WHAT IS PETRI DISH?

• The Petri dish is named


after its inventor, Julius
Richard Petri (1852-
1921), who worked at the
Imperial Health Office in
Berlin as Robert Koch’s
assistant. He created a
small, circular, flat-
bottomed dish with sides.
• A Petri dish is a
clear glass or plastic
cylinder with a lid
that is used to retain
a thin coating of
agar. Used to
cultivate
microorganisms
such as bacteria,
fungus, and others.
TYPES OF PETRI DISH BASED ON
MATERIALS
1. Glass Petri Dishes
• Lab equipment made of glass
can be sanitised and reused in
an autoclave or laboratory oven.

• Some goods are constructed


to survive repeated washings
and sanitising.

• Borosilicate and quartz glasses


have a strong thermal shock
resistance.
2. Plastic Petri Dishes

• Disposable plastic Petri plates


are an excellent alternative for
scientific work where cross-
contamination may be an issue.

• Fluoroelastomer materials
have excellent resistance to
heat, oil, and chemicals,
although they frequently operate
poorly at low temperatures.
Petri Dish in Monggo Seeds
Germination percentage is an estimate of the viability
of a population of seeds.  The equation to calculate
germination percentage is:  GP = seeds
germinated/total seeds x 100 .The germination rate
provides an measure of the time course of seed
germination.  Germination rate is determined by
calculating the GP at different time intervals after
planting and then plotting these data. 
Procedure:   

1.Obtain a petri dish and filter paper. 


 
2.Select a seed source and record the name in Table 1.

3.Count out 50 seeds (fewer if large seeds).  Soak them in water for an hour.   

4.Spread the seeds on a layer of filter paper in the dish

5.Moisten the paper with enough water so that the seeds just barely begin to float (about
10 ml).

6.Put on the lid and place the dish in the dark.

7.Record the number of seeds that germinate daily.
