Aconitase is an enzyme that catalyses the stereo-specific isomerization of citrate to

isocitrate via cis-aconitate in the tricarboxylic acid cycle, a non-redox-active process.

1. (Aconitate hydratase; EC 4.2.1.3)

2. Primary sequence-

3.  Aconitase contains a Fe4S4 cluster which converts to Fe3S4 when the

enzyme is inactive.

4. Aconitase consists of four domains, three of which are tightly packed while the
fourth is more flexible. Aconitase contains a 4Fe-4S iron-sulfur cluster. This
iron sulfur cluster does not participate in redox as most do, but holds the OH
group of citrate to facilitate its elimination. It is at this 4Fe-4S site that catalysis
occurs and citrate or isocitrate is bound.
5. PDB file number of cis-Aconitase is 1B0J
6. The cis-Aconitase enzyme belong to the class of lysase
