Importance in the Industry
Spectrophotometer
Spectroscopy is the study of the
absorption, emission, and
scattering of electromagnetic
radiation by atoms and molecules
in the gas, liquid, or solid phases.
Although the terms light,
radiation, and electromagnetic
radiation are all interchangeable,
visible electromagnetic radiation
is known as light.

WHAT IS
SPECTROSCOPY ?
WHO DISCOVERED
SPECTROSCOPY ?
The rainbow contains the spectrum nature of light, yet
it was beyond the ability of early man to comprehend
its importance. Newton did not demonstrate that the
sun's white light could be divided into a continuous
array of hues until 1666. To characterize this
occurrence, Newton used the term "spectrum." A small
aperture was used to define a beam of light, which was
then collidated by a lens, dispersed by a glass prism,
and displayed on a screen. This first spectroscope was
almost modern in design. The science of spectroscopy
began with Newton's examination of light.

Sir Isaac Newton

Generally, Sir Isaac Newton is credited with the discovery
of spectroscopy, but his work wouldn't have been possible
without the discoveries made by others before him.
PRINCIPLES
OF
OPERATION
All spectroscopic techniques work
on the same basic principle: shine a
beam of electromagnetic radiation
onto a sample and study how it
reacts. The response is normally
measured as a function of the
wavelength of the light. A spectrum
is a graph that shows the response
as a function of wavelength.

4. A radiation detector, which converts
radiant energy to a usable electrical
signal.
5. A signal processor and readout,
which displays the transduced signal.

electromagnetic absorption.
PRINCIPLES OF
The principle of operation of
the spectrophotometer is

OPERATION based on Beer-Lambert's law

which states that the amount
of light absorbed by a color
solution is directly
proportional to the solution's
concentration and the length
of a light path through the
solution.

2. Waves of specific wavelengths can be of wavelength. This is
manipulated to fall on the test solution using accomplished by diffracting the
appropriate techniques. The wavelengths of light beam into a spectrum of
incoming light can range from 1 to 2 wavelengths, detecting the

compound's absorption spectrum, or the
device, and graphing the results

Five Basic Parts of
Spectrophotometer:

1. Light Source - supply electromagnetic energy.

2. Monochromator - isolate the object of interest and eliminate the
unwanted second order radiation.
3. Sample Compartment - to accommodate the sample solution.
4. Detector - receive the transmitted light and convert it to electrical signal.
5. Digital or analogue display- to indicate absorbance or transmittance
A spectrophotometer is used in many areas of science including
microbiology, biochemistry, forensics, physics, and medical health.
You can use it to measure certain ingredients in a drug to make
sure it is effective and safe for consumers. You can measure
bacterial growth, or diagnose a patient based on how much uric
acid is present in their urine. Even non-scientists use
spectrophotometers. Wine-makers, for example, use them to
determine how much malic acid (reducing sugars) a particular wine
has in it.

Scientists can also use the

spectrophotometer to see how a reaction
has progressed. Let's say you're studying
iron deficiency, a condition that affects
over 2 billion people. Antioxidants,
vitamins, and other agents are known to
help in iron deficiency, but they don't
have a single specific chemical make-up,
and so they can't be directly measured.
However, they are known to reduce ferric
iron into ferrous iron, a more valuable

Uses of form to the human body.

Spectrophotometer
With the use of such a device, the person using a
spectrophotometer can easily acquire spectra of
shining white light on a given sample thereby
measuring the light that is returned from the
sample.
The device is used in many different fields,
especially in the production facilities and scientific
laboratories. In a production facility, a
spectrophotometer is used to check for the quality
of product such as in clothing and emission of light
during the manufacture of LED light, to name a few.
In analytical laboratories, a spectrophotometer is
used for the identification and quantifying
microscopic samples such as matching colors,
kinetics, qualifying gems and minerals, determining
the color of paint or ink, and so on.

Why is the
spectrophotometer
important?
