White cells are a group of cells whose primary purpose is
to defend against bacteria, viruses, fungi, or other
foreign substances.
The most are granular cells
 contain enzymes used for digestion and destruction of
the invading organisms.
 Myeloid : Erythroid Ratio M:E
Normal value: 4:1
indicating that four myeloid, or white blood cells are
produced for one erythroid cell
There are three stem cells in the bone marrow

Plurpotent stem cell (PSC)

Myeloid stem cell (MSC)

Lymphoid stem cell (LSC),

1-Myeloblast:
Nucleus :It has a large
nucleus with fine
chromatin,
Nucleoli:2–5 and scanty pale
blue
Cytoplasm : without granules

2-Promyelocyte
 size: Slightly larger than a
myeloblast
 Nucleoli: less clear nucleoli.
 Cytoplasm: contains both
azurophilic granules and
characteristic neutrophilic

3-Myelocyte
 Size: Smaller than a
promyelocyte
 Nucleus: the nucleus is usually
oval
 Nucleoli: without

4-Metamyelocyte
 Nucleus: The nucleus is
indented (U-shaped)
 Cytoplasm : is pale pink and

5-Band Neutrophil (stab
form):
 Similar to the
metamyelocyte but with a
curved or coiled, often S
shape ,nucleus (without
clear lobes),

6- Neutrophil :
 Nucleus : has 3
(occasionally 4 or 5) clear
lobes, separated by
chromatin threads.
 Cytoplasm: is pale staining
and contains neutrophilic
granules.

their maturation, are;
 1 -granules appear in the
cytoplasm.

differentiation into three
colors when stained by

Development steps are the following
 1-Monocytoblast --Monocyte

 GM-CSF: For regulation and stimulation of
granulocytes and monocytes
 M-CSF : For regulation and stimulation
exclusively of monocytes

 CD markers: CD33, CD13, CD14


Normal neutrophils with 3-4 lobes
Band form with tow lobes
Polysegment neutrophil or (PMNs) over 5 lobes
Eosinophil 2 lobes
Basophils 2 lobes invisible

That’s means all the mature white blood
cells which contain granular in their
cytoplasm and existing in the storage
places in three distinct cell
compartments:
• the bone marrow (production site)
• the circulation (Transport way)
• and the tissues. (activity site)
