Optical Tweezers with Linear and Nonlinear Microspectroscopy and Microscopy.

We decided to add spectroscopy to the optical tweezer system to perform


spectroscopy at the same time we manipulate particles and living cells, allowing us to
observe chemical reactions and mechanical properties changes in real time. For this we
incorporated a home made confocal spectrometer to our optical tweezer setup and succeed
to observe Raman, Hyper Rayleigh (Second Harmonic) and Hyper Raman scattering, two-
photon absorption (TPA) excited photoluminescence and force spectroscopy. The hyper
Raman and Rayleigh term comes from the expansion of the second order susceptibility

P( E ) = Po + χ (1) (q j ) : E + χ ( 2) (q j ) : EE + ... at the 2ω and 2ω ± ω j frequencies, and
their tensors are different from the Raman and Rayleigh ones, with opposite parity. In the
past hyper Raman has been observed only with giant picosecond pulses after averaging
times of hours. We observed it with seconds because our 80 MHz femtosecond Ti:sapphire
laser averages 107 more pulses than a tipical 10 Hz system used before. Hyper Rayleigh and
Raman are two photon processes and as such they are intrinsically confocal techniques with
the same advantage of the multiphoton confocal microscopy. With this system we have
been able to observe the Raman spectra of ZnSe and trapped polystirene sphere and red
blood cells (RBC) at 785 nm region, Figura 1, TPA excited luminescence at 500 nm region
and the hyper Rayleigh and Hyper Raman scattering at 400 nm of ZnSe, Figura 2. The TPA
excited luminescence spectra of a trapped DAPI stained latex microsphere clearly show the
“Morphology Dependent Resonances”, Figura 3., with good agreement with Mie
Scattering calculations for more than 15 peaks. At the same time these resonances increase
the light scattering they increase the optical force, therefore we decided to revisited 1977
Ashkin’s force spectroscopy experiment [PRL 38, 1351,1977]. By that time Ashkin used an
unstable levitation system to observe the optical forces at microspheres. We, instead, used a
stable single beam optical trap perturbed by a second tunable Ti:sapphire laser to perform
the force spectroscopy showed in Figura 4. Because we have two laser beams we can
position each one respect to the other individualy and observe the coupling to the
whispering gallery modes as a function of polarization and position. The three spectrum
showed correspond to the Ti:sapphire laser beam positioned at the edge of the microsphere
but with different polarization. As we can see for the (a) case only the TE modes are
excited, for (b) case only the TM modes and for the (c) we excited both modes. This is the
same spectrum we obtain when the sphere is illuminated by a plane wave as shown in
Figura 5. For this case the Ti:sapphire laser focus was positioned well above the Nd:Yag
focus to guarantee the ilumination field is like a plane wave.
Optical Tweezers for Red Blood Cells Rheoscopy

In this area the two important facts were Marcelo Brandão’s PhD thesis defense and the
use of double optical tweezer to understand red blood cells adhesion. Brandão’s thesis
included the results of the RBC elasticity measurements and comparison with normal
controls (HbAA) several diseases an conditons. The diseases and conditions studied were:
homozygous (HbSS) and heterozygous (HbAS) sickle cell anemia, hereditary spherocytosis
and iron deficiency anemia. These results showed that the laser optical tweezers technique
is able to detect differences in HbS red blood cells from subjects taking hydroxyurea, as
well as to differentiate red blood cells from normal controls and HbAS, sickle-cell trait,
indicating that this is a very sensitive method and can be applied for detection of drug-
response in sickle cell disease. The thesis also included the studies of storage of gamma
irradiated blood bags. The gamma irradiation is used to prevent transfusion associated
graft-versus-host disease in immunodeficient patients and this study aim was to understand
the hematological and biochemical changes induced by the irraditaion. The changes in
elasticity after 20 storage days was so high compared to non irradiated RBC, that we
suggested the rule to use irradiated blood bags up to 35 storage days to be reviewed. The
double tweezer was used to observe the adhesion of RBC’s. Figures 5.a, 5.b and 5.c show
a sequence of 6 RBC’s sticked together (rouleaux) under the forces of the two optical
tweezers. We observe that they slide one over the other very easily but they adhere very
strongly at the edges. The RBC’s adherence is an important and not well understood
phenomena in which electrostactic, RBC are negativelly charged, and van der waal’s
chemical forces play their roles. The ph and dielectric constant of the solution they are
embedded are important from the electrostatic point of view while membrane proteins and
antibodies represent the glue factor. For the first time we have a tool able to measure the
force as function of the displacement between two RBC’s and to check the existing models
and even developed a new one for the adherence force between cells and microorganisms.

Physical and Chemical Proprieties of dental hard tissue

Enamel surfaces irradiated with several wavelength and pulse widths are under
investigation in order to develop new processes for caries prevention using lasers. [Ca]/[P],
fluoride uptake, morphological and microbiological studies are the main analysis methods.
The infrared spectroscopy results show that the erbium laser (2,94µm) changes the structure
and composition of the organic matrix, OH- radical and the water composition in the
irradiated dentin. The crystalline structure of irradiated enamel with holmium laser (2.065
µm) indicates the existence of a mixture of two crystalline phases: hydroxyapatite and
tetracalcium phosphate ( Figura 7), that is more resistant to the acid produced by cariogenic
microorganisms. The dentin discoloration after thermal treatment can be due to the non-
enzymatic browning, collagen denaturation or oxidation of some chemical component;
while in the enamel, a observed white pattern is observed after thermal treatment occurs
because the water loss and consequent increase in the light scattering by the tissue.

Temperature rise due to CuHBr laser irradiation (λ=510 nm) in occlusal surface was
imaged using an IR thermographic camera. Figura 8 shows images for four instants (1 s (a),
5 s (b), 10 s (c) and 15 s (d)) during irradiation with 38.5 kW/cm2. The green marker
highlights areas where the temperature lies in the interval between 65o-70o C and suggests
isotropic heat propagation. Figura 9 shows the temperature profile along the line drawn in
the infrared image of the tooth irradiated with 5,7 kW/cm2. Image (a) refers to the initial
instant and (b), after 15 seconds of irradiation.

As mentioned in Laser Engineering and Instrumentation activities, in vitro studies are
being made on enamel irradiated with high intensity ultrashort pulses (femtosecond and
picosecond) (Figura 10). Morphological, thermal and mass removal processes are under
investigation in both temporal regimes.

Optical Diagnostic Methods


- Optical proprieties of the caries lesion differ from sound tissue. The fluorescence is used
as a new optical diagnostic method for incipient caries, with higher sensibility and
specificity than the current clinical analysis. Our aim is to characterize the demineralization
and remineralization processes of enamel lesions by optical methods. The artificial lesions
will be analysed at different levels of demineralization and remineralization process by
spectroscopy, confocal microscopy and transverse microradiography.

- Optical Characterization Of Biological Tissue By Optical Coherence Tomography (OCT)


Technique: OCT is a new non invasive optical detection technique, based in a Michelson
interferometer. It was possible to produce a tomogram of dentine-enamel junction and the
OCT system accurately depicts dental tissue, being able to detect early caries in its structure
with a lesion depth of 50µm, analyzing the tomogram of the lesion. The principal problem
of this system was the linear translator that needs a high resolution and high precision
movements (<5 µm) Figura 12.
- Pulp Vitality Test By The Laser Doppler Flowmetry. A method to diagnose the pulp
vitality state was developed, using the laser Doppler flowmetry (Figura 11). The method is
based on the discrimination of an unconventional haemodynamic parameter, the percentage
variation of the pulp flow, defined as the percentage of the pulp blood flow variation of the
interrogated tooth when compared to a homologue one. This haemodynamic parameter is
less sensitive to factors widely known as drawbacks of the technique.

Low Intensity Lasers –research


- Low-intensity laser therapy (LILT) on blood microcirculation during skin wound
healing was studied by laser Doppler flowmetry. The results indicated that there is
no immediate effect due to the laser radiation on microcirculation, although an
increasing of blood flow was observed on the 7th day post-wounding in irradiated
specimens. Also, skin samples irradiated with linearly parallel polarized He-Ne
laser beam relative to a preferential direction on animal’s back showed collagen
fibers more organized than control skin and burned skin samples presents a
significant higher attenuation coefficient than health skin samples. The attenuation
coefficient changes during skin wound repair process in a different manner if the
lesion is treated with laser or not.
- The findings in bone repair after titanium implant surgeries following LILT showed
that on the 21th and 42th days post-operatory, irradiated sites presented statistically
significant implant removal torque values higher than those from non-irradiated
sites.
- Partial results in caries prevention in vivo using LILT associated to topic fluoride
indicated that there are no morphological and chemical changes on enamel surface
after laser therapy.
- The development of new photosensitizers and the improvement of existing ones for
antimicrobial photodynamic therapy (PDT), most of them for odontopathies are
under investigation. Studies on peri-implantitis in dogs suggest that PDT using
azulene is effective to kill periodontopathogenic bacteria.

Low Intensity Lasers clinical research
- Study about LILT in the prevention of oral mucositis in patients undergoing bone
marrow transplants was carried out. The results showed that the diode 650nm laser
decreased the severity of oral mucositis as well as the degree the pain when used as
a preventative therapy in patients undergoing bone marrow transplants. This work
was developed as a cooperation between São Paulo branch and São Carlos branch
of this CEPOF.
- Effects of LILT on the velocity of canine tooth movement and consequently bone
remodeling were investigated The results indicated that all patients showed
significant acceleration of the distalization velocity on the side treated with LILT
when compared to the control.
- Effects of LILT on the ultrastructure of dentine-pulp interface after conventionally
prepared class I cavity preparation were studied. The findings suggest that laser
irradiation accelerates the recovery of the dental structures involved in the cavity
preparation at the pre-dentine level.
- Results about LILT using λ= 660 nm and λ= 780 nm on hypersensitive dentine
showed that there are not statistical significant differences between wavelengths in
pain relief after treatment

High Intensity Lasers in Restorative Dentistry, Periodontics and General Dentistry


In the study of caries prevention with Er,Cr:YSGG laser irradiation samples of
bovine enamel were irradiated with Er,Cr:YSGG laser at fluences of 2,8 J/cm2, 5,6 J/cm2
and 8,5 J/cm2. In these condition, the EDX analysis showed no significant changes in Ca/P
ratio; a maximum rinse of 1o C was detected at analysis of pulpar chamber temperature and
SEM observation showed some melted areas around microablation ones.
Adhesion and groth of cultured human gingival fibroblasts on root surfaces treated
by either irradiation with Er:YAG laser using a periodontal fiber or curet were compared,
showing that 3J/cm2 (twice) promoted faster adhesion and growth than surfaces treated with
either root planning or higher irradiation conditions.
Temperature rise was evaluated using an IR thermographic camera (ThermaCam
SC3000, Flir Systems, USA) on a tooth recovered (a) and unrecovered (b) with a H2O2
bleaching gel and irradiated with 27-46 W/cm2 of a diode laser (λ=805nm) during 30s.
Figura 13 shows the images for the highest intensity and results indicated that these
parameters are safe for teeth vitality and the temperature gradient is lower in the recovered
sample.

To measure the pulp blood flow during orthodontic forces it was developed a new
method to investigate the supply state of the pulp microcirculation during and after
orthodontic forces using the laser Doppler flowmetry. The method is based on the analysis
of the percentage of pulp flow variation during the tooth displacement, compared to the
initial flow, prior to the force application. The method allow the flow to be investigated
during long periods, indicating that the flow is substantially reduced immediately after the
force is applied (due to the strangulation of vessels), and remains reduced during at least 72
hours due to the resulting pulp inflammation, induced by the periodontic inflammation
process.

Lasers in Endodontics

In the last past decade laser technology has been considered an efficient clinical tool
in Endodontic treatment. Many studies using different wavelengths have shown the
microbial reduction capacity of high intensity power laser irradiation and recently even the
low power laser energy used with the concept of photodynamic therapy, associated with
specific dyes interacting with specific wavelengths has been efficient for the elimination of
bacteria in infected root canals. Previous studies have established save laser parameters so
that the temperature on the external root surface during the irradiation does not increase
more than 10 0C avoiding thermal damage to surrounding tissues. Furthermore, many other
in vitro research pointed out the morphologic changes of dentine surface after high intensity
laser intra canal irradiation as well as the influence of those alteration to the sealing of root
filling by dye apical infiltration. Finally, studies of periapical surgeries with laser
technology have gave the support to the clinical use. The Er:YAG laser is optimized for
hard tissue removal, performing the soft tissue incision, the osteotomy and root apical
resection, as well as the high intensity diode and Nd:YAG lasers perform the microbial
disinfection in the surgical site.

COLLABORARIONS INSIDE THE CENTER

A clinical study on oral mucosites prevention in 70 patients submitted to bone


marrow transplantation was carried out as a collaboration of IPEN, FOUSP, Hospital
Sírio Libanês, in São Paulo and Fred Hutchinson Hospital in Seatlle-USA . The laser
system used in this study was developed and customized by one of the companies in
São Carlos (MM Optics). This study was presented as a dissertation at IPEN-SP branch
of CEPOF.
A PhD study (Luciana Almeida Lopes- São Carlos branch) using culture cell
irradiated with low intensity laser is being conducted in São Paulo branch of the Center,
as a collaboration with Prof. Márcia Marques, from School of Dentistry (FOUSP),
facilities that does not exist in São Carlos.

1. Fluorescence Spectroscopy to Value Humification Degree of Organic Matter


(OM)

The humification process increases semiquinone-type free radical concentration (SFRC)


in humus and their quantification by electron spin resonance spectroscopy (ESR) has
been a good indicator of humification degree of soil organic matter. As an alternative to
relatively complex and expensive ESR spectroscopy, fluorescence spectroscopy was
used in this study to evaluate the humification degree of humic acids (HA) extracted
from Brazilian soils under different land use, tillage or cropping system. Three
fluorescence humification indexes of HA were calculated based on work done by
Zsolnay et al. (1999), Kalbitz et al. (1999), and Milori et al. (2002) (Fig.1).

The comparison of the fluorescence and ESR data clearly showed that our proposal to
evaluate humification is efficient. The increasing of resonance with structures related to
humification process (excitation in 465 nm) produces an emission spectra that is
characteristic and selective. Certainly the humification degree has a large interest in
agricultural systems. Also, it is an accessible analysis of soil organic matter stability,
because it measures stable carbon. This can be very useful for studies on carbon
sequestration by soils when associated with CO2 emission.

In conclusion, fluorescence spectroscopy demonstrates to be a useful technique to


evaluate land use and soil management effects on humification degree of soil HA. The
changes in the structure of the soil HA after a short time such as 5 years under new
management practices were easily detected by fluorescence. Compared with ESR,
fluorescence spectroscopy is relatively simple with low cost tools for studying soil
humic substances.

2. Fluorescence Induced by Laser Applied to Study OM of Whole Soil

Some results of the Fluorescence Induced by Laser (FIL) applied to study of organic
matter (OM ) of whole soils are obtained during this year. This new technique has many
advantages. The first one is to make possible this study without fractionating processes
of OM, analyzing the soil near its natural condition. The second advantage is to allow
analyses of insoluble substances of organic matter, such as humina. Third, to value
carbon concentration without produce chemical residues. And finally, this technique
answers even when the soil possess high paramagnetic metal concentrations, where the
studies with magnetic resonance techniques, such as EPR and NMR, become
impracticable. This is the case of the most common soil in Brazil (fig. 2). Moreover, this
technique has a great potential for application in field conditions and can become a
compact sensor.

3. Interactions of Chlorine with Tropical Aquatic Fulvic Acids

Aquatic humic matter in fresh water ecosystems accounts for the major part of the
dissolved organic carbon (DOC) and it is, an important precursor for harmful byproducts
in the drinking water treatment process. We were characterized Aquatic Humic
Substances (AHS), and their components humic acids (HA) and fulvic acids (FA),
extracted from a Brazilian river. The characterization was done for better understanding
of structural composition of these matrices in tropical aquatics environments, that will be
used for latter analyses involving reactions with two types of disinfectant: chlorine and
chlorine dioxide. Techniques used for spectroscopic characterization were: nuclear
magnetic resonance (13C NMR), electron spin resonance (ESR), fluorescence and
absorption of UV-Visible light, diffuse reflectance infrared Fourier transform (DRIFT)
and Fourier Transform Infrared (FTIR). Analysis of the FA samples after reaction with
Cl2 were also made (Fig.3).

4. Photolysis of Aqueous Pesticide Residues in the Presence of Humic Acids

In this work it was investigated the direct and indirect photolisys of pesticide residues, in
aqueous solutions, under UV-visible radiation (280 – 500 nm). The pesticides Atrazine
(ATZ), Imazaquin (IMZ) and Iprodione (IPR) have been used with titanium dioxide
(TiO2) as main photocatalyst and a light source composed of 200W Hg-Xe lamp.
Different kinds of Humic substances were also used as photocatalysts, to know their
behavior in reactions with pesticides and its effect over photocatalysis by TiO2. They
were used standard soil humic acid (SHA) and standard Suwannee River humic acid
(SRHA) in concentrations varying from 1 to 150 mg.L-1. UV-visible absorption
measurements indicated a fast direct photolisys for the three pesticides that, except for
ATZ, reached total degradation over 2 hours of irradiation. When 100 mg.L-1 of TiO2
was additioned, there was observed 30-40% of efficiency increase, in the photolisys rate,
of ATZ and IPR, while IMZ not increase its photolisys rate. Humic acids (HA) addition
in concentrations above 10 mg.L-1 decreased the photolisys rate for all pesticides.
Measurements with spin-trapping Electronic Paramagnetic Resonance (EPR)
spectroscopy, showed that both TiO2 and humic acids (HA) act in a similar way,
photogenerating hydroxyl radicals which would be the responsible for the attack to the
pesticides molecules. However the HA absorbs great amount of radiation below 350 nm
and also react with the photogenerated hydroxyl radical, influencing directly the
efficiency of the photolisys process, turning it strongly dependent of the nature and
concentration of the compounds in the water to be treated.
