Manipulating Cell

Migration Using
Amphiphiles
11/20/15 Meeting

Last Time
Images taken from
PDMS column
experiment
At first glance,
experiment seemed
to support hypothesis

Benzyl Alcohol
FIJI used to stitch together images. Then cells manually counted by
hand.
Total number of migrated cells: 266
Average number of migrated cells: 89

Control
Total number of cells migrated: 297
Average number of cells migrated: 99

Comparison
Only 31 more cells migrated in total in the control
This is only an increase of about 10-12%

On average, only 10 more cells migrated per well in the

control model.

Validity Issues
There are cells here
that could not be
captured in the image
The areas of each image are not
consistent and there is not a
good way to measure
Entire area covered by column
would need to be imaged which
would be very time consuming

New Experiment

Originally tried just using small pipet tips not effective

Filled pipets with PDMS, let small bit drip out bottom
New columns taped to bottom of well plate

Problems
Boundaries not very well-defined
Some wells unusable- not enough to complete
experiment

Where to go from here

Manufactured cell migration assembly too
expensive ($255 for one kit)
Two options:
1. Repeat last experiment but with more columns
2. Use larger diameter columns
