Figure 1: The relationship between Absorbance and Concentration

Equation: y = 74.835x + 0.1384
R-squared value: R²=0.9394

2. Determination of concentration of Unknown sample.

Linear equation: y = 74.835x + 0.1384
Concentration of unknown 3: x = (y – 0.1384) / 74.835
Concentration of unknown 3 followed by absorbances are:
Absorbance Concentration (mg/mL)
0.699 ( 0.699 – 0.1384)/ 74.835 = 0.00749
0.706 ( 0.706 – 0.1384) / 74.835 = 0.00758
0.705 ( 0.705 – 0.1384) / 74.835= 0.00757
Mean : x̅ =
∑ Xi = ( 0.00749 + 0.00758 + 0.00757) / 3 = 0.007549052≈ 0.00755
n
Therefore, the concentration of the unknown sample 3 is 0. 00755 mg/mL
3. Question
What can you conclude about the reproducibility of the experiment?
The R-squared value and the coefficient of variation (C.V) will be used to assess the
reproducibility of this experiment.
First, R-squared (R2) is a statistical measure that represents the coefficient of multiple
determination in a population of several samples or number of repeated experiment. The
ideal R-squared value of the regression equation is 1. If the result is closed to 1, the
experiment is more accurate. In this experiment, after determining the standard cure, the
R square value was obtained to be 0.9394, which is closed to 1. Therefore, with this
value, the relationship between Absorbance and Concentration are strongly related and
reliable, and indicating that the experiment is reproducible.
Second, using the data of unknown 3, we can calculate the C.V
We have:
Mean:

x̅ =
∑ Xi = ( 0.00749 + 0.00758 + 0.00571) / 3 = 0.007546667 ≈ 0.00755 ( mg/mL)
n

Standard deviation:

√ ∑ ( x−¿ x )2 ¿ =

2 2 2
s= (0.00749−0.00755) +(0.00758−0.00755) + ( 0.00757−0.00755 ) =
n−1 3−1
4.95x10 -5

Standard deviation 4.95 x 10−5

C.V (%) = ∗100 = * 100 = 0.66%
Mean 0.00755

Also, the coefficient of variation (C.V) can be used to demonstrate reproducibility. To

ensure that the experiment is reproducible, this figure should be less than 1%. The
calculated coefficient of variance is 0.66 %, which means the reproducibility of this
experiment is acceptable.
