Laboratory Activity #1
pH and Buffer

Introduction (one paragraph is enough)


This laboratory experiment/activity will tests . . . .
Generally, pH BUFFERS are composed of a mixture of a conjugate
base with a weak acid which can absorb excess hydrogen atoms
to maintain a stable pH value in solution. They are
subsequently ideal for calibrating electrochemical
potentiometers for low uncertainty pH measurements with
greater traceability.

Procedures: (include pictures in step-by-step format)

First is the Antilog pH, Before you can get it you need to
convert the pH value obtained above to hydronium ion
concentration. Next is the Urine, the 3 ml of urine add 3
drops of bromyhmol and next is the 3 ml of urine with a 3
drops of bromcresol last is the 3 ml of urine with 3 drops of
thymol. if the color is intermediate between yellow and deep
blue, no further rough test is necessary.

Result and Conclusion (with pictures)

Based on the result of our test is the bromthymol turned yellow and
it has a pH value of 6 and the bromcresol turned deep blue and it's
pH value is 7.6 next is the thymol it turned deep yellow. So the
result of the urine is acidic and the average pH of the urine is pH
6.

Denaturation OF Proteins

Introduction
This laboratory experiment will tests the Nature/Organization
of protein molecule and also the properties of proteins. The
test that are used in the experiment will show the change in
the protein based (that is used) in different types of
testing.

Egg Albumin Solution
Concentrated Hydrochloric Acid

Picric Acid

Procedures
a. Coagulation of Proteins

1. By Heat

Place 3 ml of egg albumin solution in a test tube and boil


gently for a few minutes. Describe the change in the albumin
solution.

Result: after heating for a few minute


the solution formed bubbles which is
positive solution.

2. Heller’s Ring Test

Place 1 ml of conc. Nitric acid in a test tube. Dilute 1 ml of egg


albumin solution with 1 ml of water and mix well. Incline the test
tube containing the nitric acid and carefully pour the 3 ml of
dilute albumin solution down the side of the tube so that it forms a
layer above the acid solution. What do you observe at the juncture
of the two liquids

Result: The nitric acid cooked the


egg and separated it the three
solution got separated with each other
and formed a cloudy white ring.

Place 1 ml. of egg albumin in a test tube and add 1 ml. of 95%
