ABSTRACT

Practicum-Internship is a requirement for all graduating BSAB student of the college. Its

objective is to expose students to different Agri-entrepreneur applying the theories,

techniques, philosophies and skills learned from various subjects.

This would also prepare them for future employment and income generation activities.

Last SY 2021-2022, the fourth year BSAB students were exposed to five production

activities, Whereas, this year, the proponents were assigned to four areas, as follows:

Zampen Native Chicken (IGP), Tissue culture Banana Lakatan production, Oyster

mushroom production, Food processing and market the available products.

The activities involved in varied agricultural production are described in this manuscript,

along with any issues that were encountered and how they were resolved. Financial

data statements are also included. This written piece was created not only for the

author's interest but also to aid readers in cultivating an open mind and a knowledge of

agriculture.

CHAPTER I

I. Introduction
Background of the study

The BSAB (Bachelor of science in Agricultural business) 4 th students of Western

Mindanao State University- College have conducted their practicum in campus.

Zampen Natuve Chicken- IGP. The Zampen Native Chicken is a chicken breed

created and develop by Western Mindanao State university (WMSU) in partnership with

the Philippine council for Agriculture, Aquatic, and Natural Resources Research and

Development (PICAARD). The name comes from its Geographical location of origin, the

Zamboanga Peninsula. The interns were able to experienced the activities in Zampen

Native Chicken-IGP that includes of collection of eggs, feeding, loading and unloading

of eggs in the incubator, cleaning and maintaining the areas, fermentation of forages,

feed formulation mixing, vaccination of chicken, deworming of chicken, slitting,

defeathering, and slaughtering of chickens.

Tissue Culture, Banana Lakatan. The Western Mindanao State University’s Tissue

culture in partnership with Department of Agriculture (DA) started the Banana Lakatan

Tissue culture production on March 2021. The component of Tissue Culture Laboratory

are the General washing area, heating room, chemical room, sterile room and

germination room. In which the interns were able to experienced the activities in the

different Tissue culture Laboratory areas.

Significance of the study

The study served as a resource, a manual, and a contribution to the students'


acquisition of various skills and knowledge in agriculture as well as their exposure to
various fields of endeavor in the future.

The research also significantly contributes by allowing the interns to improve their
marketing, technical, and planning skills through exposure to real-world projects like
Zampen Native Chicken-IGP and Tissue culture Banana Lakatan.

Internship’s Objectives

A. ZAMPEN NATIVE CHICKEN (Income Generating Project)


INTRODUCTION

The ZamPen Native Chicken is a chicken breed created and developed by Western
Mindanao State University (WMSU) in partnership with the Philippine Council for
Agriculture, Aquatic, and Natural Resources Research and Development (PCAARRD).
The name comes from its geographical location of origin, the Zamboanga Peninsula or
ZamPen.
The Zampen Native Chicken is a pure breed after undergoing a purification process.and
one of its most distinguishing characteristics is the taste of its meat. At present, the ZamPen
Native Chickens are not yet commercialized and the breeding stocks were mainly intended for
farmers who are willing and interested in raising this breed of chicken. The farmers who raise
and are willing to raise this breed are advised not to interbreed them with another breed of native
chicken as it will cause mongrelization, which may lead to decreasing its unique quality that was
developed through 10 years of research.

The current main focus of the Zampen Native Chicken is to produce more which
leads them to make a study about artificial insemination which is still currently ongoing to
know if it has a higher fertilization rate than natural breeding. Another is their study about
the feed formulation for the specific stages of the chicken growing where they use 50%
commercial feeds and 50% forages to lessen the feed expenses while still giving them
enough nutrients for growth.

INCUBATING

The main goal of incubation is to increase the hatching rate and produce healthy
hatched birds. Incubation temperature is not only the most important factor in the
hatching process, but is additionally related to the growth performance of chicks after
hatching. The eggs will be incubated for 11 days, and in the 7th day candling process
takes place. In this process unfertile and fertile eggs will be identified. The fertile eggs
will be bought back to the incubator after the candling process.

BROODING

Day-old chicks are tagged using the slitting technique following hatching day, before
being moved to the brooding area. Brooding area is where the day-old chicks were kept to
provide artificial heat. Day old chicks will stay in the brooding area for 21 days. The New Castle
Disease Vaccine will be administered to brooder chicks prior to transfer to the hardening area.

HARDENING

Chicks from brooders will be moved into the hardening area and will receive
electrolyte supplements and artificial light support. For 45 days, brooder chicks will
remain in the hardened area.

GROWING

The Hardened chicks will be moved to grower pens at the 45th day, where they
will be exposed to various weather conditions. The grower hens take several days to
properly acclimate to their new environment, before releasing hardened chicks into
grower enclosures and to prevent mortality, workers will assess the weather. At this
point, it is simple to define the pure breed zampen, which will be chosen for breeding
while the off-type zampen will be for slaughtering purposes.

period for chickens lasts between six to twelve months.

B. BANANA LAKATAN TISSUE CULTURE


I. INTORDUCTION

Background of the study

Tissue culture is the cultivation of plant cells, tissues or organs on specially formulated
nutrient media. Under the right condition the entire plant can be regenerated from a single cell or
tissue. Tissue Culture often called micro propagation is seen as important technology for
developing countries for the rapid production of quality planting materials. Unfortunately, tissue
culture is labor intensive, time consuming, and can be very costly.

The tissue culture center propagated banana (lakatan) through the use of chemicals
(medium) and banana tissue. The project was developed to rapidly reproduce a uniform or a
characteristic of preferred species of banana. The intern students were assigned in the tissue
culture center to expose them in different laboratory skills and acquire experiences about plant
propagation with technology.

The Underlying principles in plant tissue culture are simple:

a) It is necessary that the plant came from the intact plant.


b) It is necessary that the plant part be provided with the appropriate environment.
c) This can be done by providing suitable culture medium and proper culture
condition.
d) Finally, all the above activities should be done under aseptic condition.

The importance of tissue culture

a) Rapidity with which a selected species may be generated easy and economical.
b) Production of many uniform plants produced in the short period of time.
c) Characteristic of preferred species maintain the generic make-up and
morphological

Stock solution namely: MSI, MSII and MSIII

1. Assemble all glassware: beaker, spatula, aluminum foil, volumetric flask, funnel, stirring

2 Check the digital balance and arrange the chemical to be used.

3. Label the beakers according to the intended use.

4. Weigh the specified amount of each of the chemicals listed in the table.

5 Dissolve the chemicals separately in minimum volumes of distilled water either by

stirring manually or in magnetic stirrer, until dissolve.

6 Pour the fully dissolve content of each beaker in to 1.000 ml volumetric flask, using glass

funnel, consequently rinse beaker with distilled water and add the rinse into the

volumetric flask. Dilute the resulting solution to the final volume of 1 liter

7. Mixed the solution thoroughly with magnetic stirrer or agitating the last before

transferring to properly dark bottle

8. Label the bottle with the name and concentration of stock solution and date of

preparation.

9. Refrigerate the stock solution.

Note: Stock solutions are only good for 6 months

Preparation of culture media (Proliferation media)

A. Prepare and align all stock solution needed (MS I, MS II, MS III,

iron chelate and growth regulators).

B. Weigh 20g sugar and dissolve in distilled water. Weigh 7g agar


C. Measure the following stock solution add each measured stock solutions with the used of
graduated cylinder rinse graduated cylinder with distilled water and pour it into 500ml.
D. Melt the agar using microwave oven melting time in a microwave oven depends on the
amount of agar volume of the medium.
E. Mix the melted agar with the prepared stock solution and bring into the final volume of 1
liter. Measure the pH of the medium and adjust the pH level to 6.0.
F. After getting the pH, dispense the medium into suitable, autoclavable culture media like
packing jars.
G. Sterilize the dispensed media in the autoclave for 15 minutes at 15 Psi.

PRACTICES AND MANAGEMENT

The interns collected suckers from the banana plantation of WMSU AGRI. by digging
the soil around the sucker, and carefully detaching the sucker from the mother plant to
avoid cracking the corm of the sucker.
After collecting the suckers, they washed it with running water to remove any adhering
dirt and contamination.
They trimmed the leaf bracts and corm tissue up to the size sufficient enough to protect
the growing point from bleaching.
The trimmed corm was washed with soap solution.
Corm was disinfected by soaking it in formalin for 15minutes
Disinfectant was decanted and corm was rinsed three times with distilled water. The
disinfected corm was the transferred to the inoculation chamber for initial culture.
Establishing initial culture
 In the culture room, the interns further excised the corm. Making sure that the

 The corm was divided longitudinally into four parts having the shoot apex in
each section and inoculated in proliferation media.

Nursery Management

When the white cord roots have developed and the plantlets have 3-4 expanded leaves

and attained a minimum height of three to four inches, they are ready for potting out.

Acclimatization

Selected cultured banana with roots were then placed outside of the laboratory to

introduce a semi-natural-laboratory environment for at least one day and further


transplanted in

the acclimatization area to acclimatize the plantlets into a new environment before
transplanting it in the nursery.

Weeding and Watering

The seedlings in the nursery had their dead leaves and weeds pulled. In order to

obliterate possible nutritional rivals. Once daily, either in the morning or in the

late afternoon or early morning

Monday (February 27, 2023)


Acclimatized the plantlets and covered with bottles to maximize the temperature because the
plantlets are still in adjusting of the new environment.
Washed the plantlets of a running water to remove the remaining cultured media of the
germinated Banana Lakatan.
Applied fungicide to the banana lakatan plants and soaked the plantlets before to transplant.
Gathered Banana Lakatan suckers (12 pcs.)
Washed and rinsed the suckers then cut the stem.
Tuesday (February 28, 2023)
Cleaning and disinfection of a corms.
Setting up and sanitized the inoculation chamber (Laminar)
Inoculated 12 pieces of corms
Total inoculated is 24 pieces

CHAPTER III
PROBLEMS ENCOUNTERED AND SOLUTION UNDERTAKEN

 Problems Encountered
1. Zampeen Native Chicken - IGP
 Lack of feeds supply
 Unavailability of Disposal area for Dead chicks
 Damaged pens due to bad weather conditions
 Waste disposal
 No proper area for slaughtering
 Solution Undertaken
 Mixing of forages (trichantera, coco dregs, malunggay, oregano etc)
 The interns proposed to make the septic tank a disposal area for dead birds
 Pens were fixed and changed the damaged materials with stronger ones.
2. BANANA LAKATAN TISSUE CULTURE

 Lack of laborers that can speed up the production
 Some of the bottles were not totally cleaned and rinsed

 The interns rewashed the bottles to ensure that there would be no contamination

CHAPTER IV

CONCLUSION

The Bachelor of science in Agricultural Business of Western Mindanao State University


interns had accomplished the goals of their practicum in campus during the second semester of
the school year 2022-2023.

The interns successfully conducted their practicum internship, and they were able to learn
the feed formulation, feed fermentation, feeding management, artificial insemination, care, and
maintenance in every area. In the Tissue Culture Banana Lakatan production, they were able to
perform media preparation, stock solution, and establish initial culture of the Banana Lakatan, in
which they were to produced 24 pieces of the inoculated banana corms.

Furthermore, the Bachelor of Science in Agricultural Business interns conclude that thee
entire experienced and exposure of the team has helped to improved each one’s character and
personality and was able to become better through the entire process in the different areas. The
group has achieved their goals easily through motivating each other and has a unity to solved
the problems encountered.

RECOMMENDATION

Zampen Native Chicken - IGP

The proponents recommended creating a space for seclusion and waste disposal. As a
result, the surroundings become cleaner and the mortality rate of the chickens will be reduced.
Pen houses should be expanded for the chickens to have enough space and prevent stampede.

Tissue culture Banana Lakatan

The staff needs an additional employee to easily done the activities in producing Banana
lakatan and to produces mass production of Banana Lakatan.

CHAPTER V

LEARNING EXPERIENCE

The fourth- year Bachelor of science in Agricultural Business of Western Mindanao State
University interns were able to conduct their practicum and were exposed to various research-
based project at Western Mindanao State University; Zampen Native Chicken – IGP, and Tissue
culture Banana Lakatan.

The interns were able to experienced and Learned from the different areas/activities in
Zampen Native Chicken – IGP. Just like feeds fermentation and formulation, Artificial
insemination, slitting, tagging, vaccination, eggs collection, labeling, incubating, candling,
deworming, slaughtering and transferring of chickens to the next growth stage. In the Tissue
culture, the interns also able to experienced collect suckers, acclimatizing, mixing of chemicals
to make a stock solution, media preparation and inoculate corms in the bottles which was has a
media.

DEFINITION OF TERMS

Artificial Insemination (AI) – is a method and process in which the sperm was
manually and directly delivered into the female vagina in hope of getting fertilized.

Breeder – intended for breeding or reproducing.

Brooding – the period after hatching of eggs where special care and attention must be
given to ensure the health and survived.

Candling – the process of holding strong light below and above the eggs to observe the
embryo in search of signs of life or development.ee

Coco dregs – is a dried coconut.

Corm – rounded thick modified underground stem base acting as a vegetative


reproductive structure where the banana plant produces suckers.

Culture media – a growth media that could be solid, liquid, and semi-solid created to
support growth.

E-MAS or effective Microorganisms Activated solution – is a fertilizer that elicits the


growth of microbial benefits.

Feeds Fermentation – is the process of fermenting feeds.

Feed Formulation – is the process of determining the volumes of ingredients and


additives to blend to create compound feeds that meets up the target nutrient
requirements of a species and achieve the goals of production at an optimized cost.

Grower chickens – are chicken allotted to be given away to farmers who are interested
in growing of chickens.

Hardening – is the stage that allows the chick ti adapt to their surroundings and
environment to become stronger before letting loose in the range.

Hardened – the stage that chickens had fully adapted to their surroundings,
environment, and increasing their level of survival.

Incubator – a device or insulated enclosure used for the hatching of fertile eggs which

Laminar Flow – a laminar flow cabinet or tissue culture hood is a carefully enclosed
bench designed to prevent contamination.

Molasses – is a substance resulting from refining sugarcane or a bi-product.

Proliferation – a single isolated cell that divides and rapidly reproduce which gives rise
to a new plant.

Rooting – a type of culture media used for the multiplication of plants.

Stock solution – is composed of different solid solution, each weigh out in an


appropriate amount diluted in lower concentration such as distilled water.
