Patient blood

Tube method/MAJOR

1. Label two tube


2. Check the serial number of the blood component w/ the requested blood
3. Get 3 drops of Donor’s blood & wash it with 3% NSS solution (3-4 times centrifuge)

4. Discard supernatant
5. Heat with water bath (37 deg C)
6. In the second test tube, mix 1 drop of washed RBC & 1 drop of patient serum

7. Add one drop of Bovine Albumin (RAM)

8. Cover with parafilm & water bath (30 mins)

9. Wash with 3% NSS (3 to 4 times)

10. Decant supernatant

7. Add 1 drop of human anti-globulin

8. Put in slide and check under microscope

 Compatible: No agglutination
 Not compatible: agglutination
