Florence Test
Result: the semen turned into a dark brown crystal choline periodide
formed resembling haemin crystal.
False negative: low choline content or decomposed stain
False positive result: other body tissue (except vagina) may contained
choline
B. Barberios Test
Result: the reaction will depend upon spermine in semen, when few
drops of barberios reagent (alcoholic soln of picric acid) added to
spermatic fluid will produced yellow, needle shape and rhombic crystal
of spermine picrate the test is even positive even the presence of
spermatozoa.
- The limitation is pleomorphic nature of crystals

C. Acid Phosphatase Test


- A quantitative test
- The amount of acid phosphatase is estimated in a measured
specimen one square centimeter of stain material
- Give positive in old stain as well
- It will show a color purple within a short period of time.

Stained Material – it will show a purple color within 30 second to 1


minute.
Unstained material – it will show no reaction to reaction to the reagent.
