Investigating the effect of temperature on respiration rate of germinating seeds

A. Guidelines and Instruction

4. Write down your answer in Google Doc / MS Word, and turn it in through google
classroom before 9 PM on November 9, 2021.
5. Late submission will result in a deduction of 2 points/day and should not be more than 5
days. If so, the student will get 0.

B. Introduction
Respiration is the process during which simple carbohydrates, like glucose, break down
into simpler substances and liberate carbon dioxide and energy. The compound used, or
oxidized, during respiration is called a respiratory substrate. Carbohydrates, fats, and proteins
are examples of respiratory substrates, and carbohydrates are the preferred respiratory
substrate among them. The rate of respiration can be measured in terms of gas exchange, that
is consumption of the respiratory substrate oxygen or evolution of carbon dioxide. By respiration
rate we mean the uptake of oxygen per unit time.
Respiration, like most of the chemical reactions in living things, is controlled by enzymes
and so is affected by temperature. At a very high temperature, the rate of respiration decreases
with time, and at very low temperature, the respiration rate is insignificant. Respiration in plants
can be studied in moist germinating seeds that release carbon dioxide (CO2) during respiration. The
seeds are kept in an air tight conical flask. A small test tube containing potassium hydroxide (KOH)
solution is placed in the flask to absorb carbon dioxide released by the respiring germinating seeds.

C. Simulator Procedure
(as performed through the Online Labs)
1. Go to the laboratory simulator through this link:

2. Investigate the rate of respiration of germinating mustard, bean, and wheat seeds.
3. Use 20 seeds for every experiment in 5, 15, 25, 35 oC.
4. To change the type of seed, select from the drop-down list, ‘Select the seeds’.
5. You can increase or decrease the number of seeds inside the beaker by moving the
‘Number of seeds’ slider accordingly.
6. Increase or decrease the temperature using the slider, ‘Temperature’.
7. Click on the ‘Start’ button to begin the experiment.
8. You can observe that the level of water rises in the delivery tube dipped in the beaker.
9. A timer is shown. Wait till the timer stops.
10. Measure the rise of water level in the delivery tube and enter the values in the test box,
‘Rising level of water (ml)’ on the left pane.
11. Click on the submit button to verify your result. (the correct result have green check mark)
12. Record the result in the table.
13. You can redo the experiment by clicking on the ‘Reset’ button.
