The leaners shall be able to identify parts of the microscope and their functions by:
1. Identifying the parts of the compound microscope;
2. Describing the function of each part of the compound microscope.

I. CONCEPT NOTES

Microscope- is an optical instrument used for viewing very small objects, such as mineral samples or animal
or plant cells, typically magnified several hundred times.

Parts and function of the Microscope:

 Eyepiece Lens- is present at the top and is used


to see the objects under study.
 Tube- it connects the eyepiece to the objective
lenses.
 Revolving nosepiece/ Turret- it allows the
rotation of the lenses while viewing.
 Objective lenses- it is the colour coded of the
lenses, the shortest lens is of the lowest power,
and the longest lens is high power lenses.
 Coarse adjustment knob- it is used for focus on
scanning.
 Fine adjustment knob- Is used for focus on oil. It
moves the body tube for focusing the high power
lens.
 Arm- It supports the tube of the microscope and
connects to the base.
 Stage- is used for placing the slides under
observation.
 Condenser- it is for the focussing on the light of
the specimen under observation.
 Base- It provides basal support for the
microscope.
 Power switch- it turns the illumination on or off.

Mechanical- it is working or produced by machines or machinery.

Mechanical parts of a microscope

1. Body tube- holds ocular and objective lenses at the proper distances.
2. Arm- supports the body tube and stage; forms a convenient handle.
3. Revolving nosepiece- holds and permits interchange of objective lenses.
4. Stage and slide holder- supports slide
5. Stage and control knobs- move the slide; for centering and scanning images.
6. Diaphragm lever- open and closes the iris diaphragm, controls the amount of light passing
through the stage.
7. Coarse adjustment knob- aids in focusing by moving body tube up and down.
8. Fine adjustment knob- exact focusing.
9. Base- supports the microscope.

Magnifying parts- magnified the object being viewed deep on, look on microscope and it is where
Magnification happens.
1. Ocular/eyepiece- magnifies specimen by 10-15 times
2. Objectives:
a. Low-power Objective- 10-15 times-general view on specimen
b. High-power Objective- 43-100 times-detailed parts of the specimen
c. Scanning Lens- 5 times-general view of the specimen
d. Oil Immersion Objective- 10000 times-used only for viewing bacteria, needs cedar oil.

Illuminating parts- it produces light.


1. Mirror- reflects light
2. Condenser- concentrates light
3. Sub-stage Condenser- allows light to pass
4. Iris Diaphragm- controls the passage of light.

I. CONCEPT NOTES
The microscope has 3 magnifications: Scanning, Low and High. Each objective will have written the
magnification. In addition to this, the ocular lens (eyepiece) has a magnification. The total magnification is the
ocular x objective.

Procedures in Focusing Specimens


1. Always start with the scanning
objective.
Odds are, you will be able to see something on
this setting. Use the Coarse knob to focus,
image may be small at this magnification, but
you won’t be able to find it on the higher
powers without this first step. Do not use
stage clips, try moving the slide around until
you find something.

2.Once you’ve focused on scanning, switch to


Low Power. Use the Coarse knob to refocus.
Again, if you haven’t focused on this level, you
will not be able to move to the next level.

3.Now switch to High power. (If you have a


thick
slide, or a slide without a cover, do NOT use
the high power objective). At this point, ONLY
use the fine adjustment knob to focus.

4.If the specimen is too light or too dark, try


adjust the diaphragm.

5.If you see a line in your viewing field, try


twisting the eyepiece, the line should move.
