Focusing with the Microscope

I. Objectives:
1. Identify the parts and describe the function of each part of the microscope.
2. Practice proper care and handling of the microscope.
3. Develop skill in preparing specimen on glass slides for microscopic examination.
4. Develop skill in the manipulation of the microscope to observe microscopic specimen.
5. Record microscopic observations correctly.

II. Materials Needed:


1unit Light Microscope
1 pc. Scissors or cutter
5 pcs. Glass slides with cover slips
3 pcs. Threads (blue, yellow, red)
5 pcs. Small letter initial (small letter e) cut out from newspaper and magazine
5 pcs. Hair strand
1 bottle Petroleum jelly
10 pcs. Toothpick
1 bottle Pond water/Hay infusion
2 pcs. Medicine Dropper
Groups: Reuse the Chemistry Laboratory Kit and add the following: Newspaper, alcohol, masking
tape, marker
Individual Students: Gloves, Mask, Laboratory Gown

IV. Data and Observations:


Orientation
A. Dry Mounts
1. Paste the image of threads in scanner and LPO below. Draw the threads as seen in the HPO in the blank circle.

2. Paste the image of hair follicle in scanner and LPO below. Draw the hair follicle as seen in the HPO in the blank circle.

B. Wet Mounts
1. Paste the image of the letter “e” in scanner and LPO below. Draw the letter “e” as seen in the HPO in the blank circle.

V. Analysis:
A. Orientation and Magnification
1. Described the appearance and orientation of the thread.
-when we look in the microscope the appearance of the threads pilled together and the orientation of image is still the
same that the threads form like an asteris (*) as we put it in the condenser.
2. Described the appearance and orientation of the hair.
-as we look into the microscope, the appearance of hair in the scanner is that there’s a black line towards the hair and as
we watch in HPO, the hair covers with more transparent layers/tissues and the orientation of the hair is still the same as we
put in the condenser but the image goes bigger when we used the HPO.
3. Described the appearance and orientation of the letter.
-as we look into the microscope, there are something colorful dots in the appearance of the letter”e”and we put the letter
“e” in the correct position of the orientation but when we looked into the microscope, the orientation changed and the
letter “e” inverted.
4. What is the total magnification in each objectives:

5. In what direction does the image move as you move the slide slowly

B. Proper Handling and Care of the Microscope


1. Why do you have to handle and care of the microscope properly?
- As a student we should have to handle and care of the microscope properly because microscope is very expensive
it is important to make them last for a long time.
2. Did you encounter difficulties while handling the microscope? If yes, how did you address the difficulties?
- Yes, we encounter difficulties while handling the microscope because it is our first time to handle microscope and
we have not yet known the function of every parts of the microscope.

A. Focusing the Microscope


1. How many tries have you made before you get a clear focus of the letter “e”?
- For our group it takes 5 times before we get a clear focus on the letter” e” in the microscope.
2. What values have you learned while doing this part of activity?
- For our group the value that we learned while doing the activity is the value to be patience because without
patience you will never get a clear focus of the specimen.
3. Why do you need to focus specimens under the LPO first before focusing under the HPO?
- As a student we need to focus specimens under LPO first before focusing under HPO because it allows you to
center the specimen, and if it is set up correctly, then when you switch objective it should be there and only need
fine focusing. It also reduces the chance of crushing the specimen or breaking the slide or the lens as the LPO is
further away from slide than the HPO.
