you selected.

3) Then, it displays the standard deviation, accuracy, and uniformity of the selected
area at the ‘Temperature Information’ item.

[Adjustment of Scale View of X-axis]


When you need to change the scale of X-axis, double click X-axis and enter a
new value to ‘ScaleView Size’ box.

[Cancelation of the Selected Area]


1) Clearing all the selected area
Click the right button of the mouse at the ‘Temperature Graph’ window and select
‘Clear all striplines’ menu.

2) Deleting a selected area


Click the right button of the mouse at the ‘Temperature Graph’ window and select
an area you want to delete at the submenu of the ‘Clear stripline’.

4.2.5 Saving the Analyzed Result


All of the analyzed results of ‘Temperature Graph’ and ‘Temperature Information’
can be store as a file type. To copy the result, click ‘Copy Analysis Results’ button
or click ‘Save Analysis Results’ button to save it image.

5. Factory Set Procedure

To maintain the Temperature Monitoring System properly and to monitor a temperature


accurately, you have to keep the information of the Temp Recorder safely and calibrate the
probe sensor periodically.
This chapter explains how to access the factory setting values and calibrate the system
using the Temperature Monitoring software, so that it is prohibited to change the below
values without any approval from Bioneer corporation.

5.1 User Authentication in Factory Set


To access the Factory Set menu, follow the instruction below.

5.1.1 User Authentication


1) Select ‘Menu > Factory Set > Password’ menu.

2) Enter a password and click ’OK’ button.

5.2 System Calibration


Following the below procedures, you can calibrate a Temp Recorder and a Probe
Sensor by adjusting the resistances of them.
To calibrate them, you must use a special chamber which can control both
constant temperature and constant humidity and set target points of its temperature
at 30, 50, and 90 Celsius degree.

5.2.2 System Resistance Calibration


1) Performing System Resistance Calibration
Click ‘System Resistance Calibration’ button of ‘System Resistance
Information’ item. Then, it shows the information of the 8 resistance of a
Temp Recorder.

2) Finishing System Resistance Calibration


Finishing the calibration, it shows the message below. Click ‘OK’.

5.2.3 System Sensor Calibration


1) Performing System Sensor Calibration
① Set the temperature of the chamber 30 Celsius degree.
② Activate the chamber function of constant temperature and constant humidity.
③ Wait the stabilization of the chamber until constant temperature and
constant humidity.
④ Click ‘System Sensor Calibration’ button to start calibration of the probe
sensors. Then, after a moment it shows the calibrated information of the 8
sensors.
⑤ As the same manner of the previous step ①~④, calibrate 50 and 90
Celsius degree again.

2) Finishing System Sensor Calibration


Finishing the calibration, it shows “System Sensor calibrate completed.”
message. Click ‘OK’.

5.3 System Information


This chapter explains how to check the system ID and the calibration information
and how to manage the Temp Recorder.

5.3.2 Reading System Information

Click ‘Read System Information’ button at the ‘System Information’ item.
Then, it recalls the system information from the memory of the Temp
Recorder.

[Completion of reading System Information]


After reading the system information, it shows “System Information read
completed.” message. Click ‘OK’.

5.3.3 Updating System Information


[Updating System Information]
1) To update system information, change the values of ‘System
Information’ and ‘Calibration Information’ items.
2) Click ‘Update System Information’ button. Then, the modified values at
‘step 1)’ are upload to the memory of the Temp Recorder.

[Completion of updating System Information]


After updating the system information, it shows “System Information update
completed.” message. Click ‘OK’.

5.3.4 Initializing System Information


[Initializing System Information]
Click ‘Initialize System Information’ button at the ‘System Information’ item.
Then, it initializes the memory of the Temp Recorder.
* If you initialize the memory, the stored calibration data will be removed.

5.3.5 Opening System Information


1) Click ‘Open System Information’ button at the ‘System Information’ item.
2) Select a CFG file (*.cfg) which has a system information of the Temp
Recorder and click ‘OK’.
(Then, it shows all the values of ‘System Information’ and ‘Calibration
Information’ item.)

5.3.6 Saving System Information


To save the data of ‘System Information’ and ‘Calibration Information’
items to a CFG file type (*.cfg), click ‘Save System Information’ button.

6. Troubleshooting

6.1 Network Connection Problem


[Problem Cause]
If you have problems with USB Driver, network connection may not be possible
properly. Such the case, you will meet ‘Network disconnected. Please check
Network Connection and try again.’ message. To find out the problem, select a
serial port and then click ‘Connect’ button.

[Solution]
To solve this problem, you must install the USB Driver again.

1) After inserting the CD delivered with the system, open the ‘USB
Driver‘ folder and double-click ‘CDM20824_Setup.exe’.

2) At first step, a command line appears and displays ‘Installing driver....’

Appendix 1.
ExicyclerTM 96 Temperature & Power Calibration Procedure

This appendix explains how to calibrate Exicycler 96 system using the Temperature
Monitoring software and the ExiHardWareSet software.

To calibrate Exicycler 96 system, you have to follow the Temperature Calibration step
first, the Power Calibration step second, and the Temperature Record step last.
At the Temperature Calibration step first, reference temperature values for NTC sensors
are calculated through the Temperature Monitoring software. And at the Power Calibration
step second and the Temperature Record step last, the calibrated values are evaluated for
whether the values are correctly working or not.

1. Required tools and software

To calibrate Exicycler 96 system properly, the tools and software like the below are
required. Before proceeding the calibration, please check out the tools and software.

1) Removing the side panels of Exicycler 96

Remove the bolts first which are in the rear side of Exicycler 96 and remove
the side panels of it next.

2) Connecting the serial cable for download to the PC


Connect the serial cable for download to the PC like the below.

3) Connecting the serial cable for download to the ExicyclerTM96


- Remove the P1011 cable from the ExicyclerTM 96 main board.

2) Select a serial port at the ‘Configuration & Run’ menu.

3. Initializing the Calibration Data


This chapter explains how to initialize the calibration data for the reference temperature

and the Power from the memory of the Exicycler 96 using ExiHardwareSet software.

1) Click ‘Calibration’ button.


2) To recall the calibration data from the memory, click ‘Read Offset Settings’ button.
3) Click ‘Initialize Offset Settings’ button. Then, all the calibration data are removed
from the memory.

1) Click ‘Normal’ button at the ExiHardwareSet software.
2) Click ‘Temp Calibration’ button so that the first step can be processed.

5. Analyzing the Temperature Calibration Result

After completing the Temperature Calibration, Exicycler 96 gives two short beeps
sound. Then, stopping the monitoring temperature, analyze the result to calculate the
reference temperature values. For detailed analysis, please refer to page 13-17.

6. Applying the Calculated Result of the Temperature Calibration


Apply the calculated result from the Temperature Monitoring software to the
ExiHarewareSet software, following the below procedures.
1) Click ‘Calibration’ button at the ExiHarewareSet software.
2) Click ‘Read Offset Settings’ button. Then, it shows the reference temperature
values and the resistance of NTC for them.

3) Copy the calculated result values from the Temperature Monitoring and then,
paste them to the Offset item of the ExiHarewareSet software like the below
picture. (S0 = Left / S1 = Center / S2 = Right)

4) Click ‘Write OffSet’ button. Then all the calculated values for the references
temperature are stored to the memory of the ExicyclerTM96.

7. Performing the Power Calibration


7.1. Performing the Power Calibration step using the ExiHardwareSet software
1) Click ‘Normal’ button.
2) Click ‘Power Calibration’ button so that the second step can be processed.

8. Analyzing the Power Calibration Result

After completing the Power Calibration, Exicycler 96 gives two short beeps sound.
Then, stopping the monitoring temperature, analyze the result of Power Calibration.
1) Click ‘Calibration’ button.
2) Click ‘Read Offset Settings’ button. Then, it shows the calculated power values for

the Exicycler 96 including the reference temperature values and the resistance
of NTC for them also.
3) Click ‘Write Power’ button. Then all the calculated power values are stored to the

memory of the Exicycler 96.

9. Performing the Temperature Record


This is the final step for checking all the calculated values acquired from the previous
steps. In order to verify the accuracy of the temperature, this Temperature Record step
is required.

9.1. Performing the Temperature Record step using the ExiHardwareSet software
1) Click ‘Normal’ button.
2) Click ‘Temp Record’ button so that the final step can be processed.

10. Checking the Temperature Record result

After completing the Temperature Calibration, Exicycler 96 gives two short beeps
sound. Then, stopping the monitoring temperature, analyze the result for whether this

result is in the range of the specification of the Exicycler 96 or not. If not so, return
back to the first step [4. Performing the Temperature Calibration].
