STUDY THE EFFECT OF KI ON FLUORESCENCE EFFICIENCY

OF PYRENESULPHONIC ACID (PSA) & DETERMINATION OF


STERN-VOLMER QUENCHING CONSTANT

AIM
To study effect of KI on fluorescence efficiency of PSA & determine Stern-Volmer quenching
constant

MATERIALS REQUIRED
Sodium salt of pyrenesulphonic acid, potassium iodide, distilled water, micropipette, beakers,
and fluorescence spectrometer with cuvette cells etc.

OBSERVATIONS AND CALCULATIONS


Concentration of stock solution of PSA = 10 -6 M
Concentration of stock solution of KI = 10-4 M

Here, [KI] is calculated by M1V1 = M2V2 where V1 =Vol of KI(mL) ; V2 = 3mL ; M1 = 10-4 M
and M2 = [KI].

Now the experimental data points obtained to the curve y(x) = (slope).x+1 or y = 0.0064x + 1
(here, intercept = 0.9713≈1) which can be compared with the stern-volmer relationship equation,
Φof / Φf = [ Kq / ( Kf + Knr ) ][KI] + 1
Or

Φ f / Φf = [ KSV ][KI] + 1
Where, KSV = Stern-Volmer quenching constant

From comparison we get that the slope of the graph is equal to KSV .

So, Stern-Volmer quenching constant=0.0064μM-1 .
