Activity1: The effect of pH on the growth of an aquatic plant

Introduction:
Water spinach (Ipomea aquatica), locally called “kangkong”, is an herbaceous
aquatic or semi-aquatic perennial plant of the tropics and subtropics. It has a
creeping growth habit but may grow erect in water. There are two major cultivars of
water spinach. Ching Quat, also known as “green stem” water spinach, has narrow
leaves and white flowers and is usually grown in moist soils (worldcrops.org).

In this experiment, you will determine the pH tolerance of water spinach.

Note: Observe necessary precaution when conducting the


activity.

Materials
- 2 glass jars (~500mL)
- 6 stems of water spinach
- 1L distilled water
- vinegar (1⁄2 cup)

A. Procedure:

1. Label the jars no. 1 and 2


2. Prepare your treatments

3. Fill each jar with the respective culture medium

4. Prepare your test plants. Select 6 stems of water spinach with at least similar
characteristics (e.g. diameter, appearance, color, maturity, etc.). Remove all leaves and
roots. Cut all the stems with the same length (8 inches).
5. In each jar, put 3 stems, representing 3 replicates in each treatment.
6. Put the glass jars near the window.

B. Data Collection

1. Determine the pH (by researching on the internet) of distilled water and vinegar.
2. In every observation day, document your experiment/set-up.
3. Observe and write the initial conditions of the test plants in each jar (Day 0).
4. Observe the plants in each treatment every other day for a total of 9 days (Day 2-8) and
record your observations:
b. root formation – count the number of roots that grow in each plant
c. bud formation – count the number of buds that grow in each plant

Data analysis and discussion:


Analyze and discuss the data you collected on root and bud formation.

In this experiment I observed that Ph level can affect the growth of plant. The pH level
of vinegar is around 2.5, while the pH level of distilled water is 7. Water with equal
parts acid and alkaline is considered to be neutral, or have a pH balance of 7.0.
When water is more acidic, that number goes below seven. For water that is more
alkaline, the number goes above seven. The pH balance of water affects the acid. If
water is too acidic, calcium, magnesium and potassium levels are reduced.
Calcium is required for cell growth, magnesium for chlorophyll formation and
potassium for synthesizing proteins. If water is too alkaline, calcium builds up,
effectively cutting off the flow of nutrients to plants' roots. The kangkong on
treatment 1 died because the vinegar is acidic that causes the plant to not grow. The
kangkong on treatment 2 did not die because the distilled water is not acidic that
cause roots and buds’ form.

1.What are the physical changes you observed from the stem used in each treatment for the
duration of the experiment?

I observed that the stem on treatment 1 turned brown, while leaves and roots grew on treatment 2.

2. Identify the independent, dependent, controlled variables and possible extraneous


variables in the experiment.

The indipendent variables are the distilled water, and the vinegar. The dependent variable is the
growth of the kangkong. The controlled variables are the amount of water water, amount of sunlight
and wind.

Sponge Bob notices that his pal Gary is suffering from slimotosis, which
occurs when the shell develops a nasty slime and gives off a horrible
odor. His friend Patrick tells him that rubbing seaweed on the shell is the
perfect cure, while Sandy says that drinking Dr. Kelp will be a better cure.
Sponge bob decides to test this cure by rubbing Gary with seaweed for 1
week and having him drink Dr. Kelp. After a week of treatment, the slime is
gone and Gary’s shell smells better.
