Lead Standard Dilutions (SPIKING)

Standard Available:
Required spike final concentrations in
10ml:

10,000,000 ppb
0.15ppb and 0.08 ppb

M 1 V 1=M 2 V 2
0.08 ppb10 ml=10,000,000 ppbV 2 ml
V 2=0.00000008 ml

Since a pipette that could transfer such a small volume of


lead standard solution was not available, successive
dilutions had to be made.
Dilution 1:
M 1 V 1=M 2 V 2
M 2 ppb5000 l=10,000,000 ppb5 l
M 2=10,000 ppb

Dilution 2:
M 1 V 1=M 2 V 2
M 2 ppb5000 l=10,000 ppb50 l
M 2=100 ppb

This 100 ppb lead solution was used as the new standard
to spike the samples.
Highest Spike:
M 1 V 1=M 2 V 2
0.15 ppb10,000 l=100 ppbV 2 l
V 2=15 l

Lowest Spike:
M 1 V 1=M 2 V 2
0.08 ppb10,000 l=100 ppbV 2 l
V 2=8 l

V 2=0.0005 ml=0.5 l

Since a pipette that could transfer such a small volume of


Indium standard solution was not available, it was
decided to dilute the standard to 10 ppb.
Dilution 1:
M 1 V 1=M 2 V 2
10 ppb500 ml=1,000,000 ppbV 2 ppb
V 2=0.005 ml=5 l

Next, the volume of this 10 ppb standard that had to be


added to the samples to make the final Indium
concentration 5ppb was calculated:
M 1 V 1=M 2 V 2

5 ppb10 ml=10 ppbV 2 ml


V 2=5 ml

Plot graph of final spike concentration vs signal
Extrapolate to get x intercept
X intercept = concentration of lead from chocolate
