Photodiode receive the scattered light


signals and translate them into electrical
pulses, according to the electrical pulses,
two-dimension map(scattergram) of cell
size and cell internal information can be
obtained.
The X-axis reflects the cell's internal
structure. The Y-axis reflects the cell size.

Baso Channel- Impedance Method


Principle

Electrical Impedance Method is based on


the measurement of changes in electrical
resistance produced by a particle passing
through an aperture.
As blood cells are poor conductive, when
they pass through the aperture, they
cause an increased impedance, the
impedance directly proportioned to the cell
size.

SLS-I hemolysin dissolve RBC and break


WBC cell wall (except baso). Basophils keep
original form .All the other kinds of white
blood cells is shrank.According to the method
of impedance , Baso number and total
number of WBCs can be accurately counted.

Hemoglobin Channel-Colorimetry
 The diluted sample is mixed with lyse reagent, RBC dissolve, releas
ing hemoglobin.
 Hemoglobin combined with lyse reagent to form hemoglobin complex.
 The hemoglobin complex is irradiated by monochromatic light with a w
avelength of 546nm .
 Phototube is used to receive transmission light, and convert the light sig
nal into voltage signal.
 By comparing with the voltage produced by background transmission li
ght intensity before sample adding (only diluent exists), the hemoglobin
concentration can be obtained(HGB), unit is g / L.

Electrical Impedance Method is based on

Principle

The number of pulses is equivalent to the cell number


through the aperture.

The pulse amplitude is proportional to the cell size.

All electrical pulse is classified according to different


channel voltage value.

3.Installation
1.Intallation requirement
Space Requirement
Ensure enough space for instrument maintenance.
The space between the wall and the right & left side of the
instrument ≥50cm
The space between the wall and the back side of the instrum
ent≥50cm
The space between the front of the analyzer and other equip
ment ≥ 100cm;
Ensure enough space under and above the instrument for dil
uent, reagent and waste solution collecting devices.

The instrument should be protected from dust, mechanical vibration,
significant noise and power interference.
It is recommended that the electromagnetic environment assessment of the
laboratory should be conducted prior to test.
Do not use this instrument in close proximity to sources of strong electroma
gnetic radiation, as these may interfere with the proper working.
It should be placed far from the constant ON-OFF electrical devices like bru
sh-type motor, fluorescent lamp.
It should be placed far from heat and wind source, sunlight, brush-type mot
or, flickering fluorescent light and electrical contact equipment.
A well-ventilated place is a must.

2. Unpacking
Check the packaging of the instrument. Contact Dirui or
local distributor for any physical damage.

Place the package in the direction of the arrow upward.

Open the accessory box, mainframe box, check the items


according to the packing list. Contact Dirui or local distributor
for any shortage.

Login the software and find the instrument IP as picture .
Set the computer IP as 172.16.100.XXX to get the communication .

4.Adjustment
1. Sample probe

The life of puncture probe is around 30000 times

The nozzle of the probe should face to the probe base board and at
the same level with outlet of swab
If necessary ,the vertical position sensor of probe could be adjust
also .

2.Probe unit adjustment

The probe unit is fixed on the


base board with 6 screws as
picture show

Enter the “service”—“Advanced” —“Optocoupler position


detect”
Run the “close test’’ to
check the probe position on
Stat position No.3
If the probe does not face
the center of No.3
position ,adjust the screws
which fix the probe unit on
the base board to fit it .

3.Sample loader adjustment


After install or replace the sample loader ,it needs adjustment
as follow :
Enter the “service”—“Advanced” —“Optocoupler position
detect”
Run the “close test’’ Then the probe should move to the top
of No.3 STAT .
Then use paper or something to cover the second sensor of
probe unit .

Then Enter the “service”—“Advanced” —“Mechanical detect



Input 6000 after the “Y-axis’’ and run it .
Then the probe should descend to the center of No.3
STAT .
If not ,adjust the sample loader to fit it .

4.Sampling position adjustment


The correct sampling position
should be as follow :

If the position is not like above picture ,it needs adjustment


as follow :
1.Put a rack with at least 2 tubes on the loader .
2. Then Enter the “service”—“Detect” —“Mechanical check
”—
“Barcode Check’’
Then the rack will move to the mechanical hand position .
Click “Barcode Check’’ again the rack will move to sampling
position
3. If the tube passed the correct position or can not reach
the correct position as above picture show ,we need to
adjust the micro switch as follow to make it correctly .

5.Mechanical hand adjustment


If the mechanical hand has excursion in forward and backward
direction as follow ,then the adjustment is necessary

6.Preesure adjustment

5.Maintenance
1 .Regular Maintenance
Daily Maintenance:
Rinsing Equipment should be carry out every 24 hours .

It could be also setup in setting –Auto cleaning setting


interface, you may choose 10—200 as interval number base on the
sample capacity per day .

Weekly Maintenance:
If rinsing equipment is conducted everyday, the “Soaking with detergent
“ should be carry out every week .

Every 3 Months Maintenance

Clean the Filter for waste valves


Open the left cover of analyzer ,find the valve 3
assembly and remove those 4 filters .
Open the filters and clean the inner side
Close them and fasten it to avoid any leakage
Fix them on valve 3 assembly again

Necessary Maintenance
1.If live the analyzer vacantly more than 2 weeks ,the package clean
and empty should be carry out .

Replace all the reagent with distilled water

2.If the analyzer give alarm as “aperture clog” please carry out
aperture as follow

3.If the analyzer does not startup for a long time ,please prime the

4.Sheath Flow cell Maintenance


Sheath tubing maintenance needs to be carried out when the
instrument tests 7000 samples or it has been used for 3 month.
Flow Cell Soaking
a. Drain the liquid in 4-diff mix pool ( refer to user manual for
operation ) .
b. Pull out the tube 64 connected with valve SV1, and connect it
with waste container.
c. Pull out the tube 42 from connector, and connect with 5 ml
injector with detergent.
d. Push the injector gentlely, push out 4 ml detergent, push and
pull the injector repeatedly, soak for 25 mins, and then push out the
rest detergent.
e. Aspirate 5ml diluent with injector, and conduct rinsing according
to above method.

6.Upgrading

Note : Exit the program before BF-6800 information management syste

7.Troublshooting
1. 4 Diff scattergram does not appear after test or bad scattergram

Analysis :
1.Mixed sample can not reach flow cell
2. Not enough reagent reach 4 diff bath or expired reagent
3.Dirty or blocked

Solution :

Blocked or low sensitivity valve may preve


nt the test syringe from aspirating enough mi
xed sample to the pipeline between SV27 a
nd SV24.

Disconnect the relative pipe connection then


activate the pressure pump , vacuum pump
or check with syringe manually to find the de
fective valve .

Soak it with Naclo 5% for 20min to dissove
the impurity in it .
