Ninhydrin Rxn

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, glycine, and
H2O
 Add 2-3 drops 0.1% Ninhydrin Solution to each sample
 Cover each sample with a marble
 Water bath to boil for 1-2mins
 Cool
 Observe

B. Biuret Test

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, glycine, and
H2O
 Mix 0.5mL 10% NaOH to each sample
 Add 1-2 drops 0.5% CuSO4 in each sample
 Observe

C. Xanthoproteic Test

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, tryptophan,
tyrosine, phenylalanine, histidine, and H2O.
 Add 0.5mL conc. HNO3 to each sample
 Observe for heavy white precipitate is formed
 Heat and Observe for formation of yellow color
 Record color of solution
 Cool
 Make alkaline with 10% NaOH.
 Observe

D. Millon-Nasse Test

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, tyrosine, and
H2O
 Add 3 drops Millon-Nasse reagent to each sample
 Water bath to boiling for 10 mins.
 Cool in ice bath
 Add 0.5mL 0.1% NaNO2
 Warm gently
 Observe
E. Hopkins-Cole Reaction

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, tryptophan,
and H2O
 Mix 0.5mL Hopkins-Cole reagent to each sample (observe presence of violet ring)
 Layer each sample with 1mL conc. H2SO4
 Observe

F. Bromine Water Test

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, tryptophan,
and H2O
 Add 2 drops of bromine water to each sample
 Shake each sample with 0.5mL n-amyl alcohol
 Observe

G. Pauly Reaction

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, histidine,
tyrosine, and H2O
 Mix 0.5mL cold sulfanilic acid with 0.5mL cold 0.5% NaNO2 to each sample
 Cool in ice with constant shaking for 3 mins.
 Add the specified amino acid and protein samples in separate test tubes with 10% Na2CO3
 Observe

H. Lead Acetate Reaction

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, cysteine, pcs of
hair, and H2O
 Add 0.5mL 20%NaOH and 2 drops 10% Pb(OAc)2 in each sample
 Cover each sample with a marble
 Water bath to boiling for a few mins.
 Observe

I. Sakaguchi Reaction

 Prepare on separate test tubes: 0.5mL of saturated solutions of casein, albumin, arginine, and
H2O
 Mix 1mL of 20% NaOH in separate test tubes in each sample
 Add 0.5mL Molisch reagent in each sample
 Mix thoroughly
 Add 0.5mL bromine water in each sample
 Observe
