Experiment #1

Screening Tests for Bloodstains

I. Objective
Determine the color reactions of a bloodstain when subjected to different preliminary
blood tests

II. Apparatus/Materials

Test tubes, spray bottle, test tube rack, ordinary filter paper, beaker (80ml), medicine
dropper, human blood stain, chicken bloodstain, pig bloodstain, fish bloodstain

III. Procedure

1. Kastle-Meyer Test

1.1. Rub a piece of filter paper on each of the questioned bloodstain.

1.2. Add one to two droplets of Kastle Meyer reagent then add two droplets of
3% Hydrogen Peroxide.

1.3. Record and illustrate your observations.

2. Leucomalachite Green Test

2.1. Follow the same procedure as in Kastle-Meyer Test.

2.2. Add one to two droplets of Leucomalachite green reagent then add two
droplets of 3% Hydrogen Peroxide.

2.3. Record and illustrate your observations.


3. Luminol Test
3.1. Spray a freshly prepared Luminol reagent on the area of the suspected
bloodstain.
3.2. The test area must be totally dark.
3.3. Observe the color that will appear on the stained area after having been
exposed to ultraviolet light.
3.4. Record and illustrate your observations.
