Calibration of Eyepiece Graticules

How do we measure cells?

eye piece graticule

placed in the eyepiece of a microscope

scale changes with each objective lens

therefore need to calibrate
Eye Piece Graticules

Fitted in the
eyepiece

100 divisions

Arbitrary units

Needs Calibration
Stage Micrometer

10mm

100 divisions

Each is 0.1mm
To Calibrate

Superimpose the two scales

Match up two points
Count the number of epg divisions = x
Count the number of micrometer divisions = y
One micrometer division is 0.1mm

Total width = y x 0.1mm

If there are x epg divisions

So x epg divisions = y x 0.1mm

then ONE epg division = (y x 0.1) mm

If you have an object on this power you can now
find the Actual Size of the image

Count the number of epg divisions

Multiply by the calibrated width of one division
Size of drawing (mm)
Magnifcation =
Actual size of object (mm)
Using an eye piece graticule

1. eye-piece graticule 2. stage micrometer scale

100 eye piece graticule measurements:

0.25mm
1 eye piece graticule measurement:
0.25/100 = 0/0025mm (2.5µm)
Using an eye piece graticule

3. cheek cell: 20 units

cheek cell: 20 units x 2.5µm = 50µm
