1.0 PURPOSE:
To describe the standard operating procedure for calibration of micropipettes.

2.0 SCOPE:
This procedure is applicable for calibration of micropipettes used in the microbiology
laboratory.

3.0 RESPONSIBILITY:

5.0 PROCEDURE:
5.1 Receipt of micropipette:
5.1.1 Whenever a new micropipette is received it should be checked for its make, range that
is specified in the purchase order.
5.1.2 Verify if the supplier’s certificate of analysis is present.
5.1.3 Internally calibrate the micropipette given below the point 5.2 as follows.
5.1.4 After calibration that gives satisfactory result, assign the micropipette a serial number
and other details in the format: IM-II/QM/007/F2.
5.1.5 Assign the micropipette serial number as follows:-QM/MP/XX.

5.1.6 Paste the calibration label as per the format: IM-II/QM/007/F3.


5.2 Calibration:
5.3 Clean the external surfaces of Micropipette with clean dry lint free cloth.
5.4 Sanitize the Micropipette with wet cloth dipped with 70% IPA.
5.5 Wipe the Micropipette with dry cloth after sanitization.
5.6 SET the desired volume by adjusting the plunger cap by rotating in clockwise or
anticlockwise direction.
5.7 Keep clean, dry beaker over the balance pan.
5.8 Put the print to tare weight of beaker.
5.9 Fix the Micro pipette with appropriate tip firmly.
5.10 Note the temperature of the purified water used for Performance Verification of
Micropipette.
5.11 Take purified water and adjust its temperature to 25ºC pipette out the purified water.
5.12 Pipette out set volume of purified water and dispense in the beaker.
5.13 Observe the weight and print it then repeat the pipetting and weighing to totally 10
times. From the weight of water calculate the volume as per the following formula.

Three point calibrations shall be performed for the variable range pipette. E.g. 0.01ml, 0 .05 ml,
0.1ml range pipette perform calibration at set point 0.1ml, 0.5ml & 1ml.
5.15 Example calculation
Weight of Water = 100.1 mg
Density of Water at 25ºC = 0.997044 g

5.16 Recorded the observation in the format: IM-II/QM/007/F1
5.17 Acceptance Criteria: the volume variation should be within ±2% of designated set
volume.
5.18 If any deviation observed during calibration of micro pipette the analyst shall be inform
to supervisor for necessary corrective action.
5.19 Frequency of Calibration: Once in three month or after each breakdown.
