THE EFFECT OF TEMPERATURE

TO THE CATALYZE ENZYME ACTIVITY

A. Experimental Objectives:
▪ To analyze the function of catalyze of enzyme to the chemical process in our body
▪ To conclude the effect of temperature to the work of catalyze enzyme.

B. Basic Theory:
How do living cells interact with the environment around them? All living things possess
catalysts, or substances within them that speed up chemical reactions and processes. It also happen
inside of the human body. One of the examples is in our liver. Our liver has function to detoxify the
poison which inserted to our body. It produces catalysis in the form of enzyme to make the process
faster.
Enzymes are molecules that enable the chemical reactions that occur in all living things on
earth. Enzyme also has optimum temperature to work well. In this catalase and hydrogen peroxide
experiment, we will discover how enzymes act as catalysts by causing chemical reactions to occur
more quickly within living things. Using a potato and hydrogen peroxide, we can observe how
enzymes like catalase work to perform decomposition, or the breaking down, of other substances.
By doing different treatment at the temperature of potato we will also know the optimum temperature
of enzyme to work well. Catalase works to speed up the decomposition of hydrogen peroxide into
oxygen and water.

F. Apparatus and Materials

Apparatus:
- 3 reaction tube
- 3 graduated tube/beaker glass
- Knife
- Ruler
- Pipet
- Thermometer

Materials:
- Potato
- Water
- Ice cubes
 - Hydrogen peroxide (H2O2)

G. Step:
1. Peel the potato
2. Cut it down into three same size. The size is 2 cm x 1 cm x 0,5 cm.
3. Keep one section raw and at room temperature.
4. Place another section into the reaction tube filled with water then put it at the container filled
with ice cube in 10 minutes.
5. Put into the hot water for the last section in 5 minutes.
6. Measure the temperature of the three sections of potato.
7. Use pipet to put the 20 ml hydrogen peroxide to the 3 graduated tube
8. Put the three sections of potato to the different graduated tube.
9. Observe the reaction that happen and see the adding bubbles.
10. Measure the volume of bubble after 20 seconds.
11. Write in the data table.
