Bacteria Definition

“Bacteria are unicellular organisms belonging to the prokaryotic group where


the organisms lack a few organelles and a true nucleus”.

Preparation of Wet Mount of Bacteria to Observing its

The aim of this experiment is to observe the natural shape, size and
arrangement of bacteria in living condition.

Gram Stain
What is a Gram stain?
A Gram stain is a test that checks for bacteria at the site of a suspected infection such as the thorat,
lungs, genitals, or in skin wounds. Gram stains may also be used to check for bacteria in certain body
fluids, such as blood or urine.

There are two main categories of bacterial infections: Gram-positive and Gram-negative.

Examples of gram-positive organisms are:


 Cocci: Staphylococcus species, and Streptococcus species
 Bacilli: Corynebacterium species, Clostridium species, and Listeria species

Examples of gram-negative organisms are


 Cocci: Neisseria gonorrheae, Neisseria meningitidis, and Moraxella species
 Bacilli: Escherichia coil, Pseudomonas species, Proteus species, and Klebsiella species

Observations (Under Oil-immersion Objective):

Spherical (coccus)

Rod-shaped (bacilli)
