SOP FOR MEDIA STERILIZATION BY AUTOCLAVING

Objective: To provide guidelines for the sterilization of culture media using autoclaving.

Scope: This SOP applies to personnel responsible for media sterilization.

Procedure:

Preparation of Culture Media:

a. Prepare the culture media according to established protocols and recipes.

b. Dispense the media into appropriate containers, leaving adequate headspace.

Labeling:

a. Label each container with the date, media type, and any other relevant information.

b. Use autoclave-resistant labels.

Loading the Autoclave:

a. Arrange containers in the autoclave in a way that allows for even heat distribution.

b. Do not overcrowd the autoclave chamber.

Autoclave Settings:

a. Set the autoclave parameters based on the specific media and equipment being sterilized.

b. Ensure that temperature, pressure, and sterilization time meet recommended standards.

Sterilization Process:

a. Start the autoclave cycle according to the chosen settings.

b. Ensure that the autoclave reaches the desired temperature and pressure and maintains them for
the specified duration.

Cooling and Venting:

a. After the sterilization cycle, allow the autoclave to cool and depressurize before opening.
b. Use caution when venting steam, and ensure proper PPE is worn.

Quality Control:

a. Check containers for any signs of media contamination or damage.

b. Discard any media containers that show signs of contamination.

Documentation:

a. Maintain records of the autoclave cycle, including date, time, temperature, pressure, and any
deviations from the SOP.

b. Document the results of the media quality control checks.
