2 Enzymatic degradation of cellulose

Note : In the ensuing discussion the concentration rate of a particular species is


simply the rate v multiplied by the concentration

a) The rate of formation of polymers of a particular length i units ( dx ) will be
due to the rate at which the enzymes Endoglucanase (EG) and
Cellobiohydrolase (CBH) cuts the polymeric chain times the concentration
of the polymeric chain that is cut minus what is already there.

It is assumed that EG can cut chains of original length 3or greater and that each
bond in the chain has equal probability of being cut.

Hence looking ( dt ) enzyme (EG) will break a a chain lengh of length 4 to form of
a polymer of chain length 3 and 1 ,while polymers of chain length 4 disappears
dueto it being attacked .Chain length 4 can be attacked at any linkage (hence the
probability of 3) .

A Cellobiohydrolase (CBH) enzyme cuts off a single unit of length 2, from one end
(the reducing end) of the polymeric chain to produce a ( c3 )

While any ( c3 ) present will be attached and hence the concentration falls.

The rate of formation of polymers with 2 units ( dx ) will be due to the sum
of the rates at which the enzymes Endoglucanase (EG) and Cellobiohydrolase
(CBH) cuts the polymeric chain times the concentration of the polymeric chains
present minus the rate at which Beta-glucosidases (BG) removes the ( c2 )
polymer units that are formed to form glucose.

Looking closer at the formulas -

The rate of formation of ( c2 ) is


doubled because the enzyme (EG) can cut the chain at either end . This is the
“feed” material for the enzymes (EG) and (BH) to make smaller chains (in
particular ( c2 ) .Note (BH) removes ( c2 ) )

In this case when there are


chain lengths of 4 ( c4 ) the rate is doubled because the chain is “symmetrical “
and can be cut at either end.

This term is the rate at which Beta-


glucosidases (BG) removes the ( c2 ) polymer units that are formed to form
glucose. That is why it is subtracted.
c) Inspecting each term

The rate of formation of ( c1 ) is


doubled because with the enzyme (EG) can cut the chain at either end .This is the
“feed” material for the enzymes EG and BH to make smaller chains (in particular
( c1 ) )

For chain lengths of 3 ( c3 ) the


enzyme (BH) will attack a ( c3 ) polymer to form a ( c2 ) and a ( c1 ) hence the
multiplication of the rate with ( c3 )

the last term is doubled due to the fact


that the molecule is ‘’symmetrical ‘’ and there is equal chance of splitting a ( c 2 )
to form a ( c1 )
