ACTIVITY: Work instruction for Microscope

PURPOSE
To lay down procedure for the operation of Microscope.

SCOPE
This Procedure is applicable for observation of specimen biological

RESPONSIBILITY
QC in charge is responsible for supervision of the activity. QC Microbiologist is
responsible for maintenance and operation of the instrument.

PROCEDURE
1. Switch ON the instrument using ON or OFF switch
2. Set the required brightness trough the brightness control
3. Position the slide with the sample to be observed on the stage.
4. Adjust the slide to bring in to exact center of the light source.
5. Low magnification lens to be focused first followed by high magnification
lens to the center of the sample.
6. Turn the coarse adjustment knob to adjust the stage.
7. While observing through the ocular bring the object in to focus by adjusting
with the coarse knob.by adjusting fine adjustment knob sample can be
focused for better resolution.
8. Adjust the diaphragm to increase or decrease the light intensity.
9. Once the image is visible adjust the slide for the targeted item in the
specimen.
 10. To proceed from low power to high power magnification rotate the high
power objective in to position.
11. Whenever the object is to be observed under oil immersion, place one drop
immersion oil and bring the 100 X oil immersion lens in to position.
