Assessment Title DOP

Summary of the The assessment was carried out by the trainee, the Principal
situation for the Clinical Bioinformatician, and a Clinical Bioinformatician to meet the
assessment Direct Observation of Practical Skills (DOP) requirements for this
module. The evaluation involved analyzing the Quality Metrics (QM)
of Next-Generation Sequencing (NGS) data using the Sequencing
Analysis Viewer. Before the assessment, various cases were
reviewed, along with a discussion about additional QM analyses
that the bioinformatics team routinely performs. A specific case was
then selected for analysis. The trainee proceeded to conduct the
analysis, for example, determining whether the NGS data met the
QM cutoff for clinical analysis.

DOP question were given at the end of the DOP, which the trainees
answered individually.

C. Roche analysed the data accurately, detailing the various plots and their potential
impact on variant interpretation. She adeptly identified instances of low-quality NGS
Bioinformatics Illumina data and understood where to examine key metrics (e.g., error
rate or Q30) to assess if the results surpassed the thresholds set by Illumina. During the
discussions in the DOP sessions, she occasionally expressed uncertainty about the
correct responses but effectively navigated these moments by posing relevant questions.
This approach enabled her to acquire the essential information needed to accurately
address the questions.

Regarding confidentiality, it is challenging to evaluate in this DOP, as the assessment did not
provide an opportunity to assess the trainee's awareness of patient confidentiality.

In terms of maintaining records, C. Roche took notes during the explanations and discussions.
During several training seasons I had had with her, she actively has taken notes, information she
has used in this DOP.

The analysis of this data is relatively complex and high communication is needed to explain
results in simple terms. C. Roche showed his skills to do this on several occasions.

C. Roche reflected during his DOP for instance about the impact of this NGS quality metrics
review. He is aware about the impact of low QM NGS data in the following analysis and
ultimately in diagnostic provided and patient care. For instances, she explains the impact of low-
quality sequencing data correctly in the DOP questions.

In general, C. Roche successfully met the expectations for this introductory module,
demonstrating a solid foundational understanding of the subject. More importantly, she
comprehended the significance of bioinformatics within her chosen field of specialization: clinical
genomics.
