2 Description and Part Numbers
• The cell encapsulation system enables the simple, quick and reliable
encapsulation of single cells, DNA and/or functionalized beads in high
precision, monodisperse picolitre droplets.

• To encapsulate a sample, the user loads sample and reagent into the
100 µl reservoirs, closes the module, and starts the program. The
droplets are collected in the built-in output reservoir. 200 µl of reagent
and sample, containing 300,000 cells, can be converted into 3 million
droplets in 15 minutes. The module can be maintained at 4°C with the
Dolomite Temperature Control Unit, the TCU-100. Droplet production
can be monitored with the Dolomite High Speed Digital Microscope.

• The hardware can be controlled by the µEncapsulator 1 program in the


Dolomite Flow Control Centre software. The necessary reagents for
encapsulation are included in the µEncapsulator 1 Reagent Kit

8 Sample Reservoir Chip


The Sample Reservoir Chip (part no. 3200444) for the
µEncapsulator 1 module contains 2 reservoirs of 100 µl
volume for storage of biological samples at
temperature before droplet formation. Reagents are
pipetted in the inlet ports. There are 2 additional
straight through channels for carrier oil input and
emulsion output. The smooth channel surfaces and precision fabrication provide an
excellent flow path for passage of encapsulated material. The Sample Reservoir
Chip can be used in conjunction with the Droplet Chips (see Section 9) to form 2
reagent droplets.

8.2 Geometry

9 µEncapsulator Droplet Chip


The Droplet Chips for the µEncapsulator 1 module enables 2-reagent droplets
containing biological material to be generated with ease. These chips interface
directly with the Sample Reservoir Chip (part no. 3200444) via the supplied FKM
gasket. It can also be addressed independently for basic tests using the Multiflux®
Linear Connector 4-way (part no. 3000024) and Top Interface 4-way 2.15 mm (part
no. 3000237).

The 2 reagent streams meet immediately before the junction, minimising mixing prior
to the formation of droplets. The Dolomite flow focussing junction design enables
thousands of reactions per second to be carried out in volume-controlled
compartments, isolated by a flowing carrier stream.

The uEncapsulator Chips are available with different coatings (hydrophilic and
fluorophilic) and junction sizes to cover a wide range of droplet dimensions and
volumes:

10 P-Pump
The Mitos P-Pump provides pulseless liquid flow with
a precise pressure driven pumping mechanism. When
used in conjunction with Dolomite flow sensors, flow
rate as well as pressure can accurately be set and
controlled. The P-Pump operates over a wide pressure
range (0-10 bar) with excellent response time and
accuracy. Pressure driven flow is ideal for microfluidic
systems where highly stable flow is required in the
nl/min to µl/min range for applications such as droplet
formation. The design features a lockable pressure
chamber for safety, which is easy to access and
accommodates a wide range of fluid vessels.

11 Sensor Interphase/ Display


The Mitos Sensor Display can be used interchangeably
with the Mitos Flow Rate Sensors (Part Nos. 3200096-
3200100) which simply attach with a push-click action.
Easy to use, the Mitos Sensor Display provides a USB
connection and visual output for displaying micro-scale
flow rate and pressure.

Designed for use with the Mitos Sensor Display (Part
No. 3200095), the Mitos Flow Rate Sensors enable fast
and accurate measurements of ultra-low liquid rates.
The high precision thermal sensor technology provides
total media isolation and extremely low internal volume
with no moving parts.

All measurement data is fully calibrated for water and temperature compensated by
means of an internal microcontroller.

14 Temperature Control Unit
The Meros TCU-100 is an advanced temperature
controller for microfluidic chips and other devices. It
can provide extremely accurate control of a fixed
temperature from 1°C to 100°C, rapid temperature
ramps or thermocycling.
