Definition of Lipids
• Substances known as fats and oils
• Hydrophobic and soluble in ethanol like alcohol, petroleum and
chloroform
• Classified into 2 general types
➢Fats and waxes (can be hydrolyzed with ester linkages)
➢Cholesterol and steroids (cannot be hydrolyzed)
Type of Lipids
– Saturated fats
• Animal origin—meat, egg yolks, dairy products
• Vegetable oil like coconut and palm kernel oil
– Unsaturated fats
• Found in nuts, seeds, and most vegetable oils like
safflower, corn, cottonseed, sunflower oil and soybean oil
– Cholesterol
• Found in egg yolks, cream, shellfish, organ meats,
and other meats
• Plant product like olive oil, canola oil, avocado and
peanut oil
Lipids Functions
• Excellent energy reserves
• Structure of cell membranes
• Organ padding
• Body thermal insulation
• Essential fatty acids (EFA)
• Hormone synthesis
• Fat soluble vitamin absorption
Lipids in human body
• Reference male 15% body fat; reference female 25%
body fat
• Well-nourished adult meets 80% to 90% of resting
energy needs from fat
– Fat is superior to carbohydrates for energy storage for two
reasons
• Carbohydrates are hydrophilic, absorb water, and expand
and occupy more space in the tissues, and fat is
hydrophobic, contains almost no water, and is a more
compact energy storage substance
• Fat is less oxidized than carbohydrates and contains over
twice as much energy: 9 kcal/g for fat; 4 kcal/g for
carbohydrates
Energy source in human body
• Fat has glucose-sparing and protein-sparing effects
when used for energy needs
– Glucose is spared for consumption by cells that
cannot use fat, like neurons
– Protein not catabolized for fuel
• Fat-soluble vitamins (A, D, E, K) absorbed with dietary
fat
– Ingestion of less than 20 g/day risks vitamin
deficiency
Diverse functions besides energy source
– Structural
• Phospholipids and cholesterol are components of
plasma membranes and myelin
– Chemical precursors
• Cholesterol—a precursor of steroids, bile salts,
vitamin D
• Thromboplastin, an essential blood-clotting factor,
is a lipoprotein
• Fatty acids—arachidonic acid and linoleic acid:
precursors of prostaglandins and other eicosanoids
– Important protective and insulating functions
Lipids Deficiency
• Fat should comprise of 3% of total calories to prevent fatty acid
deficiency
❖Fatty acid deficiency syndromes
Dry scaly skin, dermatitis (Linoleic acid deficiency)
Hand tremors (Prostaglandin deficiency)
Inability to control blood pressure
Lipids Exceeding
• Requirements: should be less than 30% of daily
calorie intake
– Typical American gets 40% to 50% from fat
• Most fatty acids synthesized by body
– Essential fatty acids must be consumed
• To prevent overtaking, we should consume
fat breakdown (% total calories)
Less than 8% from saturated fat
10% from polyunsaturated fat
10-15% from monounsaturated fat
Cholesterol
• Plant and animal food contain sterols but only animal food contain
cholesterol
• A structural part of the cell membrane
• A precursor to vitamin D
• A precursor to bile acid
• Precursor for sex hormones such as estrogen and testosterone
• Dietary recommendation: less than 300 mg/day
• Sources – egg yolks, liver, shellfish, organ foods
Lipoproteins
• Low Density Lipoproteins (LDL) – is made by the liver and is comprised
of cholesterol that is delivered to the cells in the body
– High levels of LDL is strongly correlated with heart disease
• High Density Lipoproteins (HDL) - made by the liver and picks up
cholesterol from the cells fro recycling or excretion
– High levels of HDL is inversely correlated with heart disease It is
protective

• Levels of HDL
– High level of HDL is beneficial
– Indicates that cholesterol is being removed from the
arteries and transported to the liver for disposal

• Recommendations
– Increase your ratio of HDL to LDL
– Regular aerobic exercise
– Avoid smoking, saturated fats, coffee, stress
Lipid Metabolism
Lipids
• Triglycerides are stored in adipocytes
– constant turnover of molecules every 3 weeks
• released into blood, transported and either oxidized
or redeposited in other fat cells
• Lipogenesis = synthesizing fat from other sources
– amino acids and sugars used to make fatty acids and
glycerol
• Lipolysis = breaking down fat for fuel
– glycerol is converted to PGAL and enters glycolysis
– fatty acids are broken down 2 carbons at a time to
produce acetyl-CoA (beta oxidation)
Lipids
– Beta oxidation in the mitochondrial matrix
catabolizes the fatty acid components
• Removes 2 carbon atoms at a time which bonds to
coenzyme A
• Forms acetyl-CoA, the entry point for the citric
acid cycle
– A fatty acid with 16 carbons can yield 129
molecules of ATP
• Richer source of energy than the glucose
molecule
Lipogenesis & Lipolysis Pathways
Ketogenesis
• Fatty acids catabolized into acetyl groups (by beta-
oxidation in mitochondrial matrix) may
– enter citric acid cycle as acetyl-CoA
– undergo ketogenesis
• metabolized by liver to produce ketone
bodies
– acetoacetic acid
– -hydroxybutyric acid
– acetone
• rapid or incomplete oxidization of fats raises blood
ketone levels (ketosis) and may lead to a
pH imbalance (ketoacidosis)
