Apparatus:
❖ Light Microscope
❖ Transparent ruler with millimeter markings
Method:
1. A transparent millimeter ruler was placed on the stage, held down with the
stage clips, and observed in the desired field of view(scanning or low).
2. The metric edge of the ruler was focused on. Gentle pressure was applied to the
free end of the ruler to help adjust the thin ruler and allow for better focus.
3. The center of one millimeter marking was placed at the left edge of the field of
view and the diameter of the field was measured in millimeters
Results: TABLE SHOWING THE FIELD OF VIEW DIAMETER FOR MICROSCOPE BM 024
Field Of View
Objective Power
mm µm
X4 4.5 4500
X 10 2.0 2000
X 40 0.5 * 500
X 100 0.05 * 50
Calculation :
high power field /lower power field = lower power magnification /high power magnification
Graph: SHOWING THE RELATIONSHIP BETWEEN THE FIELD OF VIEW IN MICROMETERS AND OBJECTIVE
POWER
Precaution:
1. The microscope was held properly
Nicholas Ramnarine Lab# 2 10 th/1/2022
2. The microscope was placed in a cleared area away from water and other
electrical hazards
Source of Error:
1. Parallax error when reading ruler
2. Users vision may be damaged so accurate readings could not be found
Discussion:
The field of view is inversely proportional to objective power as shown in the graph, which means that
the higher the objective power goes the less the field of view as shown in the table.
Reflection:
During this lab I have learned how to use as well as measure the field of view for the light microscope.I
have heard this lab is difficult because of my blurred vision otherwise this lab was easy. This lab will help
scientists to manually find the field of view if they are missing equistment.
Conclusion:
The field of view of light microscope BM024 was calculated for the objective powers 4X,10X,40X and
100X .
