- Wait 15-20 seconds and exit out of the FX2_usb software.

- Reboot the CFX96 Deep well and after the startup is complete start FX2_ USB
software
- Verify communication with the CFX96 Deep well system.
- Navigate to the (Read LED Currents) menu of the FX2_USB software (step 3 through
step 6 in this procedure).
- Verify that the LED DAC values displayed are the correct values entered earlier.
- Run complete system check out.

-Goto: View > File info > Flourophore Calibrations > Expanded
-Scroll to FAM BR White
-Check the value of :dye0:60:PR-:empty; under channel 1
-Divide the value by 17,000 to get the Factor, F. F= x/value = 14640.44/17000 =
0.8612

-Record the current LED DAC in channel 1. This is in the “.pcrd” file, under Plate
Readings/”Show Raw Plate Read Data” button

-Determine the new LED DAC:

New LED DAC = [(Current LED DAC – 34) / F ] + 34
In this example:
Current LED DAC: 110
F = 0.8612
New LED DAC = [ 110 – 34) / 0.8612 ] + 34 = 122.25
