Analysis Principle-blood cell counting

Principle : Electrical impedance method-blood cell counting.

Analysis Principle-blood cell counting

WBC chamber: analog signal Digital signal

RBC are hundreds of times more than WBC, and some parts of WBC are in the same size
as RBC, so the purpose of adding lyse, is to dissolve the RBC.
Meanwhile, PLT still exist in WBC chamber, but signal of PLT is very weak, easy to be
affected by broken RBC pieces, that’s why WBC chamber don’t test the result of PLT.
Analysis Principle-blood cell counting

RBC signal
RBC chamber: analog signal Digital signal

When test RBC and PLT, WBC signal is also included. WBC result have been tested in WBC
chamber, so software can easily calculate precise RBC result.

PS: 1. RBC and PLT are tested in the same counting chamber.
2. WBC and HGB are tested in another counting chamber.
Analysis Principle--HGB measurement

Principle : Colorimetry
Lyse HGB
(Dissolved) HGB React
compound LED530nm

Colorimetry: C = F × A
HGB (C) = constant factor × lg (Blank light intensity / sample light intensity)

Sample in WBC counting chamber.

Signal: 1. Blank light intensity (Colorless Diluent in chamber)
2. Sample light intensity (Red color sample solution in chamber)
Function of diluent and lyse

Dilute blood cells and

make an even distribution.

Act as a good conductor in Dissolve RBC, then release

the electric circuit. and react with HGB.

Trouble Shooting

www.zybio.com
Trouble Shooting-Aperture clog

How does the analyzer report clog error?

During blood cell counting procedure, blood cells pass through
aperture and the analyzer count blood cells signal. This process
spent about 10 seconds. The analyzer divides this process into 8
parts. The analyzer will get RBC WBC PLT result for each part
then compare the results between these 8 parts. If it is clogged
during counting, there will have a big difference between some
parts. In this way the analyzer can judge whether the aperture is
clogged or not. Also we call it fixing time method.
Trouble Shooting-Aperture clog

How does clog error appear?

If we test sample with the analyzer, but don’t do maintenance very
well, some protein or lipid will adhere on the aperture or tubes.
After we use it for a long time, these protein and lipid will
accumulate and finally cause the aperture clogged.

How to remove clog error?

Use unclog function to remove clog with high voltage shock and
back flush. If it doesn’t work, do Whole Device Cleanser Soak
operation with probe cleanser. If it still doesn’t work, we need to
disassemble the counting chamber to clean WBC/RBC aperture
manually.

Blank result abnormal:

1. Check whether power supply is stable and whether power grounding is good?
If power is stable and power grounding is good, go for next step.
2. Temperature is high also can make PLT high result (Operation temperature:
15-35 ℃).
3. Check whether reagents are expired or contaminated. If yes, replace it.
4. Try to solve the problem by unclog, cleaning, whole device soak
function(Tubing dirty, lipid/protein may adhere on aperture.).
5. Check whether back bath tube is dirty or not. If it is dirty, clean it or replace it.
6. If there are any dust or crystal adhere to sample probe, sample probe wash
set, shielding box or counting chamber, clean it with alcohol manually.
Reagent issue

Reagent alarm :
A. Reagent empty
1. Check whether the corresponding reagent bottle is empty.
2. If yes, replace new reagent and load the barcode.
3. If reagent is enough and alarm still exist, enter Management => Status => Volt./Current
interface to check the reagent sensor voltage(0~2.5V means empty, 3.5V~5V means full.).
4. Enter Management => Service => Prime interface, run prime function to avoid bubble in tube.
B. Reagent expired
1. Enter Management => Setup => Reagent menu.
2. Select “One key elimination” to activate clear reagent expired alarm function. Dis-select
“Reagent expired” option and click OK to save.
3. Then we can click the alarm area, on the pop-up dialog box have one key clear alarm function.
Click clear to cancel the alarm and close the dialog box, then we can continue use the reagent.
C. Reagent run out
Reagent use up, need to load a new reagent.
HGB blank abnormal

HGB adjustment:
1. Enter Management=> Setup => System=> Gain.
2. Change HGB gain value to get HGB blank value around
4.5±0.1V, then save HGB gain.
Counting chamber overflow issue

WBC chamber overflow :

(Ps: Use hydraulic diagram to check it)

1. WBC bottom blocked (crystal or some particles )

2. Buffer bath blocked (strainer dirty)
3. Valve 9 blocked or defective
4. liquid pump defective
5. The tube is squeezed

RBC chamber overflow :

1. RBC bottom blocked (crystal or some particles )

2. Buffer bath blocked (strainer dirty)
3. Valve 8 blocked or defective
4. liquid pump defective
5. The tube is squeezed

Exchange RBC(J5) and WBC(J7) signal

Only one chamber’s counting result is 0: cable on main control board (Need
to disconnect J4 cable).
WBC Chamber: WBC&HGB RBC Chamber: RBC&PLT After exchange, here will have four
kinds of conditions.
For example: WBC chamber’s counting result is 0, and RBC
chamber’s counting result within range

WBC RBC Conclusion

1 Value Value Reconnect corresponding cable

2 Value 0 WBC chamber issue
