Activity-“‘Playing’ with Respiration”

Consider the impact of each substance on cellular respiration. Each substance has an influence at the
cellular level, and has no effect on hunger. Assume that the substances have no other impact on the cell
other than the one described.

1. Substance’s Effect: Impedes the binding of NAD+ to electrons.

Facilitator: Chastine Mae T. Garaygay

 Classification: Potentially effective in promoting (circle one):

Weight Loss Weight Gain

 Explanation for effect:

2,4-DINITROPHENOL
 Is an organic compound. It is a yellow crystalline solid that has a sweet musty odor.
 Is an uncoupler, or has the ability to separate the flow of electrons and the pumping of H +
ions for ATP synthesis.

Does DNP promotes weight loss or weight gain?

The substance impedes the binding of NAD + to electrons. Now what is NAD+? It stands for
Nicotinamide Adenine Dinucleotide, it is a carrier molecule that carries electrons and protons that
can be utilized by the mitochondria to produce ATP. NAD+ is converted to NADH and since there
is an uncoupler, NADH couldn't be formed to be used by the cell and the cell could not produce
ATP. When the body is not receiving enough ATP to break down the foods that we eat, it will
breakdown fats as a form of storage energy which will lead to weight loss.

2. Substance’s Effect: Makes the lipid bilayer of the inner mitochondrial membrane highly permeable to
(H+) protons.

Facilitator: Janna Alo

 Classification: Potentially effective in promoting (circle one):

Weight Loss Weight Gain

 Explanation for effect:

3. Substance’s Effect: Increases the number of free hydrogen ions or protons (H+) in the intermembrane
space of the mitochondria (between the outer and inner membranes).

Facilitator: Thalia Grace Granada

 Classification: Potentially effective in promoting (circle one):

Weight Loss Weight Gain

 Explanation for effect:

Performance Task
Learning that when glucose or another source of energy such as creatinine which is used by our
muscle cells, is degraded, it releases protons as well as energy. Then, those protons will be stored in the
internal membrane space, and when those protons go through the inter-membrane of mitochondria,
they make the ATPase enzyme work, where said enzyme takes that energy to produce ATP. Further, if
something ever happens such as preventing the protons to go through the inter-membrane of
mitochondria, then they can’t make the ATPase enzyme work, so that energy will be released as heat.
Leading for the body to “feel” that it is in need of more ATP, hence, your body cells will go “oh it
seems that I haven’t degraded enough glucose, so let’s degrade more glucose”: resulting for you to lose
weight.

4. Substance’s Effect: Partially blocks the H+/pyruvate symporter, interfering with the transport of
pyruvate into the mitochondria.

Facilitator: Shane M. Sumagaysay
