A. Add 1-2 drops of povidone iodine in 5 ml of water. Use the color produced as an
indication or basis for negative test results.
B. In small plastic cups, prepare separately a mixture of water (15ml) and a small
sample of each of the following: bread, cooked rice, table sugar (white), mashed
banana, cooked potato, soda cracker, and egg white. Label the mixtures 1-7.
C. Observe the change in color in each upon addition of 1-2 drops of povidone
iodine. A blue/ violet color is an indication of a positive test for iodine.
D. Take a close-up picture of each of your samples.

V. Discussion

1. Which among the samples are positive with the iodine test? Which are not?
Rice, Potato, Bread and Soda cracker turned out positive then Sugar, Egg White and Banana is
negative.

2. Explain the change in color reaction of starch with iodine.


The change of color in starch when iodine is added is said to be because of the formation of an
intensely colored starch-iodine complex, it is because of the charge transfer between the
Povidone- iodine and the starch, the negatively charge iodide acts as a charge donor. The
electrons in such charge transfer complexes are easy to excite higher energy by light hence
resulting to a complementary color that can be observed by the human eye.

VI. Conclusion
In conclusion, the positive samples are those that contain starch, and because starch reacts to
Povidone-iodine, the negative samples are those that do not contain starch. The banana we
used as a sample was ripe, it did not change when iodine was added. The charge transfer
between the starch and the Povidone - Iodine creates an intensely colored starch-iodine
complex, which causes the sample to change color.
