This manual is intended to provide the USER with instructions for use and to provide the SERVICE with
installation and maintenance instructions, for the following cabinet family:
The use of this equipment is limited by what is described in the paragraphs CABINET DEFINITION,
INTENDED USE and DEFINITION OF IMPROPER USE.
WARNING!
The operating manual is an integral part of the machine and it refers to the above cabinet
codes only.
Special versions of this cabinet with different code could be partially covered by this
manual, but they could also need additional instruction to add or substitute parts of this
manual.
In order to make the guide as complete and comprehensible as possible, the utmost care
has been taken in the collection and verification of the documentation contained in this
manual, but it is not designed to act as your procedure manual or act as a substitute for
validation of your own procedures.
The cabinet code is printed on the data label (see MARKING).
Unless otherwise specified, the information contained in this manual is applicable to all
codes listed above.
• Read the manual before starting ANY ACTIVITY involving the machine, including its
handling, installation and maintenance.
• The instructions given in this Manual shall be carefully read and understood before
switching this equipment on before you operate it, in order to become familiar with its
operation and function, and to avoid an IMPROPER USE.
• This manual should always be available in the operating area of the cabinet, in order to be
quickly consulted for use and service (routine checks and maintenance, and must
accompany it from the time of its manufacture until its dismantling, in the event of
relocation, sale, etc.
• In case of special cabinets derived from this model, consult the dedicated manual or the
supplementary manual referring to the code of the special cabinet.
For a better consultation, the operating manual is divided into the following sections:
◼ Part 1: containing the use instructions for the OPERATOR.
◼ Part 2: containing installation and maintenance instructions, intended solely for suitably trained
TECHNICAL personnel authorized to work on the cabinet.

1. ASSESSMENT OF RISKS
SafeMate Eco+ / Eco+ BS is a Class II Microbiological Safety Cabinet according to EN 12469:2000.
NOTE 1: notwithstanding conformity with the above standard, during operating of the cabinet some
residual risks remain and were assessed in order to indicate the measures to be taken to prevent the
hazards arising from normal use and foreseeable misuse.
The risks considered are the following:
▪ General
▪ Misuse
▪ Electric shock
▪ Biological
▪ UV-C radiations
▪ Pinch
▪ Spread of fire from the equipment
▪ Liberated gases and explosion
The analysis was done as per EN 61010-1:2010, 7.3.3 and Annex J.
The residual risks have been assessed as per the following Tab. 2:

• Prevent access to the place where the cabinet is installed to unauthorised
people or general public.

• Keep the key of the switch in a safe place to avoid the possibility of using the
cabinet by unauthorised personnel.
• Use spare UV-C lamp recommended by BioAir only.
• DO NOT plug any additional UV-C fixture to the service socket.

direct or reflected (see 5.1.4.1).

• A warning marking (disconnecting the power supply) is affixed to the filter
compartment access panel.
• Adequate information for user.
• The control panel can only be opened by authorized personnel in possession of
the key to open the control panel.

• Adequate information for user.
PRECAUTIONS TO BE TAKEN:
• Check the TLV or TWA allowed by the legislation of your Country.
• Decontamination shall be carried out exclusively by specialized and authorised
staff, following the instructions listed in par. 9 (PART 2 of this manual).
• Read advance the safety data sheet of the substance used for decontamination.
• Personal protective equipment (PPE) should be available on the spot to protect
the personnel against the risks indicated.
• DO NOT stay in the room during decontamination activity.
• Provide adequate equipment for reading the concentration of H2O2.
• Consult safety officer for precautions requested by local authorities.
• If another decontamination method is chosen, perform a risk analysis related to
the dangers deriving from this other method.

This cabinet is not suitable for operating in:
• residential areas.
On receiving the equipment make sure that the supply corresponds to what has been ordered.
In case of non-compliance report immediately to BioAir (see CONTACTS).
Also check it has not been damaged during transport.

• The operations required for installation and commissioning (unpacking, positioning,
assembly, connection to the mains power supply, installation of optional accessories and
testing) must only be carried out by specialized and authorized personnel, in accordance
with the instructions listed in PART 2 of this manual.
• Ask your dealer or BioAir for commissioning.

NOTE: the use of other ways to support the cabinet is discouraged and is under full the
responsibility of the customer.
WARNING!
This operation should be carried out by service specialized and authorised personnel.
Installation must be carried out in accordance with the instructions in the Part 2 of this
manual (see 8.3.1.1 and 8.3.1.2) and in accordance with the instructions supplied with the
accessory.
THE SAFETY OF THE CABINET DEPENDS ON THE CORRECT INSTALLATION OF THE ACCESSORIES ACCORDING
TO WHAT IS INDICATED IN THIS MANUAL AND IT IS THE RESPONSIBILITY OF THE ASSMBLER.
The support is available in the list of optional accessories in fixed or adjustable height version.
Height and other features of the stands are indicated in 3.10.1.
NOTE 1: the result of the stability test allows the use of the support in combination with the
SafeMate Eco+ / Eco+ BS cabinet as per height limits shown in Tab. 3.
The stability tests were made in accordance with EN 61010-1 paragraph 7.4.
NOTE 2: the choice should be defined at installation phase. Any variations must be carried out
by qualified and authorised technical personnel.

• The user should not connect the cabinet to the mains before commissioning and final
testing.
• The first connection should be carried out only by authorised personnel in accordance with
the instructions in the Part 2 of this manual (see 8.5).
The cabinet is designed to operate in the range defined by the technical data sheet (see 3.1)
and in the environmental conditions indicated in 3.4.
• Check that the mains voltage matches what shown on the data plate of the cabinet.
• Use a dedicated power line equipped with a ground connection that complies with local
regulations.
• The magneto-thermal protection to be used on the power line is indicated in Part 2 of this
manual (see 8.5).
• If using an uninterruptible power supply (UPS), read the information in Part 2 of this
manual (see 8.5).
3. Connect the plug (Pic. 5 – item 2) of the supply cord (2.3.1) directly to the
dedicated mains socket (2.3.2).

BioAir may supply on request the following optional detachable supply cords:

An unsuitable or defective mains supply cord compromises the safety of the cabinet.
PRECAUTIONS TO BE TAKEN:
• Do not cut the cord to replace the plug or connector.
In case of incompatibility with mains socket or damage of the cord,
plug or connector, ask the maintenance staff to replace the complete
cable with a certified type and with the characteristics indicated in
Tab. 9.

The appliance inlet (Pic. 10) is placed on the right top of the
control panel (see Pic. 15 – item 1) and consists to a male
connector (IEC 60320 C14) with a fuse holder for two main
fuses (Pic. 11).
The main fuses are replaceable from the outside, located in
the inlet appliance.
A label close to the appliance inlet reports the values of the Pic. 10 Pic. 11
fuses (see Pic. 12).
Fuses rating are also available in the table of specifications
(see 3.1):

On the cabinet there are two lines for technical gases and vacuum.
To take advantage of these utilities, specific (optional) taps must be installed, supplied separately in the
installation KIT (see 5.1.5).
On the flammable gas line must be installed the solenoid valve provided in the dedicated installation KIT.
WARNING!
The operation should be carried out by service specialized and authorised personnel.
Installation must be carried out in accordance with the ruling standards and the instructions
given in both Part 2 of this manual (see 8) and in those su supplied with the accessory.
The installer must issue a test report for the activation of these lines.
THE SAFETY OF THE CABINET DEPENDS ON THE CORRECT INSTALLATION OF THE ACCESSORIES ACCORDING
TO WHAT IS INDICATED IN THIS MANUAL AND IT IS THE RESPONSIBILITY OF THE ASSMBLER.
• Both in the case of installation and removal of this modification, the cabinet should be
validated by qualified personnel.
CHANGES LIKE THIS OR DIFFERENT CABINET CONFIGURATIONS WILL VOID ANY APPROVAL
MARKING IF APPLICABLE.

WARNING!
• Before the first use of the cabinet an efficiency test (installation testing) shall be carried out
as per table 5 of EN 12469 standard and Part 2 of this manual, paragraph 8.7, by a qualified
and authorised staff, with the issue of an installation report to be kept along with the
documentation of the cabinet.
This makes it possible to exclude any damage caused by transport or the installation itself,
which could affect the good operating and the safety of the cabinet.
The test should be performed by qualified and authorised staff, with the issue of relevant

4. OPERATING PRINCIPLE
SafeMate Eco+ / Eco+ BS is a cabinet of the type “VERTICAL LAMINAR AIRFLOW WITH PARTIAL
RECIRCULATION”, which provides protection for the operator, for the environment and for the product.
The effectiveness of this cabinet depends on several factors:
◼ location and installation test;
◼ training of personnel involved in the use;
◼ proper use and correct working procedures;
◼ correct handling of biological agents according to current regulation;
◼ routine and periodic maintenance; and
◼ routine performance testing (see 10.1.1).
The working area (Pic. 14 – item 1) is crossed
by a vertical unidirectional air flow
(DOWNFLOW, Pic. 14 – item 2).
This airflow is part of what supplied by a
motorblower (Pic. 14 – item 3), which blows into
a plenum (Pic. 14 – item 4) distributing the air
as follows:
➢ about 70% (Pic. 14 – item 11) conveyed
into the working chamber through an
absolute filter HEPA H141 (Pic. 14 – item
5) to create a sterile area in laminar flow
(LAF) in which the product is protected
from external and cross contamination;
NOTE 2: in BS type cabinets the exhaust airflow also passes through a second filter in line with
the first.
Both airflows are mixed by the suction of the fan, forming 100% of the (contaminated) airflow in the rear
intake duct (Pic. 14 - item 10) to repeat the cycle (Pic. 14 - items 11 and 12).
All potentially contaminated areas within the BSC that are at positive pressure are surrounded by zones of
negative pressure relative to the workroom atmosphere.

The front window is made of laminated safety glass (two layers) and is part of this cabinet
enclosure: in addition to being a protection against UV-C radiations, constitutes a decisive factor
for the containment efficiency of frontal barrier (INFLOW).

The breakage of the window compromises both the UV-C radiations protection and the air
barrier containment.
The front window (Pic. 19 – item 4) is sliding and it’s electrically

For this reason the working position height can not be changed.

1. push the key (UP) for at least 2 seconds, since the window rises again;
2. then push the key (DOWN) and try to lower the window;

On the line for non-flammable pressurized fluids, the tap provided in the list of accessories
must be mounted inside the work chamber (see 3.11).
The tap is also suitable for vacuum (see also 5.1.5, 3) and compressed air.
Fluids used must be non aggressive towards materials of which the valve is made (body in
brass, gaskets in PTFE).
On the flammable gas line (see also 5.1.5, 1 and 2) the following devices supplied with the
kit provided in the accessories list (see 3.11) must be mounted in line:
◼ the specific tap, inside the work chamber and on the same line as the solenoid valve,
Activation of the solenoid valve is only possible in MODE 1, by pressing the dedicated key on
the keypad (see also 5.2.1.3), provided that no alarm is running.
Once activated, the solenoid valve opens and it is possible to use the gas by acting on the tap.
To prevent HEPA filter damage and possible risk of fire (see 1.12), the solenoid valve
automatically stops the gas supply in the following events (EN 12469 standard, annex A.9):
➢ shutdown of the cabinet;
➢ sudden interruption of the power supply (BLACKOUT);
➢ lack of airflow;
➢ activation of any alarm.
NOTE 1: termination of alarm conditions and automatic or manual restart of the cabinet
after blackout will not reactivate the valve.
Restore can only be performed manually with ceased alarm.
NOTE 2: the provision of gas supply is not recommended. Flames from gas burners
interfere with laminar airflow within the cabinet and may compromise containment.
NOTE 3: where practicable, disposable loops or use of electrical loop sterilizers are

During ECO MODE (see also in 5.2, MODE 1) the cabinet cannot be considered
safe for working.
During this mode the alarm light (see 5.1.7.3) is on.
• BEFORE activating ECO MODE any sample still lying in the working area must be
put in a safe condition.
This mode saves energy in cases where, without any operator activity (eg a waiting period
between one activity and another), it is necessary to maintain the sterility of the working area
and still guarantee the front barrier.
In this way the return to MODE 1 for a new activity takes place without having to wait for the
start-up procedure.
• Check neither your arms nor objects are across the front aperture while
activating this mode.
NOTE 2: if the front window is jammed, or gets jammed during the activation of ECO
MODE, the cabinet will automatically return to MODE 1 and display the screen of Pic. 49.
NOTE 3: in case of BLACK OUT, when the mains power is restored, the cabinet will restart
in MODE 1 (see sequence in point 2 below), but showing the screen in Pic. 48 at the end of the

2. To exit ECO mode (restoring of MODE 1), press (MODE key) for 2 seconds.
An acoustic and visual alarm is activated to signal the automatic upward movement of the
front window. The window stops when the working position is reached.
At the same time the airflow will be increased to reach the configured value.

material separate from the sterile one, place the latter far from the working area and put the wastes

Pay special attention when removing cultures that have been accidentally poured inside the working
area: they could favour the growth of fungi and the spreading of their spores with consequent
contamination of the cabinet. For cleansing and decontamination, see par. 7.
◼ Keep the cabinet on for at least 10 minutes once the work is over, in order to remove any
contaminating substance from inside.

The exhaust air can be left free in the environment, but in the required cases can be further
filtered and/or ducted outside.
The MSC shall not be connected directly to any other air-handling system.
NOTE: it may be provided exhaust the air to the atmosphere through an indirect connection,
and possibly to further purify the exhaust air, e.g. through an absorbent filter.
These modifications can be obtained through the installation of accessories designed by BioAir for
these needs. Below are the available accessories:
WARNING!
The requirement and feasibility of this type of modifications should be carefully
assessed by competent personnel in order not to compromise the performance of the
cabinet together with the safety of the operator, the environment and the product:
◼ in the case of an external ducting, an adequate air intake must be guaranteed in
the room;
◼ in the case of an additional filter, the increase in pressure drop on the exhaust
must be compensated by an adequate and balanced additional suction.
◼ Read also information in PART 1 of this manual (see 2.5).
For this type of requirements contact BioAir, which, directly or through local distributors, will
provide assistance for the specific needs of the laboratories, proposing the appropriate
solutions.
NOTE: the installation of these accessories must be carried out after the cabinet has been
tested, as the cabinet performance must first be checked in normal conditions. Subsequently
the validation must be made with the new configuration, carrying out again the tests related
to the values of the airflows and to the alarm interventions.

NOTE 2: in addition to the instruments required for the checks (see specific paragraphs), it is
advisable to have the following instruments

The test is based on the comparison between the particles detected upstream and downstream the
filter.
In order for the test to be valid, it is necessary that the concentration upstream of the filters is
sufficiently high to have a statistical confirmation. For this reason, particles having a particle size
within a given diameter must be produced by aerosol (see Tab. 10).
This concentration is conventionally defined as 100%.
The oils that can be used for aerosols must produce particles with a diameter of less than 1 μm
(MPPS).

leave the cabinet switched on for at least 15 minutes.

The decrease of the average value of INFLOW must not exceed 20% and must not be
less than 0,4 m/s.
10. Read the LAF value indicated by the hot wire anemometer.
The decrease of LAF vs the mean value should not exceed 20%.
NOTE 2: the value shown on the display always represents an average value, therefore
when checking the maximum and minimum LAF values, this cannot coincide with what is
indicated by the anemometer.
Normally the gap is equal to the difference between the average value and the value being
checked. Tab. 14 shows how to interpret the alarm value shown on the display.

9. BIOLOGICAL DECONTAMINATION
The decontamination is a chemical and physical process for destroying all the kind of microorganism.
The decontamination of a MSC cabinet is MANDATORY before any service or maintenance operation,
replacement of the filters or any other operations that require access to parts potentially contaminated of
the cabinet and that could present a biological risk against the maintenance operator and external
environment, before moving or disposal of the cabinet and after any emergency.
The choice of the cabinet decontamination method must fall in the risk assessment of the process and of
the activities carried out in the laboratory and must therefore consider the pathogens that are used.
Formaldehyde, which has always been used as an effective contamination agent and proposed in the
guidelines set out in Annex J of the European Standard EN 12469: 2000, has been recognized as a
carcinogen and at the issue date of this revision of the manual, several alternatives for decontamination
are under investigation.
An alternative decontamination procedure will be described below, which uses hydrogen peroxide (H 2O2)
as decontamination agent.
The procedure has been validated through the use of chemical and biological indicators and it has been
demonstrated the ability to sterilize samples with a concentration of 106 spores of Bacillus
stearothermophilus.
You can use another method of decontamination at the discretion of the laboratory manager.
For this eventuality a function of the cabinet is available (see 9.2.4), which could facilitate the procedure.
In any case it is important that the decontamination procedure is considered into the risk assessment of
the process and activities performed in the laboratory and that a validation of the procedure is performed
using the right chemical and biological indicators (BI) that have a behaviour similar to the pathogenic
agents that are used in the MSC; in particular the BI's should be chosen in order to have a 6-log
reduction and should be positioned in the places where it is more difficult the diffusion of the
decontamination agent (for example in corners of working area, in air diffusion plenum, after exhaust
filter).
This procedure must be validated by a method demonstrating compliance with the following requirements:
1) Decontamination effectiveness through the use of chemical and biological indicators correct and
positioned at critical points.
2) Safety of the operator and of the surrounding environment which should therefore not be
contaminated with dangerous substances.
3) Compatibility of the used agent with the mechanical and electrical components of the cabinet.
For the procedure with H2O2, BioAir has considered all the aspects listed above in the definition of the
following decontamination cycle.

The solution of hydrogen peroxide and silver is nebulised into particles smaller than 5 μm. In
technical jargon the micronebulization (dry mist), that is the process through which a liquid is
reduced into tiny particles.
The emitted particles are airborne and, remaining suspended, reach all free surfaces.
The cabinet is wrapped in a transparent plastic cover and sealed in order to guarantee the
effectiveness of the process and not to disperse the particles generated by the nebulisation of
H2O2 into the environment.

11. Coat the cabinet with the plastic bag (Pic. A34 - item 2) supplied in the decontamination kit
(see 9.1.2.2) by trimming it in such a way as to eliminate the excess parts
(This operation is used to avoid the formation of pockets in which decontaminant residues
may remain in high concentrations).
12. Make the necessary holes to make the following cables come out:
◼ exit hole of the cabinet power cord (Pic. A34 - item 3); and
◼ exit holes of the control and supply cables of the nebulizer (Pic. A34 - item 4).
13. Carefully seal all the engraved parts for the arrangement of the bag and the cable passage
holes with adhesive tape.
14. Carefully seal the junction of the plastic bag with the cabinet along the entire lower perimeter
with adhesive tape (Pic. A34- item 6).
15. Check carefully that there are no damages (even small scratches) to the containment bag
and in case seal them with adhesive tape.
16. Apply the "CAUTION DECONTAMINATION IN PROGRESS" warning on the containment bag
and on the outside of the access door to the room.
17. Be sure that there is no risk that an HVAC plant will recirculate air into an adjacent room with
presence of personnel.

The doses and times indicated in Tab. 16 for the decontamination cycle with this method
refer only to the volume created by the containment bag.
If for some reason it is necessary to act on larger volumes, the times and the doses
must be recalculated through a new validation of the necessary procedure.
5. Press the PROGRAM MODE key on the external keyboard of the nebulizer again (Pic. A35 -
item 4) to confirm the program.
6. Make sure that there is no personnel in the room affected by this decontamination.
WARNING!
During the whole decontamination phase, equip yourself with adequate portable
instrumentation to check the level of H2O2 in the environment remains within allowed
NOTE 3: if this filling lasts for longer than 150 seconds (therefore the pump remains
active for longer than 150 seconds) it is assumed the bottle is empty (no functional block is
provided for).
The display will show this condition with the message: ALM BOTTLE.
To reset the signal it is necessary to replace the bottle and then restart a cycle of
decontamination. The bottle release method is described in 9.2.3.1.
Once the decontaminant load has been
completed, the display shows the message in
Pic. A38 with the start of H2O2 dispensing and
the set time decrease.
Once the program minutes/seconds have
elapsed, the START/STOP LED turns off while the Pic. A38
display returns to display the message in Pic.
A36.
9. Leave the cabinet switched on and wait for 60 minutes.
10. Start the second decontamination cycle by pressing the STAR/STOP key.
11. Leave the room.
12. After the second cycle, wait 15 minutes and then switch off the cabinet.
In this condition the dispensing phase of decontamination cycle is ended.
NOTE 4: in case of interruption of mains electrical supply, when power is reinstated the
unit goes back to the state (idle-on or stand-by) that it was in at the time of power failure.
If a decontamination cycle is performed during the interruption, it is aborted.
All the data set remain stored. The decontamination is to be repeated.
13. Leave the cabinet sealed and wait 12 hours to allow the H2O2 concentration to be reduced.

Even if the cabinet is sealed and the recorded level of H 2O2 during validation test is lower
than allowed limit, consider always that something could go wrong and so use all the
necessary precautions before to enter into the room.
Before proceeding with this operation after decontamination cycle, use the necessary
precautions: it is recommended the use of PPE (mask, gloves, glasses, etc.) and verify
by means of suitable equipment that the H2O2 concentration in the room be lower than

after 4 hours.
5. Once the concentration level is stabled within the limits, remove the plastic bag, the hose and
the chemical indicators.
In this stage still wear protective devices.
6. Run the cabinet in MODE 1 for at least 15 minutes.
7. Check that the concentration level of H2O2 in the cabinet and in the room is less than the

9.2.4.1 PROCEDURE
1. Turn on the cabinet in MODE 2 (see
PART 1, 5.2.2).
2. Close the front window.
8. Elapsed this time press the keys e and hold them for Pic. A43
30-40 seconds.
The service socket is turned off and the fan starts. Display will
show the message FUMIGATION RECIRCULATION (Pic. A43).
9. Release the keys.
The fan stops. Display shows FUMIGATION IN PROGRESS (Pic.
A41).
10. Leave the cabinet in this status for the time established for the Pic. A44
contact phase.
11. Past this time restart the cabinet.
PURGE mode is activated: the fan starts (FUMIGATION PURGE
Pic. A44) at reduced velocity and the front window will be
partially opened (few centimetres).
12. Remove the switch key and leave the cabinet running in
FUMIGATION PURGE for the time established for the purge
Pic. A45
phase.
In case of blackout, when the power is reactivated the display
will show the message of Pic. A45.
13. After PURGE phase turn off the cabinet and remove the used
equipment.

pericoli derivanti da uso normale e prevedibile uso improprio.

• L’utilizzatore non deve collegare la cabina alla rete prima dell’avvenuta messa in funzione e
collaudo finale.
• La prima connessione deve essere eseguita solo da personale autorizzato in conformità alle

La cabina è progettata per funzionare nel campo definito dalla scheda tecnica (vedi 3.1) e
nelle condizioni ambientali indicate in 3.4.
• Controllare che la tensione di rete corrisponda ai dati di targa della cabina.
• Utilizzare una linea elettrica dedicata, dotata di collegamento di terra conforme alle norme
locali.
• La protezione magnetotermica da utilizzare sulla linea elettrica è indicata nella Parte 2 di
questo manuale (vedi 8.5).
• In caso di utilizzo di un gruppo di continuità (UPS), leggere le informazioni nella Parte 2 di
questo manuale (vedi 8.5).
Procedere quindi come segue:
1. Collegare il connettore femmina (Fig. 5 – pos. 1) del cavo di alimentazione (2.3.1) all’ingresso di
alimentazione (Fig. 7 - pos 1).
2. Inserire il gancio di blocco (Fig. 6) nell’inserto filettato (Fig. 7 - pos 2) avvitandolo fino ad aderire
alla parte superiore del connettore in modo leggermente forzato (a mano).

Sulla cabina sono predisposte due linee per gas tecnici e vuoto.
Per usufruire di tali utenze è necessario installare i rubinetti specifici (opzionali), forniti a parte in KIT
d’installazione (vedi 5.1.5).
Sulla linea gas combustibile deve essere installata l’elettrovalvola fornita nel KIT d’installazione dedicato.
ATTENZIONE!
L’operazione deve essere eseguita da personale tecnico specializzato e autorizzato.
L’installazione deve avvenire in conformità alle norme vigenti e alle istruzioni riportate sia nella
Parte 2 di questo manuale (vedi 8), sia in quelle fornite con l’accessorio.
L’installatore deve rilasciare un rapporto di collaudo per l’attivazione di queste linee.

ATTENZIONE!
• Il primo utilizzo della cabina deve essere preceduto da un test di efficienza (test di
installazione) secondo tabella 5 della norma EN 12469, e secondo il paragrafo 8.7 (Parte 2 di
questo manuale), effettuato da personale qualificato ed autorizzato, con emissione di un
rapporto di installazione da custodire insieme alla documentazione della cabina.
Ciò consente di escludere eventuali danni causati dal trasporto o dall'installazione stessa, che
potrebbero influire sul buon funzionamento e sulla sicurezza della cabina.
Il test deve essere eseguito da personale qualificato e autorizzato, con emissione del relativo
rapporto da conservare insieme alla documentazione della cabina.
NOTA: la cabina è collaudata in fabbrica con la configurazione “RIPIANO CHIUSO”, perciò in caso sia
stata fornita la cabina con “RIPIANO FORATO”, i dati presenti sul rapporto di collaudo (Certificato di Qualità)
relativi alla griglia di LAF (flusso laminare verticale) andranno verificati ed eventualmente aggiornati.
ATTENZIONE!
• Non utilizzare questa MSC se priva di rapporto di installazione.
Una volta terminata l’installazione eseguire le procedure finali prima di iniziare il lavoro:
• accendere la cabina in MODO 2 (vedi 5.2.2) e aprire la zona di accesso frontale sollevando il
vetro tramite l’apposito pulsante (vedi 5.1.7.8);

4. PRINCIPIO OPERATIVO
SafeMate Eco+ / Eco+ BS è una cabina di tipo “A FLUSSO LAMINARE VERTICALE A PARZIALE RICIRCOLO”
che fornisce protezione per l’operatore, per l’ambiente e per il prodotto.
L’efficacia di questa cabina dipende da diversi fattori:
◼ ubicazione e test d’installazione;
◼ formazione del personale addetto all’uso;
◼ uso adeguato e procedure di lavoro corrette;
◼ manipolazione corretta degli agenti biologici in accordo alle norme vigenti;
◼ manutenzione ordinaria e periodica; e
◼ verifica periodica delle prestazioni (vedi 10.1.1).
La zona di lavoro (Fig. 14 – pos. 1) viene
attraversata da un flusso d’aria unidirezionale
verticale (DOWNFLOW, Fig. 14 – pos. 2).
Questo flusso d’aria è parte di quanto fornito
da un motoventilatore (Fig. 14 – pos. 3), che
soffia in un plenum (Fig. 14 – pos. 4)
distribuendo l’aria nel seguente modo:
➢ circa il 70% (Fig. 14 – pos. 11)
convogliata nella camera di lavoro
attraverso una filtro assoluto HEPA H141
(Fig. 14 – pos. 5) per creare una zona
sterile in flusso laminare (LAF) nella
quale il prodotto è protetto da
contaminazioni esterne e crociate;
volume diretto verso l’interno attraverso l’apertura frontale (INFLOW, Fig. 14 – pos. 8) al fine di
formare una barriera di protezione per l’operatore.
NOTA 2: nelle cabine in versione BS il flusso d’aria espulso attraversa anche un secondo filtro in
serie al primo.

Tutte le aree potenzialmente contaminate all'interno della MSC, che sono a pressione positiva, sono
circondate da zone di pressione negativa rispetto all'atmosfera del laboratorio.

PRECAUZIONI DA ADOTTARE:
• In caso che avvenga un consistente versamento di liquidi contaminati, nei casi di
assistenza tecnica e manutenzione che prevedono sostituzione filtri o
motoventilatore, e prima dei controlli periodici è necessaria una decontaminazione
efficacie e completa della cabina, con un metodo validato (vedi par. 9 per
informazioni e per il metodo di decontaminazione proposto da BioAir).
NOTA 2: il fatto che la lampada UV produca luce non è direttamente correlato al proprio
potere effettivo di neutralizzazione di microrganismi. L’effettiva efficacia può essere controllata
con apparecchiature adeguate.
Su richiesta è possibile far eseguire un test di efficienza delle radiazioni emesse dalla lampada
UV-C: contattare il vostro distributore BioAir.
ATTENZIONE!
Si sconsigliano permanenze lunghe o postazioni di lavoro che possano essere
raggiunte dalla luce UVC diretta o riflessa.

• In caso fosse indispensabile permanere nello stesso locale durante il

Sulla linea per fluidi pressurizzati non combustibili, deve essere montato all’interno della
camera di lavoro il rubinetto con il kit previsto nella lista degli accessori (vedi 3.11).
Il rubinetto è adatto anche all’uso per il vuoto ordinario (vedi 5.1.5, 3) e aria compressa.
I fluidi utilizzati non devono essere dannosi per i materiali di cui è composto il rubinetto (corpo
principale in ottone, guarnizioni in PTFE).

Durante il MODO ECO (vedi anche in 5.2, MODO 1) la cabina non può essere
considerata sicura per attività.
Durante questa modalità la spia rossa di allarme (vedi 5.1.7.3) è accesa.
• PRIMA di attivare il MODO ECO, qualsiasi campione ancora presente nell'area di
lavoro deve essere messo in condizioni di sicurezza.
Questa modalità consente di risparmiare energia nei casi in cui, senza alcuna attività
dell'operatore (es. un periodo di attesa tra un'attività e l'altra), è necessario mantenere la
sterilità dell'area di lavoro e garantire comunque la barriera frontale.
In questo modo il ritorno al MODO 1 per una nuova attività avviene senza dover attendere la
procedura di avvio.
NOTA 2: se il vetro frontale è bloccato, o si blocca durante l'attivazione del MODO ECO, la
cabina tornerà automaticamente in MODO 1 visualizzando la schermata di Fig. 49.
NOTA 3: in caso di BLACK OUT durante il MODO ECO, al ritorno dell’alimentazione di rete
la cabina si riavvierà in MODO 1 (vedi sequenza nel punto 2 di seguito), mostrando però al
termine del riavvio la schermata di Fig. 48.
2. Per uscire dal MODO ECO (ripristino del MODO 1), premere (tasto MODO) per 2
secondi.
È attivato un allarme acustico e visivo per segnalare il movimento automatico del vetro
frontale verso l’alto. Il vetro si ferma al raggiungimento della posizione di lavoro.
Contemporaneamente il flusso d'aria sarà aumentato per raggiungere il valore configurato.
Durante il ripristino del MODO 1 il display mostra la Fig. 46 con allarme acustico e visivo
attivo fino a quando la cabina non sarà in condizioni di sicurezza operativa.
L'illuminazione si accende.
L'elettrovalvola rimane disattivata.
Le prese elettriche rimangono nello stato in cui si trovano.
Raggiunte le condizioni di sicurezza operativa l'allarme si spegne e sul display compare la
schermata di Fig. 42.

◼ Evitare di far passare materiale contaminato sopra a quello sterile, tenere separato il materiale
contaminato da quello sterile, collocare quest’ultimo lontano dalla zona di lavoro e mettere gli scarti
in un angolo posteriore della zona di lavoro.
◼ Non collocare nella cabina apparecchiature di grandi dimensioni e ingombranti e non posizionare
apparecchiature sulle griglie di aerazione per non disturbare i flussi di aria.
◼ Non ostruire le asole di ripresa dell’aria nella camera di
lavoro (vedi 5.1.6.3) con oggetti (vedi Fig. 64) e in
particolare fare attenzione a non ostruire la griglia
frontale con le maniche dell’indumento da lavoro.
◼ Utilizzare la cabina solo quando la ventilazione è in
funzione (MODO 1), con il display funzionante e in
assenza di allarme (acustico o visivo).
◼ Limitare le attività all’interno della stanza dove si trova
la cabina per non disturbare il contenimento della
barriera d’aria ed il flusso laminare (LAF), con
conseguente diminuzione del livello di protezione per
l’operatore e per il prodotto.
Ogni volta che le porte del laboratorio vengono aperte e
chiuse, si producono correnti d'aria.

◼ Ripetere la disinfezione delle superfici interne dopo che
la cabina è stata svuotata da materiali e gli strumenti.
Porre particolare attenzione all’eliminazione di colture accidentalmente rovesciate nella zona di
lavoro: queste possono generare la crescita di funghi e la diffusione delle loro spore, con
conseguente contaminazione della cabina. Per la pulizia e la decontaminazione vedi par. 7.
◼ Mantenere la cabina in funzione per almeno 10 minuti una volta terminato il lavoro, per permettere
l’eliminazione di qualsiasi sostanza contaminante.
◼ Sottoporre la cabina a controlli periodici secondo quanto indicato dal fabbricante (vedi 10.1.1) o
dalle autorità di regolamentazione, se più restrittive.

La procedura con radiazioni UV-C NON garantisce una reale riduzione della contaminazione
microbica, poiché è efficace solo per le superfici direttamente esposte ai raggi.
Questo metodo NON può sostituire quelli convenzionali di decontaminazione di una MSC.
Nei seguenti casi utilizzare un metodo di decontaminazione valido:
➢ prima di accedere a zone potenzialmente contaminate;
➢ ogni volta che avvenga una consistente fuoriuscita di liquidi contaminati;
➢ prima di una manutenzione (sostituzione di filtri e motoventilatore);
➢ prima dei controlli periodici;
➢ prima di un trasloco o smantellamento;
➢ dopo qualsiasi emergenza.
• L’operatore deve identificare il metodo di decontaminazione efficacie contro l’agente
patogeno in questione.
ATTENZIONE!
La rottura delle lampade UV e fluorescenti costituisce un rischio di lesioni alla pelle e agli occhi,
oltre a rappresentare un rischio per la salute dovuto ai vapori di mercurio, metallo contenuto
nelle lampade anche se in minime quantità, che possono sprigionarsi e disperdersi
nell’ambiente.

L’aria espulsa può essere lasciata libera in ambiente, ma nei casi richiesti può essere filtrata
ulteriormente e/o canalizzata all’esterno.
La MSC non deve essere collegata direttamente a nessun altro sistema di trattamento dell'aria.
ATTENZIONE!
L’esigenza e la fattibilità di questo tipo di modifiche devono essere attentamente
valutate da personale competente per non rischiare di compromettere le prestazioni
della cabina insieme alla sicurezza dell’operatore, dell’ambiente e del prodotto:
◼ nel caso di una canalizzazione all’esterno, deve essere garantita un’adeguata ripresa
d’aria nel locale;
◼ nel caso di un filtro aggiuntivo, l’aumento della perdita di carico sull’espulsione deve
essere compensata da un’adeguata e bilanciata aspirazione supplementare.
◼ Leggere anche le informazioni nella PARTE 1 di questo manuale (vedi 2.5).
Questi accessori permettono di convogliare l’aria espulsa dalla cabina nei condotti a sezione
circolare di diametro 200 mm. Vengono installati sul tetto della cabina dove sono presenti i punti
di fissaggio predisposti. Si raccomanda di non ridurre il diametro della canalizzazione.
Per questo tipo di esigenze contattare BioAir, la quale, direttamente o attraverso i distributori
locali, fornirà l’assistenza per le necessità specifiche dei laboratori, proponendo le soluzioni
adeguate.
NOTA: l’installazione di questi accessori deve essere successiva al collaudo della cabina,
poiché per prima cosa devono essere verificate le prestazioni della cabina in condizioni normali.
Successivamente dovrà essere fatta la validazione con la nuova configurazione, effettuando
nuovamente i test legati ai valori dei flussi d’aria e agli interventi di allarme.

Le verifiche necessarie sono le seguenti e devono essere eseguite in MODO 1 (vedi PARTE 1, 5.2.1),
tranne quella delle soglie di allarme, da eseguire in modalità MANUALE:
◼ Verifica dell’integrità dei filtri HEPA (vedi 8.8).
◼ Verifica della velocità di barriera frontale (vedi 8.9.1).
◼ Verifica della velocità del flusso verticale (LAF) (vedi 8.9.2).
◼ Verifica dei flussi d’aria con fumo (vedi 8.9.3).
◼ Verifica delle soglie di allarme (vedi 8.9.5).
◼ Verifica della pulizia dell’aria in zona di lavoro (vedi 8.9.4).
(NON RICHIESTO dalla norma di prodotto, opzionale solo su richiesta preventiva).
NOTA 1: le procedure di prova devono essere eseguite secondo quelle definite nel rapporto di
prova di fabbrica. I risultati delle prove di flusso laminare e barriera devono essere coerenti con
quelli del rapporto di prova emesso dalla fabbrica (Certificato di qualità), in cui sono indicate le
prestazioni e gli strumenti usati.
NOTA 2: oltre agli strumenti richiesti per le verifiche (vedi paragrafi specifici), si consiglia di
avere a disposizione i seguenti strumenti:

◼ multimetro digitale per misurare il vero valore efficace (true RMS) in AC;

NOTA 3: le verifiche sulle cabine in versione BS differiscono solo nel test di integrità dei filtri
(vedi 8.8.2).
Il test si basa sul raffronto tra le particelle rilevate a monte e a valle del filtro.
Affinché il test sia valido, è necessario che la concentrazione di particelle a monte dei filtri sia
sufficientemente alta da avere un riscontro statistico. Per questo motivo devono essere prodotte
tramite aerosol delle particelle la cui grandezza sia entro un diametro stabilito (vedi Tab. 10).
Questa concentrazione è definita convenzionalmente come 100%.
Gli oli utilizzabili per l’aerosol devono produrre particelle di diametro inferiore a 1 μm (MPPS).

L’anemometro deve essere sostenuto da un supporto appropriato in modo da poterlo lasciare in
posizione senza che il braccio dell’operatore possa creare disturbo alla misurazione.
La velocità dell’aria di BARRIERA (VINF) è la velocità media dell’aria che, entrando nella cabina
attraverso l’apertura frontale, protegge l’operatore.
Il valore è ottenuto per calcolo partendo dalla misura
della velocità dell’aria espulsa (VEXH) secondo EN 12469,
allegato G.
Verificare le condizioni indicate in 8.9.
Effettuare questa misura mediante un anemometro a
KI (vedi 2)
NOTA 2: il fattore KI si ottiene dividendo l’area della sezione in cui si effettua la misura per
l’area della sezione trasversale dell'apertura frontale (AREA DELLA BARRIERA, vedi PARTE 1, 3.6).
In Tab. 12 sono elencati i valori di KI secondo le taglie e le sezioni di espulsione previste per la
cabina e per gli accessori con canali di diametro 200 mm.
In caso di canalizzazione con sezione diversa, ricalcolare il fattore KI.
1. Calcolare la media delle misurazioni effettuate alla bocca di uscita dell’aria (VEXH).
2. Calcolare la velocità media della BARRIERA frontale (VINF) con la seguente formula:
VINF = VEXH x KI

verso l'interno, senza riflusso fuori dalla cabina o penetrazione del fumo sopra o sulla
superficie di lavoro.
◼ Il flusso d'aria all'interno dell'area di lavoro della cabina deve essere verso il basso
(nessun riflusso), senza fuoriuscita verso l'esterno della cabina ai lati e nella parte
superiore della finestra.

9. DECONTAMINAZIONE BIOLOGICA
Per decontaminazione si intende un processo chimico o fisico in grado di distruggere tutte le forme di
microrganismi viventi.
La decontaminazione di una cabina MSC è OBBLIGATORIA prima di qualsiasi lavoro di assistenza tecnica o
manutenzione, sostituzione filtri o altre operazioni che richiedano l'accesso a parti potenzialmente
contaminate della cabina e che potrebbero presentare un rischio biologico per il manutentore e l'ambiente
esterno, prima di un trasloco o dismissione o dopo qualsiasi emergenza.
La scelta del metodo di decontaminazione della cabina deve rientrare nella valutazione del rischio del
processo e delle attività svolte in laboratorio e deve quindi tenere in dovuta considerazione gli agenti
patogeni utilizzati.
La formaldeide, da sempre usata come agente di contaminazione efficace e proposta nelle linee guida di
cui all'allegato J della Norma Europea EN 12469:2000, è stata riconosciuta come agente cancerogeno e
alla data di emissione di questa revisione del manuale, diverse alternative per la decontaminazione sono
oggetto di indagine.
Di seguito sarà descritta una procedura di decontaminazione in alternativa, che come agente di
decontaminazione utilizza perossido d’idrogeno (H2O2).
La procedura è stata convalidata mediante utilizzo di indicatori chimici e biologici ed ha dimostrato la
capacità di sterilizzare campioni con una concentrazione di 106 di spore di Bacillus Stearothermophilus.
È possibile utilizzare un altro metodo di decontaminazione a discrezione del responsabile del laboratorio.
Per questa eventualità è prevista una funzione della cabina (vedi 9.2.4), che potrebbe agevolare la
procedura.
In ogni caso è importante che la procedura di decontaminazione sia considerata nella valutazione del
rischio del processo e attività svolte in laboratorio e che una validazione della procedura venga eseguita
utilizzando i corretti indicatori chimici e biologici (BI) aventi un comportamento simile gli agenti patogeni
che vengono utilizzati nella MSC; in particolare i BI devono essere scelti in modo da avere una riduzione di
6-log e devono essere posizionati in luoghi in cui è più difficile la diffusione dell'agente decontaminante (ad
esempio negli angoli della zona di lavoro, nel plenum diffusore dell'aria, dopo filtro di espulsione).
Tale procedura deve essere validata mediante un metodo che dimostri il rispetto delle seguenti
prescrizioni:
1) Efficacia di decontaminazione mediante utilizzo di indicatori chimici e biologici corretti e
posizionati nei punti critici.
2) Sicurezza per l'operatore e per l'ambiente circostante che non deve quindi essere contaminato
con sostanze pericolose.
3) Compatibilità dell'agente utilizzato con la componentistica meccanica ed elettrica della cabina.
Per la procedura con H2O2, BioAir ha preso in considerazione tutti gli aspetti sopra riportati nella
definizione del seguente ciclo di decontaminazione.

Le dosi e i tempi indicati in Tab. 16 per il ciclo di decontaminazione con questo metodo si
riferiscono solo al volume creato dal sacco di contenimento.
Se per qualche motivo è necessario agire su volumi maggiori, i tempi e le dosi vanno
rideterminati attraverso una nuova validazione della procedura necessaria.
5. Premere nuovamente il pulsante PROGRAM MODE sulla pulsantiera esterna del nebulizzatore
(Fig. A35 - pos. 4) per confermare il programma.
6. Accertarsi che non ci sia personale nella stanza interessata a questa decontaminazione.
ATTENZIONE!
Durante tutta la fase di decontaminazione munirsi di strumentazione portatile adeguata
per verificare che il livello di H2O2 nell’ambiente resti entro i limiti permessi (vedi 9.1.2.4
e relativa ).
7. Avviare il primo ciclo di decontaminazione premendo il tasto STAR/STOP (Fig. A35 - pos. 5).
Il LED relativo lampeggia fino a fine ciclo.
È emessa una breve segnalazione acustica intermittente per 15 secondi (countdown) ad
indicare che è stato attivato il ciclo.
Il countdown permette di lasciare l’ambiente in cui è posta la macchina, una volta premuto il
tasto START/STOP, prima che inizi il ciclo vero è proprio (erogazione del prodotto).
8. Abbandonare la stanza.
Al termine del countdown il display visualizza il messaggio di Fig. A37.
NOTA 3: se il carico del serbatoio ha una durata superiore a 150 secondi (quindi la
pompa resta attiva per più di 150 secondi) si presuppone che la bottiglia sia vuota (non è
previsto il blocco funzionale).
Il display visualizza questa condizione tramite la scritta: ALM BOTTIGLIA.
Per resettare la segnalazione è necessario sostituire la bottiglia quindi riavviare un ciclo di
decontaminazione. Il metodo di sgancio della bottiglia è descritto in 9.2.3.1.
Terminato il carico di decontaminante il display
visualizza il messaggio di Fig. A38 con inizio
dell’erogazione di H2O2 e del decremento del
tempo impostato.
Una volta trascorsi i minuti/secondi di
programma il LED START/STOP si spegne Fig. A38
mentre il display torna a visualizzare il
messaggio di Fig. A36.
9. Lasciare la cabina accesa e attendere 60 minuti.
10. Avviare il secondo ciclo di decontaminazione premendo il tasto STAR/STOP.
11. Abbandonare la stanza.
12. Terminato il secondo ciclo attendere 15 minuti e poi spegnere la cabina.
In questa condizione la fase di erogazione del ciclo di decontaminazione è terminata.

Anche se la cabina è sigillata ed il valore di concentrazione di H2O2 registrato durante i test
di validazione è inferiore al limite consentito, considerare sempre che qualcosa potrebbe
accadere e compromettere la sicurezza.
Prima di proseguire in questa operazione dopo il ciclo di decontaminazione, usare le
necessarie precauzioni: si raccomanda l'uso dei DPI (maschera, guanti, occhiali ecc.) e
verificare mediante idonea strumentazione che la concentrazione di H 2O2 nella stanza

metodo alternativo di decontaminazione che deve essere validato dall’utilizzatore.
9.2.4.1 PROCEDURA
1. Avviare la cabina in MODO 2 (vedi
PARTE 1, 5.2.2).
2. Chiudere il vetro frontale
3. Premere i pulsanti , e

4. Premere il pulsante INVIO (SOCKET).
Il display mostra il messaggio di richiesta password (Fig. A40).
5. Impostare la password corretta tramite i pulsanti SU (LIGHT)
and GIÙ (UV).
La password per la fumigazione è 9.
6. Premere il pulsante . Fig. A41
Se la password è corretta, il display mostrerà il messaggio
FUMIGAZIONE IN CORSO (Fig. A41); la presa elettrica di
servizio è attivata per far partire la prima fase del ciclo
(erogazione del decontaminante).

ATTENZIONE!
• I filtri HEPA devono essere decontaminati in situ prima dello smaltimento.
• Il requisito e il metodo di decontaminazione devono essere scelti mediante
valutazione del rischio (vedi anche par. 9).
Una volta rimossi devono essere sigillati e identificati come RIFIUTO PERICOLOSO.
Pertanto devono essere trattati da aziende qualificate e autorizzate allo smaltimento di tali rifiuti.
◼ Sigillare ogni singolo filtro in un sacco di politene.
◼ Utilizzare contenitori adatti per lo stoccaggio temporaneo.
◼ Identificare il contenitore con etichetta adeguata riportante il tipo di rifiuto.
◼ Avviare le corrette procedure di smaltimento in base alle norme locali in vigore.
