3.3 Analysis Procedure

This section introduces the analysis procedure of the LW C100plus by actions, action
positions, testing flow and measuring points.

3.3.1 Actions
The LW C100plus finishes all the tests by performing the following steps repeatedly:

 The reaction disk rotates to carry the cuvettes to the first set of wash probes and

wash them automatically.

 The reaction disk rotates 87 cuvettes positions (during rotating the absorbance of the

reaction liquid is measured) and then pauses; rotates to R1 dispensing position,

pauses; rotates 90 cuvettes positions, then pauses. This whole process is called one

period.

 During the 1st and the 81st period, R1, sample and R2 are dispensed and the reaction

is monitored. Absorbance measurement is performed once in each period.

 In the 81st period, the cuvettes are carried to the first set of wash probes and then

washed. After that, the next cycle begins.

Chapter 4 Board Description

The LW C100plus consist mechanically of the following boards and assembly:
 Main board
 Rear board
 Pump/Valve drive board
 Fluid level detection board
 AD collection board
 Power supply assembly

4.1 Main Board

4.1.1 Overview
The main board is the control center of the hardware system and consists of six functional units. Each of
these functional units is controlled by a CPU and communicates with each other, thus constituting the entire
control system.

LW C100plus Service Manual 26

Chapter 4 Board Description

4.1.2 Main Unit

(1) Functional block diagram

(2) Function description

 Communication: The main unit communicates with PC via the RS232C serial port, and also communicates
with the five control units via the Complex Programmable Logic Device (CPLD) and the serial port
extension module.
 Interruption: The probe unit, mixer unit, sample/reagent disk unit and temperature unit have an interrupt
signal respectively, and share the same interrupt of the main unit CPU with CPLD via the serial port chip
ST16654. The main unit sends data to the extension address of the corresponding serial port. After
receiving data from a control unit, the relevant serial port sends an interrupt signal to the main unit. Each
of the four serial ports has an interrupt output. The photometric unit uses a single interrupt to
communicate with the main unit.
 Logic: The logic function is used for three purposes, which include: 1) interrupt control of the four
interrupt signals; 2) register extension of the RESET/PSEN port for downloading the control unit program;
and 3) extension of the address latch.
 Extension of SRAM: A 32K Static Random Access Memory (SRAM) is extended for data buffer. The
lower 8-bit address bus of the SRAM is obtained via the data bus of the address latch CPU within the
CPLD.
 Reset: The system provides two resetting modes: power-on reset and manual reset. By using manual reset,
all chips will be restarted.

Chapter 4 Board Description

4.1.3 Reaction Disk and Photometric Unit

(1) Functional block diagram

(2) Function description

 Communication: The photometric unit receives commands from the main unit via a serial port.
 Control of reaction disk: controlling rotation of the reaction disk and collecting photometric data of
corresponding channel.
 Control of AD collection: When the photometric unit outputs a control signal for AD conversion, an AD
conversion is started, and the photometric unit reads the converted photometric data via the extended
Serial Port Interface (SPI) of the CPLD.
 Control of motors and light source: The photometric unit outputs the control signals of stepper motors,
stepper pulse and light source to the power driving board.
 Signal detection: The photometric unit also detects the signal of the reaction disk initial position sensor.
 Description of photometric data collection: When receiving a photometric data collection command from
the main unit CPU, the reaction disk and photometric unit carries the corresponding cuvette to the 340nm
wavelength for channel 340, and then starts AD conversion and collects 8 data for each channel. When
collection of the channel is finished, the photometric unit proceeds to the next channel and collects data
for the same cuvette. After data collection of the cuvette is complete, a new collection cycle starts when
the next cuvette reaches to the 340nm. During the process of AD data collection, the light spot of each
channel should stay in the same cuvette for the same collection cycle.

Chapter 4 Board Description

 AD collection: The photometric signals output from the eight detection circuits are filtered and amplified,
and then gated through multiple channels.
 Photometric conversion: The eight conversion circuits convert the light intensity signal transmitted
through the reaction cuvette into voltage signal, and then send it to the input terminal of the AD converter.
The AD conversion board receives control signal from the main board, collects the processed eight
photometric signals, and then sends the AD values of light intensity to the main board for processing.

4.4.2 Eight Photometric Conversion Circuits

The eight light rays are respectively converted by a photometric conversion board of the corresponding
wavelength. The light transmitted for different wavelength is of different intensity, among which ultraviolet ray
is the weakest while visible light is the strongest. The gains for photometric conversion boards vary with
wavelength, and therefore, cannot be exchanged.

4.4.3 Circuit of AD Collection Board

The AD collection board consists of the signal processing circuit, multiplexing and AD amplifying circuit.
(1) Signal processing circuit
The signal processing circuit consists of eight filters that are used to input photometric signal and the gain
adjustment circuit. The following figure shows the principles of the signal processing circuit:

Flat cable
Conversion circuit Primary Secondary
Amplifier Multiplex
filter filter

Manual gain adjustment

The dotted frame shows the structure of the eight-channel signal processing circuit. The input photometric
signal is filtered through two stages and amplified on the AD collection board. Manual gain adjustment is
applied on the amplifying circuit in order to make the output values of all signals reach the maximum. As the
light source is attenuated to decrease the power and the amplitude of output signals, manual gain adjustment
may help make the output values of all signals reach the maximum.
(2) Multiplexing and AD collection
The eight processed photometric signals are transmitted to the input end of the multiple selector and then
routed to corresponding channel for AD conversion. The AD converter is of 16-bit for resolution.
(3) Power supply of AD collection board
The AD collection board is powered by an analog power supply of ±15V and a digital power supply of 5V,
which are all introduced via the main board.
The 5V of the AD977AR is stabilized linearly from the 15V power supply of the AD collection board,
which is transmitted via a five-wire shield cable.

4.5 Fluid Level Detection Board

4.5.1 Overview
The analyzer is capable of fluid level detection, which prevents the probe from entering reagent or sample
too deep to cause carryover and influence the test result, and from aspirating no reagent or sample to bring
incorrect test result due to insufficient reagent or sample volume.

4.5.2 Structure of the Probe

change of phase. The phase detector then outputs an error voltage, which is then filtered by the low-pass filter.
When obtaining the change, the band-pass filter shapes it and then sends it to the CPU of the main board. The
fluid level detection board then outputs a positive pulse signal.
(4) Power supply of fluid level detection board
The fluid level detection board is powered by 12V, which is provided by the main board and converted to
5V and 9V. 9V is used to power the phase-locked loop and 5V to power other elements on the board.

4.6 Power Supply Assembly

4.6.1 Overview
The entire system is powered by the power supply assembly with the operating voltage within
100V-240V.
The power supply assembly provides the following voltages: 24V, 12V and 5V. The figure below shows
the principle of the power supply assembly.

4.6.2 Description of Power Supply Assembly

The commercial power 110V or 220V is first filtered and protected against leakage and then converted into
four power supplies.
As shown in the figure of section 4.6.1, the power supply assembly has four outputs.
 12V for power supply 1 to power the light source.
 5V for power supply 2 to power the main board and the rear board.
 12V for power supply 3 to power the rear board, the fluid level detection board and the pump/valve
driving board.
 24V for power supply 4 to power the heater, refrigerator, cooling fans and stepper motors. Power supply
2-4 join together to provide power supply (5V, 12V and 24V) for the rear board.

Chapter 9 Appendices

9.1 Terminology

9.1.1 AD Value
The light that reaches the detector generates current, which is amplified and then transferred

to photovoltage (analog signals) after passing through the fixed resistor. The photovoltage is

converted to corresponding numeric value via the AD (analog-digital) converter. The obtained

numeric value is an AD value.

9.1.2 Dark Current

Dark current refers to the AD value obtained when the lamp is turned off, e.g. no signal light is

transmitted. It is equivalent to the circuit background and should be subtracted during absorbance

calculating.

9.1.3 Water Blank

Water blank refers to the absorbance of a cuvette filled with purified water..Absorbance is a

relative concept and based upon the standard absorbance value. In the LW C100plus, the

absorbance of the water blank is 0 and should be subtracted for calculating any other absorbance.

9.1.4 Measuring Point

The specific moment for photoelectric measuring is called a measuring point and indicated by

a numeric value. Each measuring point is strictly determined and closely related with one another.

In the LW C100plus, each reaction process is divided into 71 measuring points. The interval

between two is 24s in fast mode and 36s in normal mode.

9.1.5 Absorbance
Absorbance is the logarithm of minus quotient between the transmitted light intensity and the

input light intensity. In the LW C100plus, transmitted light intensity is the AD value of a reaction

cuvette filled with distilled water; the final absorbance is the product of the calculated value ×

20000.

9.1.6 Reaction Curve

Reaction curve consists of several points with measuring point as the X-coordinate and
LW C100plus Service Manual 102
Chapter 8 Troubleshooting
absorbance as the Y-coordinate. The following figure shows a typical reaction curve of the
LW C100plus.

9.1.7 Response
Response refers to the absorbance change before and after the reaction, or the absorbance

change rate during the reaction process.

9.1.8 Calibration
Calibration is a sample measurement, during which one or more samples (or calibrators) with

given concentration (or activity) are measured to get its response. According to the set calibration

method (linear or nonlinear), an optimal curve is fit based on the given concentration and the

calculated response and used to generate an equation. By using the curve, a sample can be

measured to get its response, which is then employed to calculate its concentration or activity.

9.1.9 Calibration Curve

Calibration curve is a curve fit by the optimal equation based on several points with

concentration (or activity) as the X-coordinate and response as the Y-coordinate.

9.1.10 Calibration Parameter

Calibration parameter refers to any other item rather than concentration and response in a

calibration formula.
