Research activities
My group has made very significant contributions in the area of kala-azar and cancer
research. During 1994-2004, WHO was deeply concerned about large-scale treatment
failure of anti-kala-azar drug sodium antimony gluconate (SAG)
(www.who.int/tdr;TDR/SWG/04). Our pioneering work provided immunological basis for this
SAG-unresponsiveness of kala-azar patients by showing that antimony-resistant
Leishmania donovani attenuates anti-leishmanial efficacy of SAG by inhibiting dendritic
cells (DCs) (PLOS PATHOGENS-2010). Another key invention from my laboratory with
direct medical implication is that Runx proteins promote anti-leishmanial immunity by
enhancing CD40-mediated type-1 T-cell responses (PLOS PATHOGENS-2020). This
study is equally significant for other pathogenic infections and has been featured by the
British Society for Parasitology (https://plos.altmetric.com/details/96796194/twitter). My lab
has also made seminal contributions in cancer biology. Our work revealed c-Src as
important target to reduce TIM-3 receptor’s expression on DCs and augment anti-tumor
immunity (J Biol Chem-2011, J Immunol-2014, J Immunol-2016). Notably, TIM-3 is an
emerging “check-point” receptor in cancer. Our work on TIM-3 was acclaimed as
outstanding by Prof. Vijay Kuchroo (Harvard Medical School), who discovered TIM-3.
Indeed, our study showing TIM-3 as inhibitory receptor on DCs (J Immunol-2014) has
overturned the prevailing paradigm that TIM-3 drives DC activation. Our another work on
c-Src and c-MET (Int J Biochem Cell Biol-2011) prompted the researchers of MD
Anderson Cancer Center to conduct first human Phase-I trial for evaluating anti-cancer
efficacy of crizotinib and dasatinib (Invest New Drugs-2018;doi:10.1007/s10637-017-0513-
5). Thus, my group’s work has unveiled many unique mechanisms and potential
therapeutic targets to combat kala-azar and cancer.

Beside fundamental research, my group at CSIR-IMTECH has successfully developed the


biosimilar of adalimumab, which is world’s top selling drug and used for treatment of
rheumatoid arthritis. Because adalimumab-based therapy is largely unaffordable by many
Indian patients, our work has tremendous societal significance.
