EFFECT OF TEMPERATURE ON ENZYME ACTIVITY
Introduction:
The activity of an enzyme measures the rate at which enzymes converts substrate
to products. One factor that affects the activity is the temperature. As the temperature is
increased, the molecules move faster and collide more frequently. The substrate and
enzyme collide as temperature is increased until it reaches its optimum temperature that
is the temperature at which the enzyme works at its best. Each enzyme has its own
optimum temperature. Human enzymes usually have optimum temperature at 37oC.

Objectives: At the end of the activity, the student can :


1. Determine the effect of temperature on reaction rate below and
above the optimum.
2. Determine the optimum temperature of the enzyme used.

Reagents: Saliva, 4 ml 1% starch, 4 drops iodine in KI, 4 ml benedict’s reagent.


Materials: Thermometer, 8 test tubes (20 ml), test tube rack, brush, holder, tripod,
wire gauze, 250 ml beaker, 10 ml graduated cylinder, dropper, alcohol lamp
Procedure:
1. Prepare 5 test tubes. Place 1 ml of 1% starch and add 10 drops of saliva to each
tube. Mix thoroughly.
2. Place the first tube in ice water, the 2nd tube leave at room temperature, the 3rd
tube in 400C , the 4th tube at 60oCwater bath and the 5th tube boil for 2 minutes..
3. Leave the 4 tubes in their respective temperatures for 30 minutes. The 4th tube
allows to stand for 30 minutes after heating for 2 minutes.
4. Test the contents of each tube with iodine and benedict’s tests. (Follow procedure
in activity 8).
